Four frequently used extractants (H2O, 0.1 M NaCl, 0.016 M KH2PO4, and 0.5 M NaHCO3) as well as different extraction conditions have been tested for sulphate extraction from gypsum‐free agricultural soils. Water is the preferable extractant for soils with pH > 6. Two extraction steps have to be carried out for complete extraction (> 95%). A 0.016 M KH2PO4 solution was found to be the most efficient extractant for soils with a pH < 6 within a single extraction step. A shaking frequency of 170 min‐1 and a duration of extraction of 4 hours are the optimized conditions for the sulphate extraction with H2O and KH2PO4 solution.  相似文献

The labile organic carbon(C) and C-related enzymes are sensitive indicators capturing alterations of soil organic matter(SOM),even in a short-time scale.Although the effects of crop husbandry and land use change on these attributes have been well studied,there is no consensus about how plant phenology may impact them.This study aimed to determine the short-term effect of six distinct phenological stages(PS-1:full bloom;PS-2:fruit set;PS-3:pit hardening;PS-4:physiological maturity;PS-5:60 d after physiological maturity;and PS-6:fall) of peach on the changes in soil organic carbon(SOC) fractions of different oxidizability,labile C pools,and C-cycle enzyme activities in soils,for two consecutive years(2015 and 2016) in the North-Western Himalayas(NWH).Peach rhizosphere soils were sampled at the topsoil(0–15 cm) and subsoil(16–30 cm) layers,along with rhizosphere soils from adjacent perennial grasses,which served as a control.Values for most of the assessed parameters,including very labile C,labile C,microbial biomass C,permanganate oxidizable C,dissolved organic C,mineralizable C,amylase activity,and carboxymethyl-cellulase activity,were significantly(P ≤ 0.05) higher at PS-3 than at other phenological stages of peach.Conversely,a sudden decline in these soil variables was recorded at PS-5,followed by a slight buildup at PS-6,particularly in the topsoil of the peach orchard.Short-term changes in organic C fractions of different oxidizability,influenced by peach phenological stage,significantly(P ≤ 0.05) affected C management index,C pool index,and lability index.Both the C management index and lability index showed their highest values at PS-3 and their lowest values at PS-5,clearly indicating short-term accretion and depletion of SOC,in tandem with the peach phenological events.Principal component analysis suggested that a composite of soil indicators,including microbial biomass C,dissolved organic C,amylase,and invertase,could help detect short-term changes in SOC content.It is concluded that peach phenological events had a major impact on the short-term variations of the studied soil variables,which could be attributed to changes in the above-and belowground plant residues,as well as the extent of nutrients and water acquisition.  相似文献

Phalaris arundinacea sowie Bromus inermis unter Beregnung in der Regel gesicherte Mehrerträge an Trockensubstanz. Auch die Ertragsverteilung zwischen den einzelnen Aufwüchsen dieser Bestände konnte durch Zusatzwasser deutlich ausgeglichener gestaltet werden. Spatial relationships between root length density of Coffea arabica (coffee RLD) and soil nutrient- related factors at plot scale in a coffee- Erythrina poeppigiana system was studied by geostatistics. In a 24 × 29 m area, (organic and conventional management), coffee and Erythrina fine roots and soil chemical properties were sampled on an irregular grid in the topsoil. A factor analysis explained 83 % of the total variation of the soil attributes. Soil factors were identified: Chemical fertility (CF), Micronutrients, Organic matter, and Acidity (Ac). Based on the spherical model, all the attributes presented a strong spatial structure. The scale of spatial correlation for CF was lesser than for Ac, but similar to coffee RLD. Erythrina RLD had a short-range variation. Patchy areas of high spots of coffee RLD were greater in organic plot. Cross-semivariogram analysis estimated a correlation between soil factors and coffee RLD over a scale of 5.50 m; but 4.23 m with Erythrina RLD. Nutrients linked to P, Zn, exchangeable bases and acidity soil affected the scale of spatial aggregation pattern of coffee RLD. The spatial response of coffee RLD suggests a differential nutrient uptake strategy for acquiring soil nutrients induced by the quality of organic and inorganic fertilizer inputs. The fact that coffee RLD had higher scale of spatial variation than Erythrina RLD and a negative spatial correlation indicate that pruned Erythrina trees are not so competitive for acquiring shared nutrients in an agroforestry system.  相似文献

