The amino acid composition and the physicochemical and functional properties of quinoa protein isolates were evaluated. Protein isolates were prepared from quinoa seed by alkaline solubilization (at pH 9, called Q9, and at pH 11, called Q11) followed by isoelectric precipitation and spray drying. Q9 and Q11 had high levels of essential amino acids, with high levels of lysine. Both isolates showed similar patterns in native/SDS-PAGE and SEM. The pH effect on fluorescence measurements showed decreasing fluorescence intensity and a shift in the maximum of emission of both isolates. Q9 showed an endotherm with a denaturation temperature of 98.1 degrees C and a denaturation enthalpy of 12.7 J/g, while Q11 showed no endotherm. The protein solubility of Q11 was lower than that of Q9 at pH above 5.0 but similar at the pH range 3.0-4.0. The water holding capacity (WHC) was similar in both isolates and was not affected by pH. The water imbibing capacity (WIC) was double for Q11 (3.5 mL of water/g isolate). Analysis of DSC, fluorescence, and solubility data suggests that there is apparently denaturation due to pH. Some differences were found that could be attributed to the extreme pH treatments in protein isolates and the nature of quinoa proteins. Q9 and Q11 can be used as a valuable source of nutrition for infants and children. Q9 may be used as an ingredient in nutritive beverages, and Q11 may be used as an ingredient in sauces, sausages, and soups. 相似文献

Florida ranks first in citrus production, with nearly 68% of all U.S. citrus growing in the season 2005-2006. Most of the citrus groves are located from central to south Florida, and agricultural irrigation permitting is regulated by three of Florida's five water management districts. Most of the permitting for citrus production in Highlands, Polk and Hillsborough counties is conducted by the Southwest Florida Water Management District (SWFWMD), and quantities are based on the District's AGMOD computer program. In 2003, the SWFWMD implemented new permit criteria so that permitted amounts were more representative of actual water use. This paper compares grower reported citrus irrigation water use in Highlands, Polk and Hillsborough counties from 1994 through 2005 with permitted and theoretical irrigation requirements calculated by a daily water balance. Two different sets of crop coefficients (Kc's) developed for citrus in Florida were compared in the daily soil water balance calculation of theoretical irrigation requirements. The percentage of irrigated area considered in this study ranged from 40 to 60% to simulate a range of grower practices. Meteorological data from two weather stations and additional rainfall information from 50 locations within the three counties was used in the water balance. Missing and error values in the meteorological historical record data were filled with weather generators. The multiannual average water consumption (including cold protection water use) from growers ranged from 243 (Hillsborough) to 406 mm (Highlands) and the multiannual average permitted irrigation requirement (without cold protection) ranged from 295 to 557 mm. The simulated gross irrigation requirements under different scenarios of location-Kc-wetted area were variable but mostly lower than the limits established by the district, except for some scenarios in Polk County, whose maximum simulated irrigation value reached 578 mm year−1. This article contributes to the literature on firm demography and regional development in at least three different ways. First, consumption, rather than employment, which is the most common variable seen in literature, is used to measure the impact of firm demography on regional development. Second, while the literature is mainly focused on the relationship between new business formation and regional development, we investigate both entry and exit flows of firms. Third, we decompose each of these flows into spatial and sectoral components. The empirical investigation looks at the Italian regions with reference to the period 2004–2009. Ashy stem blight (ASB) caused by Macrophomina phaseolina (Tassi) Goidanich (Mp) is a devastating seed-transmitted disease in common bean in the tropics. The identification of resistant cultivars throughout the cropping season contributes to disease management. Resistance is found in the primary and tertiary gene pools. Our objectives were to determine (1) the reaction of Phaseolus spp. genotypes to two Mp isolates at vegetative and reproductive stages, (2) the area under disease progress curve (AUDPC), and (3) resistant plants per genotype at harvest. Twenty-three genotypes from different origins were planted in the greenhouse in 2016 and 2017. One less-aggressive Mp (PRJD16) and one more-aggressive (PRI16) isolate were inoculated one and three times, respectively, by the cut-stem method. ‘Beníquez’, ‘Othello’, and ‘Verano’ were highly susceptible (mean scores >8.0, and AUDPC values from 264.6 to 300.8) to both isolates. BAT 477 and NY6020-4 were intermediate (5.6 and 6.2; AUDPC: 161.6 and 187.1) to PRJD16 and susceptible (7.4 and 8.2; AUDPC: 209.4 and 235.1) to PRI16. Resistant genotypes (mean scores ≤3) were not identified in this study. However, A 195, ‘Badillo’, and ‘PC 50’ possessed lower mean scores (4.3–5.4) and AUDPC values (126.4–150.9) to both isolates. Furthermore, A 195 had the highest percentage of resistant plants (55.6%) followed by PC 50, I9365-31, and PI 321637 (27.8%) to PRJD16 at harvest. Thus, the identification of resistant parents across Phaseolus species is necessary to increase the levels of ASB resistance in common bean cultivars throughout the entire cropping season. 相似文献

