In a natural environment, seminal plasma provides spermatozoa with protection against reactive oxygen species. Storing semen in cooling conditions requires diluting it with various buffer solutions. Therefore, the protective role of seminal plasma is not sufficient enough. Semen obtained from five male specimens was diluted with the Kobayashi buffer solution at a 1:9 ratio. To determine the influence of antioxidants on semen storage, a buffer solution was used, as before, with the addition of 1 % albumin, 1 mM vitamin C, 1.5 mg ml?1 vitamin E, 5 mM sodium citrate, 5 mM glutathione and 5 mM cysteine. After the preparation of such tests, the parameters of spermatozoa motility were measured every 3–5 days, using the CASA system (Image House CRISMAS Company Ltd.). Among all used antioxidants, the best effects were observed after the addition of glutathione to semen. After 17 days of storage, the percentage of motile spermatozoa in the samples preserved with glutathione addition was 57 %, while without antioxidant addition, it was 44 %. Furthermore, the addition of cysteine and albumin also resulted in the lengthening of the life span of perch sperm cells. The presence of the remaining antioxidants (vitamins C and E, and sodium citrate) did not have any positive influence on spermatozoa viability, and in these samples, no motile spermatozoa were observed after 12 days of storage. Our data show that dilution of perch sperm with buffered solution might be a promising method for short-term storage.  相似文献

The study encompasses three reproduction seasons. Tench spawners caught during the spawning season originated from carp ponds (domestic stock) and a lake (wild stock). Fish were reproduced under controlled conditions after hormonal stimulation with GnRHa-containing pellets combined with metoclopramide (Ovopel) or carp pituitary homogenate (CPH). As a result of hormonal stimulation, eggs were obtained from a larger number of females originating from the lake (71.7%) than those originating from the pond (58.3%), although no other statistical differences were found. A similar relationship was recorded for the spermatozoa motilities (range from 72 to 76%). The obtained results indicate that both investigated reservoirs are suitable for tench broodstock management due to the fact that synchronization of ovulation among different stocks is easy to achieve. For this purpose, among the tested spawning agents, Ovopel could be recommended as being slightly more effective.  相似文献

Studies that provide representative insights for determining yield through its related traits during the ontogeny of modern cultivars subjected to sources of environmental variation are limited for different crops, including wheat. Most of the empirical evidence on the relationships between the yield of small grain cereal crops and its contributing traits has been obtained under dry or semidry conditions. The aims of this paper were to (1) illustrate how an path analysis can be used to clarify and interpret the relationships between grain yield (GY), yield components, and other yield-related traits of 25 winter wheat cultivars subjected to sources of environmental variation and (2) determine how the yield-related traits contribute to the yield variation. The data used in this analysis were generated from multi-environment trials across wheat-growing areas in Poland. Using Ward’s clustering procedure was capable of identifying the most critical predictor traits of the yield components and their contributions to cultivar-focused GY variation. Our findings document, confirm, and improve the basic biological understanding of how to grow modern wheat cultivars for high GY through effectively stimulating the improvement of yield-related traits through the optimization of developmental stage-based agronomic strategies. Our results confirmed empirically that modern European wheat cultivars grown in a temperate climate require favorable conditions, the use of appropriate N fertilizer and growth regulators, and the application of fungicide to protect against leaf diseases and to provide conditions that effectively increase the time to anthesis, the Leaf Area Index per spike at anthesis, and the grain filling duration, and reduce plant height and flag leaf disease severity, thus leading to a high GY. A high yield level is obtained by the performance of preferred yield-related traits that can maintain the three yield components at relatively high levels.  相似文献

Soil testing is used to help make fertilizer recommendations for greater yields and profits. But the increase of soil‐sampling density raises costs of sample collection and analyses. The aim of this study was to compare grid‐cell sampling densities (1, 2, and 4 ha) in terms of the estimation accuracy of macronutrients (P, K, Mg) availability and pH and to investigate how sampling density affects the amount of fertilizers and lime recommended and correctly applied to winter wheat (Triticum aestivum L.). The distribution of liming requirements and available nutrients were quite similar for the 1‐ and 2‐ha grids but notably different for the 4‐ha grid. However, the whole‐field average values of pH and P, K, and Mg concentrations in soil obtained for different sampling densities were very similar, thus placing, respectively, the soil of the studied area in the same class of liming needs and nutrient availability. The range and estimation errors of these parameters decreased with sampling‐grid size increase. The amount of lime and fertilizers to be applied on the field and the portion of a field correctly limed or fertilized depended on the soil chemical property considered. If one treats the 1‐ha grid as the reference and the most correct soil‐sampling approach, 2‐ha grid offered the greatest part of the field to be adequately fertilized with lime, P, and K. However, fertilization with Mg was much more appropriate if the recommendation was based on 4‐ha, than on a 2‐ha soil‐sampling grid. To gain an insight into soil variation and soil process occurring at small scale, laboratory and geostatistical analyses on individual soil samples may be necessary in some cases. Possibly, such costly research can deliver relevant information which could be then applied into farmer's practice.  相似文献

