Rapid diagnosis of tuberculosis in cattle reacting positive in antemortem assays is crucial in countries where eradication programs are operated to confirm the presence of the infection in tuberculosis-free herds. This study evaluated the accuracy of histopathologic examination by hematoxylin and eosin and Ziehl-Neelsen (ZN) staining applied in this framework, when suspected lesions are caused by low infectious doses and are detected in early stages of the disease. For this purpose, histologic methods were compared with mycobacterial culture as reference test on suspected lymph node samples from 173 cattle reacting positive in antemortem tests. Histopathology demonstrated high sensitivity (93.4%) and specificity (92.3%), while ZN sensitivity and specificity were respectively 33.9% and 100%. There was good agreement between histopathology and bacterial culture, suggesting that histopathologic examination is a reliable tool for rapid diagnosis in countries where active tuberculosis eradication programs allow the prompt identification and elimination of reactor cattle. Histopathology permits identification of typical mycobacterial lesions and its differentiation from other causes.  相似文献

Putative genetic markers have been associated with ETEC F4 (Mucine 4 [MUC4]; MUC4GG;CG as susceptible; MUC4CC as resistant) and F18 (Fucosyltransferase 1 [FUT1]; FUT1GG;AG as susceptible; FUT1AA as resistant) resistances respectively. In this study, 71 post‐weaning pigs were followed from d0 (35 days old) to d42 (77 days of age) to investigate the effect of MUC4 or FUT1 genotypes on the mid‐jejunal microbiota composition, pigs expression of genes related to inflammation (IL8, GPX2, REG3G, TFF3, CCL20 and LBPI) and glycomic binding pattern profile (Ulex europaeus agglutinin I [UEA] fucose‐binding lectin and peanut agglutinin [PNA] galactose‐specific), and on blood plasma targeted metabolomics profile, faecal score and performance parameters of growing healthy pigs. The MUC4 and FUT1 resistant genotypes improved the pigs’ growth performance and had firmed faecal score susceptible genotypes in d0–d21 period. Pigs with MUC4GG genotype had a higher jejunal expression of genes relate to immune function (CCL20 and REG3G) than MUC4CG and MUC4CC pigs (p < 0.05). MUC4CG pigs had higher expression of TFF3 (implicated in mucosal integrity) than MUC4GG and MUC4CC (p < 0.05). FUT1 influenced the alpha‐ and beta‐jejunal microbial indices. The FUT1AA group had a higher number of operational taxonomic units (OTUs) belonging to Lactobacillus genus, while FUT1GG group had a higher number of OTUs belonging to Veillonella genus. MUC4CC pigs had lower scores for UEA on brush borders and goblet cells in villi than MUC4GG (p < 0.05). FUT1AA pigs had lower UEA positivity and higher PNA positivity on brush borders and goblet cells than FUT1AG and FUT1GG (p < 0.05). Both FUT1 and MUC4 influenced the metabolic profile of healthy pigs. Results highlight the role of MUC4 and FUT1 on pig intestinal homoeostasis and improved the knowledge regarding the potential interaction between host genetics, gut microbiota composition and host metabolism in a healthy status.  相似文献

Conventional in vitro fertilization has not yet been implemented in the equine species. One of the main reasons has been the inability to develop a culture medium and incubation conditions supporting high levels of stallion sperm capacitation and hyperactivation in vitro. Although different culture media have been used for this purpose, human tubal fluid (HTF) medium, widely used in the manipulation of human and mice gametes, has not been reported so far in stallion sperm culture. The first part of this study aimed to compare HTF and Whitten's media on different stallion sperm quality and capacitation variables. Additionally, the effect of procaine, aminopyridine and caffeine in both media was evaluated on sperm motility parameters at different incubation times. Integrity and destabilization of the plasma membrane were evaluated by merocyanine 540/SYTOX Green (MC540), mitochondrial membrane potential (?Ψm) using tetramethylrhodamine methyl ester perchlorate (TMRM), acrosome membrane integrity by PNA/FITC and tyrosine phosphorylation by P‐tyrosine mouse mAb conjugated to Alexa Fluor® by flow cytometry. Motility parameters were evaluated using the integrated semen analysis system (ISAS®). We found no differences between Whitten's and HTF media and incubation time in terms of sperm viability, uninduced acrosome membrane damage or mitochondrial membrane potential at 30‐ and 120‐min incubation. Membrane fluidity (MC540) increased in both media at 30‐ and 120‐min incubation compared to noncapacitating conditions. Similarly, tyrosine phosphorylation increased in both media in capacitating conditions at 2‐ and 4‐hr incubation compared to noncapacitating conditions. Although procaine showed the best result in terms of sperm hyperactivated motility in both media, aminopyridine also showed parameters consistent with the hyperactivation including an increase in curvilinear velocity and decrease in straightness. In conclusion, HTF medium and aminopyridine equally support capacitation‐related parameters in stallion sperm.  相似文献