It is difficult to obtain fossil data from the 10% of Earth's terrestrial surface that is covered by thick glaciers and ice sheets, and hence, knowledge of the paleoenvironments of these regions has remained limited. We show that DNA and amino acids from buried organisms can be recovered from the basal sections of deep ice cores, enabling reconstructions of past flora and fauna. We show that high-altitude southern Greenland, currently lying below more than 2 kilometers of ice, was inhabited by a diverse array of conifer trees and insects within the past million years. The results provide direct evidence in support of a forested southern Greenland and suggest that many deep ice cores may contain genetic records of paleoenvironments in their basal sections.  相似文献

Iron(V)-oxo species have been proposed as key reactive intermediates in the catalysis of oxygen-activating enzymes and synthetic catalysts. Here, we report the synthesis of [Fe(TAML)(O)]- in nearly quantitative yield, where TAML is a macrocyclic tetraamide ligand. Mass spectrometry, M?ssbauer, electron paramagnetic resonance, and x-ray absorption spectroscopies, as well as reactivity studies and density functional theory calculations show that this long-lived (hours at -60 degrees C) intermediate is a spin S = 1/2 iron(V)-oxo complex. Iron-TAML systems have proven to be efficient catalysts in the decomposition of numerous pollutants by hydrogen peroxide, and the species we characterized is a likely reactive intermediate in these reactions.  相似文献

Tick-borne encephalitis (TBE) in animals is not well understood yet. TBE virus (TBEV) serology in several host species could be valuable for epidemiological analyses in the field as well as for the detection of clinical cases. However, performance and suitability of the available test systems are not well assessed. Therefore, we evaluated two commercial TBEV-ELISA kits in a pilot study and compared them for their suitability in veterinary applications. For this purpose, we tested 163 field collected goat sera and evaluated the results by serum neutralization test (SNT) as "gold standard". Twenty-eight SNT positive sera (17.2%) were detected. The best suited ELISA kit was used for determination of a species-specific cutoff for horses, cattle, sheep, goats, pigs, mice, dogs, rabbits and monkeys with defined sera from animals without known or with improbable contact to TBEV. The level of non-specific ELISA results does not only differ between animal species but may also be influenced by the age of the tested animals. The number of sera which tested false positive by ELISA was higher in older than in young sheep. In order to obtain defined polyclonal sera as references, two dogs, cattle, goats, sheep, rabbits and pigs each, as well as one horse and 90 mice were immunized four times with a commercially available TBEV vaccine. In conclusion, our results demonstrated that commercial TBEV-ELISA kits are suitable for application in veterinary medicine for both, verification of clinical TBE cases and epidemiological screening. However, positive ELISA results should be verified by SNT. Only a very low number of false negative ELISA-results were found.  相似文献

In 2009, a novel swine-origin H1N1 influenza A virus (S-OIV), antigenically and genetically divergent from seasonal H1N1, caused a flu pandemic in humans. Development of an effective vaccine to limit transmission of S-OIV in animal reservoir hosts and from reservoir hosts to humans and animals is necessary. In the present study, we constructed and evaluated a vectored vaccine expressing the H1 hemagglutinin of a recent S-OIV isolate using equine herpesvirus 1 (EHV-1) as the delivery vehicle. Expression of the recombinant protein was demonstrated by immunofluorescence and western blotting and the in vitro growth properties of the modified live vector were found to be comparable to those of the parental virus. The EHV-1-H1 vaccine induced an influenza virus-specific antibody response when inoculated into mice by both the intranasal and subcutaneous routes. Upon challenge infection, protection of vaccinated mice could be demonstrated by reduction of clinical signs and faster virus clearance. Our study shows that an EHV-1-based influenza H1N1 vaccine may be a promising alternative for protection against S-OIV infection.  相似文献
