A large outbreak of canine salmonellosis in military watch dogs due to Salmonella Montevideo and/or Salmonella Give was only recognized through a monitoring program, implemented by the military forces. In 51 out of 80 exposed dogs (63.8%) from four kennels Salmonella was isolated from fecal samples. The prevalence per sampling day ranged from 5.6 to 77.8%. Clinical disease with mild diarrhea but without fever appeared only in nine dogs (11.3% of study population, 16.9% of cases) from one kennel (***P < 0.001). Risk analysis identified "dog food A" (odds ratio 3.33 [1.12 < OR < 10.06] *P < 0.05) and "dog food C" (odds ratio 3.40 [1.07 < OR < 11.22] *P < 0.05), two commercial dehydrated dog feeds, as suspected infectious sources. Feeding "dog food C" alone or together with "dog food A" was significantly associated with clinical disease (***P < 0.001). S. Montevideo and S. Give with similar plasmid profiles and PFGE-restriction patterns were isolated from the suspected dog feeds and fecal samples. This outbreak demonstrates not only that Salmonella infections in dogs occur without clinical symptoms but also that large outbreaks occur after feeding dehydrated dog feeds. Especially in households with known risk group patients this finding demonstrates a particular risk for their owners, as they often live in close contact with their animals.  相似文献

In February 2007, sweet orange trees with characteristic symptoms of huanglongbing (HLB) were encountered in a region of S?o Paulo state (SPs) hitherto free of HLB. These trees tested negative for the three liberibacter species associated with HLB. A polymerase chain reaction (PCR) product from symptomatic fruit columella DNA amplifications with universal primers fD1/rP1 was cloned and sequenced. The corresponding agent was found to have highest 16S rDNA sequence identity (99%) with the pigeon pea witches'-broom phytoplasma of group 16Sr IX. Sequences of PCR products obtained with phytoplasma 16S rDNA primer pairs fU5/rU3, fU5/P7 confirm these results. With two primers D7f2/D7r2 designed based on the 16S rDNA sequence of the cloned DNA fragment, positive amplifications were obtained from more than one hundred samples including symptomatic fruits and blotchy mottle leaves. Samples positive for phytoplasmas were negative for liberibacters, except for four samples, which were positive for both the phytoplasma and 'Candidatus Liberibacter asiaticus'. The phytoplasma was detected by electron microscopy in the sieve tubes of midribs from symptomatic leaves. These results show that a phytoplasma of group IX is associated with citrus HLB symptoms in northern, central, and southern SPs. This phytoplasma has very probably been transmitted to citrus from an external source of inoculum, but the putative insect vector is not yet known.  相似文献

Synchronization of the oestrous cycle of gilts using altrenogest treatment has been found to increase ovulation rate. The current experiment investigated if the increase in ovulation rate after altrenogest treatment is related to increased follicle size at the end of altrenogest treatment compared with late luteal phase follicles. Crossbred gilts (n = 15) received altrenogest during 18 days [20 mg Regumate (Janssen Animal Health, Beerse, Belgium)], starting 5-7 days after onset of first oestrus. Control gilts (n = 15) did not receive altrenogest. At days 10-12 of the oestrous cycle [i.e. in the presence of corpora lutea (CL)], average follicle development was 2.51 +/- 0.20 mm (assessed with ultrasound) in altrenogest-treated gilts and 2.58 +/- 0.16 mm in control gilts (p > 0.10). During the last days of altrenogest treatment (i.e. when CL had gone into regression), average follicle size had increased to 3.01 +/- 0.31 mm (p < 0.05). Subsequent ovulation rate was 16.6 +/- 1.7 in altrenogest treated gilts and 15.1 +/- 1.2 in control gilts (p < 0.05). Altrenogest treatment resulted in increased follicle size after regression of the CL, showing that suppression of follicle growth by altrenogest alone is less severe than suppression by endogenous progesterone (either with or without altrenogest). Altrenogest treatment also resulted in a higher ovulation rate. However, it is unclear if the increased follicle size and higher ovulation rate after altrenogest treatment are causally related, as the relation between the two on an animal level was not significant.  相似文献