Forty-one apple samples, representing eight of the most widely cultivated varieties in western Europe, were collected in Trentino, Italy. Samples were extracted from fresh fruit with a mixture of acetone/water to achieve a good extraction of polyphenols, including proanthocyanidin oligomers which were analyzed by normal-phase HPLC. Up to 20 compounds including catechin, epicatechin, B2 procyanidin, hydroxycinnamates, flavonols, anthocyanins, and dihydrochalcones were analyzed by reversed-phase HPLC and LC-MS. Total polyphenol content was independently measured with an optimized Folin-Ciocalteu assay. The mean content of total polyphenols lay between 66.2 and 211.9 mg/100 g of FW depending on the variety. With chromatographic analysis, it was possible to explain the whole amount of total polyphenols measured by the FC assay. Flavanols (catechin and proanthocyanidins) are the major class of apple polyphenols (71-90%), followed by hydroxycinnamates (4-18%), flavonols (1-11%), dihydrochalcones (2-6%), and in red apples anthocyanins (1-3%). 相似文献

A novel electronic nose based on solid-phase microextraction (SPME) coupled with a surface acoustic wave (SAW) sensor array has been used to analyze different quality virgin olive oils. A mathematical model was designed with 37 samples to distinguish lampante from the other virgin olive oils categories (extra-virgin and virgin), because lampante-virgin olive oils cannot be consumed without a previous refining process. The model, successfully validated with a test set of 16 samples, was able to classify 90% of the samples correctly. Eight volatile products characterizing strawberry aroma, which is generated from the oxidative degradation of linoleic and linolenic acids by a lipoxygenase (LOX) pathway, were examined because of their antifungal activity against Colletotrichum acutatum, one of the causal agents of strawberry anthracnose. In this study, the effects of aldehydes, alcohols, and esters on mycelial growth and conidia development were evaluated. (E)-Hex-2-enal was found to be the best inhibitor of mycelial growth [MID (minimum inhibitory doses)=33.65 microL L(-1)] and of spore germination (MID=6.76 microL L(-1)), while hexyl acetate was the least effective of all volatile compounds tested (MID=6441.89 microL L(-1) for mycelial growth and MID=1351.35 microL L(-1) for spore germination). Furthermore, the antifungal activity of (E)-hex-2-enal on susceptibility of strawberry fruits to C. acutatum was also confirmed. The presence of these molecules in jars containing strawberry fruits inoculated with a suspension of spores inhibited the fungus growth and prevented the appearance of symptoms. Moreover, a study of the effects of (E)-hex-2-enal on conidial cells of C. acutatum was evaluated by transmission electron microscopy. This volatile compound altered the structures of the cell wall and plasma membrane, causing disorganization and lysis of organelles and, eventually, cell death. 相似文献
