Quantification of microbial activities involved in soil organic carbon (SOC) decomposition is critical for the prediction of the long-term impact of climate change on soil respiration (SR) and SOC stock. Although the temperature sensitivity of SR is especially critical in semi-arid regions, such as North West Tunisia, where the SOC stock is low, little research has been carried out in these environments. More needs to be known about factors, such as SOC availability that influence temperature sensitivity. In this study, soil samples were incubated with and without glucose addition for 28 days after a 28-day pre-incubation period. Pre-incubation and incubation was carried out at 20 °C, 30 °C, 40 °C and 50 °C. Respiration measurements were taken with temperature, glucose addition and incubation time as independent variables. The highest pre-incubation temperature reduced the temperature sensitivity of SR during the subsequent incubation period, both with and without glucose addition. Soil samples pre-incubated at 50 °C had the lowest SR at all subsequent incubation temperatures and the lowest temperature sensitivity of SR, even after glucose addition. However, after glucose addition, the effect of a high pre-incubation temperature on soil respiration lasted only two days. Measuring the water-soluble carbon (WSC) in soil samples suggested that the high pre-incubation temperature may have killed part of the microbial biomass, modified microbial communities or solubilized SOC. For quantifying the possible effect of global warming, in particular heat waves, on soil respiration in the soil studied, the results indicate a moderate response of soil respiration to temperature at high temperatures, as shown by Q10 close to 1.7, even in the range 40-50 °C.  相似文献

This study elucidates the aetiology and epidemiology of monilia disease of quince caused by the fungus Monilinia linhartiana in Spain. Disease incidence and the dynamics of apothecial development and ascospore discharge were quantified and the pathogen was characterized using morphological and molecular methods. The pathogen did not produce conidia or apothecia on agar media but produced conidia on leaves showing symptoms and apothecia on mummified young quince fruit. Monilinia linhartiana was not pathogenic on ripe quince fruit but was readily isolated from developing, mummified fruit (pseudosclerotia). Phylogenetic analysis based on 5·8S‐ITS region sequences placed M. linhartiana in the Disjuntoriae section of Monilinia species infecting rosaceous hosts. Studies during 2004–2008 in four commercial orchards in southern Spain determined two major infection periods for the disease. The first coincided with the unfolding of the first leaves and resulted in leaf blotch and shoot blight. The second coincided with flowering and led to mummification of developing young fruit. Foliar infection was apparently initiated by airborne ascospores produced on pseudosclerotia that overwintered on the soil surface, while flower infection was probably initiated by conidia produced on leaf lesions. Incidence of diseased shoots ranged from 1 to 91% and was correlated with calculated inoculum potential, based on the density and maturity of apothecia formed on pseudosclerotia. This epidemiological study has made it possible to characterize the life cycle of monilia disease on quince in southern Spain, which will help the development of new control strategies.  相似文献

Pesticides have been used in agriculture to enhance food production by eradicating unwanted insects and controlling disease vectors, nevertheless occupational exposure to high levels of these compounds can lead to neurodegenerative disorders, characterized by serious oxidative and neurotoxic effects. However, there is a lack of consensus as to which determinations are best used to quantify future risks arising from xenobiotic exposure and natural antioxidant interventions. Our study aims to determine the potential ability of selenium and/or vitamin E, used as nutritional supplements, to alleviate oxidative stress in cerebral cortex tissue induced by dimethoate, an organophosphorus pesticide. Adult Wistar rats were exposed either to dimethoate (0.2 g/L of drinking water), dimethoate + selenium (0.5 mg/kg of diet), dimethoate + vitamin E (100 mg/kg of diet), or dimethoate + selenium + vitamin E, for 30 days. Exposure to dimethoate increased malondialdehyde levels, protein carbonyl groups and advanced oxidation protein products, while Na+K+-ATPase, acetylcholinesterase and butyrylcholinesterase activities decreased in the cerebral cortex. An increase in glutathione peroxidase, superoxide dismutase and catalase activities and a decrease in glutathione, non-protein thiols and vitamin C levels were observed. Administration of selenium and/or vitamin E through the diet in dimethoate treated rats ameliorated the biochemical parameters cited above. The histological findings confirmed the biochemical results. The model of this study that we employed characterized the relationships between dimethoate-induced neurotoxicity and its alleviation by natural antioxidants like selenium and vitamin E. These elements may be considered beneficial for the protection of cerebral cortex against injury induced by dimethoate.  相似文献