Three levels (0, 1 and 2%) of conjugated linoleic acid (CLA) were combined with two levels (low and high) of monounsaturated fatty acids (MUFA) for pig feeding. Productive, carcass and meat quality traits were studied. Large White ♂ × Landrace × Large White ♀ gilts (= 288) weighting 70 kg were randomly allotted to 6 different feeding treatments and fed to a final average weight of 107 kg. Loins were taken from 48 animals (8 animals randomly selected from each treatment). No differences due to dietary CLA, MUFA or CLA × MUFA interaction were found on average daily gain, average daily consumption, feed conversion ratio, carcass yield, backfat thickness, loin weight, loin pH and loin colour. A significant increase in intramuscular fat content (= 0.010) and in saturated fatty acids (SFA) (< 0.001), and a decrease in MUFA (= 0.001) and desaturase indices were found as consequence of dietary CLA, regardless the MUFA level. Therefore, dietary CLA, MUFA and their interaction did not influence productive and carcass traits of pigs. However, the use of CLA for swine feeding increased the intramuscular fat content and modified the fatty acid profile, regardless the MUFA level of the diets.  相似文献

Agricultural intensification is a major cause for biodiversity loss. It occurs at field scales through increased inputs and outputs, and at landscape scales through landscape simplification. Agri-environment schemes (AES) of the European Common Agricultural Policy (CAP) aim at reducing biodiversity loss by promoting extensification of agricultural practises mostly at field scales. We present a conceptual model for the relationship between landscape complexity and ecological effectiveness of AES based on (a) non-linear relationships between landscape complexity and abundance and diversity at field scales and (b) four possible interactive scenarios between landscape- and field scale effects on abundance and diversity. We then evaluated whether and how effectiveness of AES interacted with landscape-scale effects of intensification along a landscape complexity gradient established in central Spain. Pairs of cereal fields with and without AES but with the same landscape context were selected in three regions differing in landscape complexity. Effectiveness of AES was measured as differences between paired fields in species richness and abundance of five target groups (birds, grasshoppers and crickets, spiders, bees and plants). Landscape metrics were measured in 500–m radius circular plots around field centres. Positive, negative and no effects of landscape complexity on effectiveness of AES were found, suggesting that effects of complexity on effectiveness of AES changes from positive to negative along gradients of landscape complexity. Effectiveness of AES for improving biodiversity was then constrained by landscape. Compulsory measures aimed at enhancing or maintaining landscape complexity would enhance the effectiveness of AES for preserving biodiversity in farmed landscapes. For M. Rodents play an important role in rangelands through the engineering of extensive burrow systems, which provides key habitats for many animal and plant species. We have analyzed the long‐term variation in the abundance and distribution of rodent burrows in grazing ecosystems of southern Russia (Kalmykia) under the landscape change from desert to steppe caused by the drastic reduction of livestock after the collapse of the USSR in the early 1990s. We conducted burrow surveys in the “desert” (1980) and “steppe” (2017) periods on 19 3‐km transects. We found considerable changes in burrow abundance and distribution, as well as evidence of desert habitat fragmentation and isolation caused by the expansion of tall‐grass communities. Burrows of the open‐dwelling diurnal ground squirrel (Spermophilus pygmaeus), the dominant and the keystone species during the “desert” period, almost completely disappeared from the rodent burrow network by 2017, indicating significant habitat loss. In contrast, the burrows of the folivorous social vole (Microtus socialis) which was rare in the 1980s, became abundant and ubiquitously distributed. The burrow density of the desert‐dwelling psammophilous midday gerbil (Meriones meridianus) decreased, while the distances between occupied patches increased, indicating desert habitat fragmentation and loss of population connectivity. Burrows of the folivorous tamarisk gerbils (M. tamariscinus) were recorded only sporadically in both 1980 and 2017. The observed changes in the rodent burrow network, the key component of grazing ecosystems, correlate with rodent species ecology and can have long‐term and important consequences for ecosystem functioning.  相似文献
