Arabinoxylans (AX), xylanase, and xylanase inhibitors have an important role in many cereal food processing applications. The effects of genotype, growing location, and their interaction (G × L) on AX, apparent xylanase activity, and apparent xylanase inhibition activity of Triticum aestivum xylanase inhibitor (TAXI) and xylanase inhibiting protein (XIP) were investigated for six hard red and six hard white spring wheat genotypes grown at three locations. Difference in total AX level among genotypes was not determined to a significant level by genotype. Instead, variability in total AX content was largely dependent on G × L. However, total AX content was significantly different between the two wheat classes. For bran xylanase activity, 25% of the variability could be attributed to G × L interaction. Moreover, there was significant difference between the bran xylanase activities in the two wheat classes. Bran TAXI activity and XIP activity were significantly different among genotypes. Genotype contributed 72% to the variability in TAXI activity and 39% in XIP. However, no significant difference was observed among the two wheat classes for TAXI or XIP activity. These results indicate that TAXI might be a stable parameter in segregating wheat genotypes with varying xylanase activity.  相似文献

In dogs, secondary polycythemia (SP) may be associated with polyuria and polydipsia (PU/PD). The pathogenesis of this PU/PD has not yet been explained. We hypothesized that hyperviscosity and increased blood volume in SP might affect vasopressin (VP) release, resulting in PU/PD. This hypothesis was tested in 2 dogs with SP caused by renal neo-plasia and PU/PD. Osmoregulation of VP release was studied by a modified water deprivation test and by investigating the VP response to hypertonic saline infusion.
Water deprivation test results were consistent with an inability to produce concentrated urine despite increasing plasma osmolality. During hypertonic saline infusion, the osmotic threshold of VP release was markedly increased in both dogs, resulting in a delayed VP response to increasing plasma osmolality. The sensitivity of VP release was low normal in both dogs. We conclude that blood hyperviscosity and increased blood volume led to impaired VP release and polyuria.  相似文献

In 19 wheat-milling fractions total pentosan content, calculated as 0.88 x (% L-arabinose + % D-xylose), varied between 1.44 and 30.66% on dry matter (dm). It increased with ash content once the latter exceeded 0.6% (dm basis). Water-extractable arabinoxylans were recovered by saturating water extracts to 65% ethanol. Their contents in the milling fractions varied between 0.35 and 1.38%, and above 0.6% ash content also increased with this parameter. Their L-arabinose-to-D-xylose ratios ranged between 0.65 and 0.39, with the lowest values found for the fractions with highest ash content, indicating that the ash-rich tissues contain more arabinoxylans that are less branched. (1)H NMR spectroscopy revealed that the decrease in L-arabinose-to-D-xylose ratio was accompanied by an increase in unsubstituted xylose residues and a decrease in disubstituted xylose residues, while the contents of monosubstituted xyloses were virtually constant.  相似文献

Strains of fungi from different origins, including isolates of the natural microflora of barley, were screened for their ability to modify barley starchy endosperm cell walls in situ. In an initial step, fungi were selected that degrade the major component of the cell walls, that is, (1-->3),(1-->4)-beta-D-glucan, in vitro on artificial media. Nongerminating, sterilized barley, obtained by gamma-irradiation, was inoculated with such fungi and subjected to solid state fermentation under conditions resembling those of a traditional malting process. For some strains of fungi, a clear correlation between the production of endo-beta-glucanase and the friability of the treated kernels was found. Image analysis of Calcofluor stained longitudinal sections of barley kernels fermented with the endo-beta-glucanase producing strains showed that starchy endosperm cell walls were modified. As malt quality is inversely related to its (1-->3),(1-->4)-beta-D-glucan content, the selected strains have high potential to be used as starter cultures during malt production, contributing to the processing quality of the final product.  相似文献

Pseudomonas CMR12a was previously selected as an efficient biocontrol strain producing phenazines and cyclic lipopeptides (CLPs). In this study, biocontrol capacity of Pseudomonas CMR12a against Rhizoctonia root rot of bean and the involvement of phenazines and CLPs in this ability were tested. Two different anastomosis groups (AGs) of Rhizoctonia solani, the intermediately aggressive AG 2-2 and the highly aggressive AG 4 HGI, were included in growth-chamber experiments with bean plants. The wild-type strain CMR12a dramatically reduced disease severity caused by both R. solani AGs. A CLP-deficient and a phenazine-deficient mutant of CMR12a still protected bean plants, albeit to a lesser extent compared with the wild type. Two mutants deficient in both phenazine and CLP production completely lost their biocontrol activity. Disease-suppressive capacity of CMR12a decreased after washing bacteria before application to soil and thereby removing metabolites produced during growth on plate. In addition, microscopic observations revealed pronounced branching of hyphal tips of both R. solani AGs in the presence of CMR12a. More branched and denser mycelium was also observed for the phenazine-deficient mutant; however, neither the CLP-deficient mutant nor the mutants deficient in both CLPs and phenazines influenced hyphal growth. Together, results demonstrate the involvement of phenazines and CLPs during Pseudomonas CMR12a-mediated biocontrol of Rhizoctonia root rot of bean.  相似文献