ObjectiveTo determine the pharmacokinetics and pharmacodynamics of the neurosteroidal anaesthetic, alfaxalone, in horses after a single intravenous (IV) injection of alfaxalone, following premedication with acepromazine, xylazine and guaiphenesin.Study designProspective experimental study.AnimalsTen (five male and five female), adult, healthy, Standardbred horses.MethodsHorses were premedicated with acepromazine (0.03 mg kg?1 IV). Twenty minutes later they received xylazine (1 mg kg?1 IV), then after 5 minutes, guaiphenesin (35 mg kg?1 IV) followed immediately by IV induction of anaesthesia with alfaxalone (1 mg kg?1). Cardiorespiratory variables (pulse rate, respiratory rate, pulse oximetry) and clinical signs of anaesthetic depth were evaluated throughout anaesthesia. Venous blood samples were collected at strategic time points and plasma concentrations of alfaxalone were assayed using liquid chromatography-mass spectrometry (LC/MS) and analysed by noncompartmental pharmacokinetic analysis. The quality of anaesthetic induction and recovery was scored on a scale of 1–5 (1 very poor, 5 excellent).ResultsThe median (range) induction and recovery scores were 4 (3–5) (good: horse slowly and moderately gently attained recumbency with minimal or no rigidity or paddling) and 4 (1–5) (good: horse stood on first attempt with some knuckling and ataxia) respectively. The monitored cardiopulmonary variables were within the range expected for clinical equine anaesthesia. The mean ± SD durations of anaesthesia from induction to sternal recumbency and from induction to standing were 42.7 ± 8.4 and 47 ± 9.6 minutes, respectively. The mean ± SD plasma elimination half life (t1/2), plasma clearance (Clp) and volume of distribution (Vd) for alfaxalone were 33.4 minutes, 37.1 ± 11.1 mL minute?1 kg?1 and 1.6 ± 0.4 L kg?1, respectively.Conclusions and clinical relevanceAlfaxalone, in a 2-hydroxypropyl-beta-cyclodextrin formulation, provides anaesthesia with a short duration of recumbency that is characterised by a smooth induction and satisfactory recovery in the horse. As in other species, alfaxalone is rapidly cleared from the plasma in the horse.  相似文献

State-and-transition models (STMs) are being developed for many areas in the United States and represent an important tool for assessing and managing public and private rangelands. Substantial resources have been invested in model development, yet minimal efforts have been made to evaluate the utility of STMs for rangeland assessment and management. We interviewed 47 rangeland professionals, equally divided between managers and researchers, in four ecoregions to determine their perceptions of the purpose, development, and strengths and weaknesses of STMs to assess the status of the STM framework. Our analysis identified three primary perspectives regarding the purpose of STMs: a decision-making tool for land managers, a means to represent the complex dynamics of rangeland ecosystems, and an effective communication tool. These diverse views of STM purposes were associated with differing perspectives concerning model development that identified five major issues in need of further development and refinement: 1) the relative importance of management practices and ecological processes in driving transitions, 2) the criteria used to define thresholds, 3) the appropriate level of model complexity, 4) the respective roles of expert knowledge and ecological data in model development, and 5) processes for model review and revision. We recommend greater dialogue among researchers and managers to further clarify STM terminology and develop standard protocols for model development and validation. Mechanisms are critically needed to assure peer review and revision of existing models so that STMs are continually updated to reflect current understanding of rangeland dynamics.  相似文献

