High resolution Fourier transform mass spectrometry (HRFTMS) and nuclear magnetic resonance (NMR) spectroscopy were employed as complementary metabolomic tools to dereplicate the chemical profile of the new and antitrypanosomally active sponge-associated bacterium Actinokineospora sp. EG49 extract. Principal Component (PCA), hierarchical clustering (HCA), and orthogonal partial least square-discriminant analysis (OPLS-DA) were used to evaluate the HRFTMS and NMR data of crude extracts from four different fermentation approaches. Statistical analysis identified the best culture one-strain-many-compounds (OSMAC) condition and extraction procedure, which was used for the isolation of novel bioactive metabolites. As a result, two new O-glycosylated angucyclines, named actinosporins A (1) and B (2), were isolated from the broth culture of Actinokineospora sp. strain EG49, which was cultivated from the Red Sea sponge Spheciospongia vagabunda. The structures of actinosporins A and B were determined by 1D- and 2D-NMR techniques, as well as high resolution tandem mass spectrometry. Testing for antiparasitic properties showed that actinosporin A exhibited activity against Trypanosoma brucei brucei with an IC50 value of 15 µM; however no activity was detected against Leishmania major and Plasmodium falciparum, therefore suggesting its selectivity against the parasite Trypanosoma brucei brucei; the causative agent of sleeping sickness.  相似文献

OBJECTIVE: To characterize ground reaction forces (GRFs) and determine whether there were correlations between forces and passive coxofemoral joint laxity in puppies. ANIMALS: Fifty-one 16-week-old hound-breed dogs. PROCEDURE: Force-plate gait evaluation and distraction radiographic imaging were performed. Ground reaction forces evaluated included x (mediolateral), y (craniocaudal breaking and propulsion), and z (vertical) peak force and impulse. Z-plane limb loading and unloading rates, loading interval, and weight distribution and y-plane stance time breaking and propulsion percentages were calculated. One-way ANOVA with the Duncan multiple range test was used to evaluate differences in gait variables among limbs. The relationships of left, right, highest, and mean distraction index (DI) with individual limb data of each dog were evaluated with the Spearman rank correlation. Left and right DIs were compared by means of linear regression analysis. RESULTS: Mean +/- SEM DI was 0.67 +/- 0.02. Left and right DIs were strongly correlated, but there were no significant relationships between DIs and gait variables. Most fore- and hind limb gait variables differed significantly, whereas paired fore- and hind limb gait variables did not. Asymmetry was most pronounced in the x- and y-planes. CONCLUSIONS AND CLINICAL RELEVANCE: GRFs were consistent with those of clinically normal mature dogs, supporting an absence of association between GRF and DI in young dogs. The GRFs and elucidation of the relationship between GRFs and DI may be useful for future studies in immature dogs.  相似文献

Fishing can drive changes in important phenotypic traits through plastic and evolutionary pathways. Size‐selective harvest is a primary driver of such trait change, has received much attention in the literature and is now commonly considered in fisheries management. The potential for selection on behavioural traits has received less study, but mounting evidence suggests that aggression, foraging behaviour and linked traits can also be affected by fishing. An important phenomenon that has received much less attention is selection on reproductive phenology (i.e., the timing of breeding). The potential for this type of “temporal selection” is widespread because there is often substantial variability in reproductive phenology within fish populations, and fisheries management strategies or fishermen's behaviours can cause fishing effort to vary greatly over time. For example, seasonal closures may expose only early or late breeding individuals to harvest as observed in a range of marine and freshwater fisheries. Such selection may induce evolutionary responses in phenological traits, but can also have demographic impacts such as shortened breeding seasons and reduced phenotypic diversity. These changes can in turn influence productivity, reduce the efficacy of management, exacerbate ongoing climate‐driven changes in phenology and reduce resilience to environmental change. In this essay, we describe how fisheries management can cause temporal variability in harvest, and describe the types of selection on temporal traits that can result. We then summarize the likely biological consequences of temporally selective fishing on populations and population complexes and conclude by identifying areas for future research.  相似文献

The polyphagous shot hole borer (PSHB), Euwallacea nr. fornicatus (Coleoptera; Scolytinae) has become a serious threat to the avocado industry and several shade tree species in Israel. Branch wilting and tree mortality is the outcome of PSHB galleries. Understanding the relationship between avocado trees and the PSHB is required for considering management strategies. In Israel, 52 tree species from 26 botanical families were attacked by the PSHB, but only 12 species were suitable for beetle reproduction. All examined avocado cultivars were attacked, but ‘Hass’ most severely. Large and medium diameter avocado branches were more resistant to PSHB, compared to thin branches. Effectively, gallery density increased as branch diameter decreased. Concomitantly, in large and medium diameter branches, extensive sugar exudation occurred and beetle attack rarely progressed to the formation of natal galleries, whereas minimal sugar exudation was observed in thin branches. This was more evident in those that were weakened by repeated attacks followed by successful beetle colonization. PSHB prefers and successfully colonized branches that had been previously attacked by its conspecifics, and reproduction was much higher in these branches, as opposed to initial attacks. Lesion frequencies increased from late spring (April) until late summer (September). Avocado branches at the early stages of beetle colonization may be identified by sugar exudation at the base of the thin branches. The main approach for reducing damage caused by the PSHB is sanitation, achieved by the removal of colonized branches and intact infested pruned slash.  相似文献

Mango malformation disease (MMD), caused by Fusarium mangiferae, is a major constraint to mango production, causing significant yield reduction resulting in severe economic impact. The present study characterizes fungal localization in planta during initiation and development of vegetative and floral malformation. Young mango trees were artificially inoculated with a green fluorescent protein (GFP)‐expressing strain of F. mangiferae. Shoots and buds were sampled periodically over a period of more than a year and localization of the GFP‐expressing fungi was determined using confocal microscopy. Fungal localization appears to be epiphytic: mycelia remained in close contact with the plant surface but did not penetrate the tissue. In vegetative malformation and in young inflorescences, the fungus was confined to protected regions between scales, young leaf bases and buds. Fungal colonization was only very rarely detected on open leaves or on exposed shoot sections. In developed flowers, mycelia were localized mainly to protected regions at the base of the flower organs. Upon development of the inner flower organs, specific mycelial growth occurred around the anthers and the style. Mycelial penetration through the stylar tract into aborting carpels was observed. For several months, mycelia were confined to the surface of the organs and were not detected within plant tissues. Only at later stages, transient saprophytic growth of the fungus was detected causing the malformed inflorescences to senesce and collapse, concurrent with dispersion of conidia. Implications of the present study on MMD in natural field infections are discussed.  相似文献

It is increasingly recognised that land degradation monitoring and assessment can benefit from incorporating multiple sources of knowledge, using a variety of methods at different scales, including the perspectives of researchers, land managers and other stakeholders. However, the knowledge and methods required to achieve this are often dispersed across individuals and organisations at different levels and locations. Appropriate knowledge management mechanisms are therefore required to more efficiently harness these different sources of knowledge and facilitate their broader dissemination and application. This paper examines what knowledge is, how it is generated and explores how it may be stored, transferred and exchanged between knowledge producers and users before it is applied to monitor and assess land degradation at the local scale. It suggests that knowledge management can also benefit from the development of mechanisms that promote changes in understanding and efficient means of accessing and/or brokering knowledge. Broadly, these processes for knowledge management can (i) help identify and share good practices and build capacity for land degradation monitoring at different scales and in different contexts and (ii) create knowledge networks to share lessons learned and monitoring data among and between different stakeholders, scales and locations.