The aim of the study was to optimize burbot juveniles rearing in recirculating aquaculture system. In experiment 1 (17 °C, photoperiod 24L:0D), the fish (initial: body weight [W] = 15.36 ± 3.72 g, standard length [SL] = 12.48 ± 1.09 cm) were divided into four groups (I, II, III and IV). Different feeding levels were applied: 1, 2, 3 and 4 % of biomass daily (counted based on dry feed weight). The feed conversion ratio (FCR) and specific growth rate (SGR) were recorded. In experiment 2 (17 °C, feeding level of 2 % of biomass day?1), fish (W = 5.24 ± 2.43 g and SL = 8.54 ± 1.24 cm) were divided into two groups where different light conditions were applied (I: 24 h light [1,800 lx] and II: 24 h darkness [4 lx]). In experiment 1, the highest SGR was recorded in group II (1.93 % day?1), whereas the lowest SGR (1.27 % day?1) and final W (P < 0.05) was in I group. The lowest (P < 0.05) FCR (0.63) was in group II. In the remaining groups, FCR was similar (0.68–0.70, P > 0.05). The feed consumption in group I reached 100 %, in group II, it was 71.3 % (P < 0.05) and it was the lowest in groups III (39.26 %) and IV (36.93 %). In experiment 2 no differences in the growth and survival rate were recorded (final SL between 14.16 and 14.19 cm, P > 0.05; W between 23.33–23.35 g; P > 0.05). The results from experiment 1 indicate that the feeding 2 % of biomass day?1 was the most efficient. Also, it was proven, for the first time, that there was no effect of using different constant light conditions.  相似文献

Different Shewanella species are isolated both from healthy and from diseased fish. To date, contemporary methods do not provide sufficient insight to determine species and detail differentiation between tested strains. Bacteria isolated from cultured (n = 33), wild (n = 12) and ornamental (n = 6) fish, as well as several reference strains, were tested by 16S rRNA gene sequencing, ERIC‐PCR and pulsed‐field gel electrophoresis (PFGE) assays. Our study indicates that isolates collected from freshwater fish were genetically diverse. Based on 16S rRNA gene sequences, bacteria were clustered into groups S. putrefaciens, S. xiamenensis and S. oneidensis. Some isolates were classified only to genus Shewanella; thus, 16S rRNA gene analyses were not enough to determine the species. ERIC‐PCR revealed 49 different genotype profiles indicating that the method might be useful for differentiation of Shewanella isolates irrespectively to species identification, contrary to PFGE which is not suitable for Shewanella typing.  相似文献

Milt of the Leuciscus idus L. was collected from five experimental groups, and selected parameters of its quality were analysed for 36 h (group II), 60 h (group III), 84 h (group IV) and 108 h (group V), respectively, after hormonal stimulation with Ovopel (1 granule kg?1 of body weight). The control (group I) fish were not subjected to hormonal stimulation. The highest milt volume was obtained from the fish in group IV (0.70 ± 0.55 mL), where the largest volume of milt expressed per kilogram was also obtained (3.03 ± 1.94 mL kg?1). Significant differences were also found in milt volumes obtained between group I and groups III (P<0.01) and IV (P<0.05). The highest percentage of motile spermatozoa was found in the milt of group IV (59%); significant differences were found between group I and groups II (P<0.01) and III (P<0.001). The value of osmotic pressure of seminal plasma was the highest in group IV (203.19 ± 37.63 mOsm kg?1), and the lowest in group I (118.31 ± 41.13 mOsm kg?1). Parameters determining milt quality and quantity indicate that the period of 60–84 h after hormonal stimulation with Ovopel is optimal for obtaining milt from ide.  相似文献

Abstract

The Kujawy and Pomorze Province with 127 rural and municipal-and-rural communes in 19 districts is a typical agricultural region, with the highest plant farming intensity in Poland. The diversity and dominance of crop plantations in the agroecosystems in communes were calculated based on percentage share of 21 kinds of crops in the cropland structure. The algorithms applied in ecological research of biodiversity were used: the Shannon–Weaver diversity index (H’) and Simpson's dominance index (λ). The diversity (H’) of crops in arable land in the Kujawy and Pomorze Province ranges from 1.68 to 2.54. The diversity of crop plantations in the region was bigger in the communes in which there are: better soil, a higher share of arable land in the agricultural acreage and cropland in the arable land as well as with a higher share of barley, corn for grain and sugar beet in the cropland structure. The highest dominance index (λ) for crops in the whole province is 0.291. The index of dominance of agricultural crops in this region increases with an increase in the share of: woodland in the total area, meadows and pasture in the agricultural acreage and rye in the cropland structure.  相似文献