The aim of this work was to explore leaf characteristics underlining the difference in the sensitivity of pea cultivars (cv. Kelvedon, Douce and Lincoln) to Fe deficiency. Plants were grown in a greenhouse under controlled conditions in continuously aerated solution. Three treatments were used: 30 μM Fe (+Fe), 0 μM Fe (−Fe); direct deficiency and 30 μM Fe + 10 mM NaHCO3 (+Fe+Bic); indirect deficiency for 12 days. Growth parameters, iron status, potassium content, chlorophyll fluorescence and photosynthetic capacity were studied. Our results showed that Fe deficiency led to a significant decrease of chlorophyll index (SPAD readings) and bivalent iron content in all Pisum sativum cultivars. The lower reduction was observed in Fe-deficient plants of Kelvedon and Douce. In addition, shoot length and whole plant dry weight were not affected by Fe deficiency in the latter cultivars. Both tolerant cultivars showed higher accumulation of potassium content in their leaves compared with the sensitive one. Moreover, both chlorophyll fluorescence ratios (Fv/Fm and Fv/F0) were significantly decreased in all cultivars under both Fe deficiency treatments. The photosynthetic electron transport activity was reduced in the sensitive cultivar, especially in the absence of iron. The adverse effect of bicarbonate-induced Fe deficiency on the above mentioned parameters were more pronounced than that of the direct one. The capacity of both tolerant cultivars to preserve adequate chlorophyll synthesis, photosynthetic capacity and plant growth under iron-limiting conditions is related to the suitable nutrition of their leaves in ferrous iron, due to (at least partially) their higher potassium content.  相似文献

Barbarine sheep is the dominant breed in Tunisia. This fat-tailed breed present in all production systems is characterised by metabolic and digestive adaptation to the contrasting environment conditions prevailing in the country (heat stress, water deprivation, salinity etc.). The fat tail (1.5 to 7 kg) is an energy reservoir that is used in periods of feed scarcity. Moreover, the rumen of this breed harbours a microflora enabling it to valorize low-quality roughages and native range vegetation. Barbarine sheep could make benefit from a wide range of local feed resources like fodder shrubs (e.g. cactus and oldman saltbushes), tannin-containing shrubs (e.g. Acacia cyanophylla) and agro-industrial by-products (e.g. olive cake, bran etc. conserved in the form of feed blocks or pellets). It has a very shallow anoestrous, the intensity of which is dependant upon nutrition conditions rather than day length as temperate breeds. Productive and reproductive performances of late pregnant–early suckling, pre-mating ewes and also rams of this breed are not affected when some alternative feed resources replace common feedstuffs which are expensive and cannot afford regularly to smallholders. In brief, the merits of the Barbarine sheep in the production systems and other main adaptive mechanisms of this breed are discussed in this paper. The prospective of Barbarine sheep husbandry in the system dynamics, climate change and the evolution of the socioeconomic context are also envisaged.  相似文献

Proteases have a significant role in the life cycle of parasites and the pathogen-host relationship, being regarded as important virulence factors. In the parasitic nematode Hysterothylacium aduncum proteolytic activity was measured during in vitro development from third larval stage (L3) to mature adult, using DQ red casein as a fluorogenic substrate. Proteolytic activity was detected in all the developmental stages studied and at all pH values within the range employed (2.0-7.5). The assay with specific inhibitors permitted the determination of metalloprotease activity, and, to a lesser extent, that of aspartate- and cysteine-protease. Serine-protease activity was the lowest of those studied. In L3 recently collected from the host fish (L3-0 h), the greatest activity was found at an optimum pH of 4.0 and was mainly inhibited by 1,10-phenathroline (metalloprotease inhibitor). This metalloprotease activity in L3-0 h (infective stage) may be related to the invasion of the host tissues by this larva. In the other developmental stages, the greatest protease activity was found at pH 5.5, although at pH 4.0 a lower activity peak was detected. On the other hand, our data show that the proteolytic activity of the nematode varies according to the presence of pepsin (an aspartic-protease) in the culture medium. Thus, at pH 4.0, activity was greater in the absence of pepsin, with increasing aspartic-protease activity. Together with the detection of aspartic-, cysteine- and metallo-protease (enzymes involved in digestion in invertebrates) in all the developmental stages of the parasite taking place in the digestive tract of the host fish, this allows us to suggest that the pepsin in the culture medium mimics the predigestion conditions in the habitat of the worm within the host and that the activity detected may have, amongst others, a digestive function.  相似文献