The effect of an extract of Yucca schidigera on the control and infection process of the apple scab pathogen, Venturia inaequalis, was examined and compared with the chemical resistance inducer, acibenzolar-S-methyl (ASM). In seedling assays, both materials significantly reduced apple scab symptoms and pathogen sporulation on leaves and both showed similar control efficacies as the reference treatment, sulphur. Whereas yucca extract and sulphur gave significant inhibition of conidial germination in vitro, ASM did not inhibit germination. Histopathological studies of the infection process of V. inaequalis in apple leaves showed that the yucca extract primarily acted by inhibiting pre-penetration events and penetration itself. In contrast, the ASM treatment significantly inhibited more stages of the infection process (pre-penetration, penetration and post-penetration events). These observations suggest that the yucca extract acted mainly by a direct fungitoxic effect whereas ASM, as expected, acted as a resistance inducer. However, expression studies of two genes encoding the PR proteins, PR1 and PR8, in apple seedlings indicated that yucca extract may also affect plant defence as expression of both genes was up-regulated following yucca treatment, to a level similar to that observed after treatment with ASM. The fungitoxic effect of sulphur on V. inaequalis was also confirmed in this study.  相似文献

We report picosecond time-resolved x-ray diffraction from the myoglobin (Mb) mutant in which Leu29 is replaced by Phe (L29Fmutant). The frame-by-frame structural evolution, resolved to 1.8 angstroms, allows one to literally "watch" the protein as it executes its function. Time-resolved mid-infrared spectroscopy of flash-photolyzed L29F MbCO revealed a short-lived CO intermediate whose 140-ps lifetime is shorter than that found in wild-type protein by a factor of 1000. The electron density maps of the protein unveil transient conformational changes far more dramatic than the structural differences between the carboxy and deoxy states and depict the correlated side-chain motion responsible for rapidly sweeping CO away from its primary docking site.  相似文献

To study the influence of potassium (K) fertilizer rate on soil test K values, crop yield, and K-leaching in sandy soils, four long-term fertilizer experiments (0–60–120–180 kg K ha?1 a?1) were initiated in 1988 in northern Germany on farmers fields. Clay content of the plow layer was about 4%, and organic matter between 2% and 5%. Plant available soil K was estimated with the double lactate (DL) method. Small grain cereals (rye and barley) did not respond to K fertilization in the 7-year period even though the soil test value of the K-0 plots decreased from ca. 90 to ca. 30 mg KDL kg?1 within 3 years. This value remained almost constant thereafter. Crop removal (including straw) of 75 kg K ha?1 a?1 was therefore apparently supplied from nonexchangeable K fractions. Compared to the optimum, no K application reduced the yield of potato by up to 21%, and that of white sugar yield up to 10%. Maximum potato yield was obtained by annually applying 60 kg K ha?1 which resulted in a test value of 60 mg KDL kg?1 soil. Maximum potato yield was also obtained at 40 mg KDL kg?1 soil, however, with a single application of 200 kg K ha?1. Similar results were obtained with sugar beet. This indicates that for maximum yield, even for K demanding crops, it is not necessary to maintain KDL values above 40 mg K kg?1 soil throughout the entire crop rotation. Soil test values increased roughly proportional to the K fertilizer level. About 120 kg fertilizer K ha?1 a?1, markedly more than crop K removal, was required to maintain the initial KDL of 90 mg kg?1. The K concentration of the soil solution in the top soil measured after harvest was increased exponentially by K fertilizer level and so was K leaching from the plow layer into the rooted subsoil. The leached quantity increased from 22 kg K ha?1 a?1 in the plot without K application to 42.79 and 133 kg Kha?1 a?1 in plots supplied with 60, 120 and 180 kg K ha?1 a?1 respectively. Soil test values around 100 mg KDL kg?1 on sandy soils, as often found in the plow layer of farmers fields, lead to K leaching below the root zone that may exceed the critical K concentration of 12 mg K T?1 for drinking water.  相似文献