Endoxylanases seriously affect the rheological properties of durum wheat (Triticum durum Desf.) semolina spaghetti doughs prepared with, and as evaluated, by the farinograph. Under the experimental conditions, control doughs (34.9% moisture content) made from two semolinas (semA and semB) yielded a maximal consistency of 525 and 517 farinograph units (FU), with, respectively, 19.4 and 16.4% of the total level of arabinoxylans (TOT-AX) being water-extractable (WE-AX). When 75.4 Somogyi units/50 g of semolina of the endoxylanases from Trichoderma viride (XTV), rumen microorganisms (XRM), Bacillus subtilis (XBS), and Aspergillus niger (XAN) were used, the maximal consistencies at 34.9% moisture decreased for semA to 467, 436, 448, and 417 FU, respectively. This was accompanied by increased WE-AX contents of 60.8, 71.2, 70.7, and 73.0%, respectively. Similar results were observed for semB. By reducing the total water content of doughs, it was possible to recover the maximal consistency of the original doughs. Both the decrease in maximal consistency and the amount of water to be omitted were significantly related to the decrease in molecular weight (MW) of the WE-AX and the percentage of WE-AX solubilized as a result of the enzymic action. At the same time, it was clear that endogenous endoxylanase inhibitors were present in the durum wheat semolinas and that they inhibited the endoxylanases used to different degrees. Part of the differences in effects between the different endoxylanases (decrease in maximal consistency, amount of AX solubilized, MWs of the WE-AX, and amount of water that could be omitted) could be ascribed to the differences in inhibition of the endoxylanases by endogenous inhibitors.  相似文献

The relationship between syruping in refrigerated doughs upon prolonged storage and different aspects of arabinoxylan (AX) hydrolysis was investigated using Triticum aestivum xylanase inhibitor (TAXI) and different xylanases in the dough formula. Dough characteristics were evaluated with strong emphasis on the AX population and its fate as a function of storage time. Selective reduction of part of the flour endogenous xylanase activity in dough by added TAXI reduced dough syruping after 12 and 20 days of storage by 50%, providing straightforward evidence for the involvement of xylanases and, thus, AX in the syruping phenomenon. Addition of xylanases with different inhibitor sensitivities [an inhibition-sensitive Bacillus subtilis xylanase (XBS(i)) as well as a noninhibited mutant (XBS(ni)) thereof] to dough confirmed the importance of xylanases in dough syruping, on one hand, and the power of wheat flour TAXI to constitute a significant barrier against xylanase-mediated dough syruping, on the other hand. Use of xylanases with different substrate selectivities [an Aspergillus aculeatusxylanase (XAA) versus XBS(ni)] showed degradation of water-extractable AX (WE-AX) and solubilized AX to low molecular weight molecules rather than the conversion of water-unextractable AX (WU-AX) to high molecular weight water extractable components to be the main factor influencing dough syruping.  相似文献

Different concentrations (1.2-3.6%) of maltodextrin preparations with average degrees of polymerization (DP) varying between 4 and 66 reduced the differential scanning calorimetry (DSC) staling endotherm in baked and stored (7 days, 23 degrees C) bread doughs from 3.4 mJ/mg to values within a 3.0-1.9 mJ/mg range. Commercial enzymes used in industrial practice as antistaling agents for bread also reduced amylopectin retrogradation. This suggested that the maltodextrins used are promising antistaling components and that the staling of bread and amylopectin retrogradation are related phenomena. In addition, the results obtained suggest that starch hydrolysis products resulting from enzymic attack may well be responsible for the antistaling effect induced by antistaling enzymes.  相似文献

Zusammenfassung Von 380 Winter- und 1400 Sommergersten des Sortiments Gatersleben wurde in Halle das Verhalten gegen die virulente Rasse 24 vonPuccinia striiformis West, untersucht. Als hochresistent erwiesen sich 5 Winter- und 43 Sommerformen. Ein großer Teil dieser Sommergersten, darunter auch wertvolle Kombinationen von Gelbrost- mit Zwergrost- bzw. Mehltauresistenz, stammen aus Aethiopien. Resistenz zeigten weitere 88 Proben oder ca. 5% des Sortiments. In 12% lagen Gemische anfälliger mit resistenten Pflanzen vor. 80% der Gersten waren mäßig bis stark anfällig für Gelbrost.
Summary Stripe rust reaction of 380 winter and 1400 spring barley varieties of the Gatersleben collection was tested in the greenhouse as well as in the disease nursery by artificial infection with the virulent race 24 ofPuccinia striiformis West. Five winter and 43 spring barley varieties have proved to be highly resistant. A large number of the latter were collected in Ethiopia, among them are useful combinations of stripe rust with dwarf rust resistance and mildew resistance, respectively. Resistance was shown by other 88 varieties or by about 5% of the total. Susceptible as well as resistant plants were found in 12% of the samples. 80% of the varieties were moderately or highly susceptible to stripe rust.
