The objectives of this study were to investigate the relationships between body weight and morpho-structural indices to predict body weight from their orthogonal body shape characters using principal component analysis and to morphologically classify the chicken genotypes using multivariate discriminant analysis. Data used were from 273 randomly selected 12-weeks-old indigenous chickens of normal-feathered (NF), frizzle-feathered (FF), naked-neck (NN) and Anak Titan (AT) genotypes. Phenotypic correlation among body weight and most biometric traits ranged from 0.227-0.876, -0.7-0.901, 0.034-0.968 and -0.207-0.849 for NF, NN and AT chickens, respectively. Factor analysis with varimax rotation of interrelated traits revealed three principal components which accounted for 83.1%, 74.4%, 78.8% and 76.5% of the total variance in NF, FF, NN and AT chickens in the order listed. Breast girth, keel length, thigh length, shank length and wing length were found to be the most discriminating variables to separate the chicken genotypes. The longest distance (72.54) occurred between AT and NF genotypes while the shortest distance (4.27) was recorded for FF and NN genotypes. Classification results showed that 85.2% of AT genotype was correctly classified into their source population. However, 22.7% of NF was misclassified as NN, while 33.3% of NN was misclassified as NF chickens. These results suggest that there is high rate of gene flow between these two indigenous chicken genotypes. Information obtained from this study may be considered useful in breed improvement programmes for selection, characterization, conservation and better management of Nigerian indigenous chickens. 相似文献

The automated analysis of total white blood cell count and white blood cell differentials is routine in research and clinical diagnosis in mammalian species. In contrast, in avian haematology these parameters are still estimated by conventional microscopic procedures due to technical difficulties associated with the morphological peculiarities of avian erythrocytes and thrombocytes. Both cell types are nucleated and fairly resistant to cell lysis, a prerequisite for automated leukocyte quantification and differentiation by commercial instruments. By using an anti-CD45 monoclonal antibody in combination with selected subset specific markers we have established a simple (no-lyse no-wash single-step one-tube) flow cytometry based technique for high precision chicken blood cell quantification. EDTA-blood samples are diluted, spiked with fluorescence beads and incubated with a mixture of fluorochrome conjugated chicken leukocyte specific antibodies. We demonstrate that total leukocyte numbers as well as thrombocyte, monocyte, T-cell, B-cell and heterophilic granulocyte numbers can be determined by flow cytometry in a single step without prior cell lysis, cell separation or cell washing steps. Importantly, we also show that blood samples can be fixed prior to cell staining which enables shipping of samples making the technology widely available. Comparison of this technique with conventional microscopy revealed superior precision. By comparing leukocyte differentials of two chicken populations and during immune system development after hatch we demonstrate that large sample numbers can be analysed within hours. This technique will help to overcome previous restrictions in immune status analysis in chickens in experimental systems, during vaccine testing and health status monitoring in chicken flocks. Advances in avian genomics should facilitate the development of appropriate tools for other avian species in the future which will make this technique broadly applicable. 相似文献

Objective-To assess joint kinematics in dogs with osteoarthritis of the hip joints during walking up an incline or down a decline and over low obstacles and to compare findings with data for nonlame dogs. Animals-10 dogs with osteoarthritis of the hip joints (mean ± SD age, 6.95 ± 3.17 years; mean body weight, 34.33 ± 13.58 kg) and 8 nonlame dogs (3.4 ± 2.0 years; 23.6 ± 4.6 kg). Procedures-Reflective markers located on the limbs and high-speed cameras were used to record joint kinematics during walking up an incline or down a decline and over low obstacles. Maximal flexion, extension, and range of motion of the hip joints were calculated. Results-Osteoarthritis of the hip joints reduced extension of both hip joints and flexion of the contralateral hind limb, compared with flexion of the lame hind limb, during walking down a decline. Walking up an incline resulted in decreased extension of the stifle joint in both hind limbs of osteoarthritic dogs; extension was significantly decreased for the lame hind limb. During walking over low obstacles, maximal flexion of the stifle joint was increased significantly for the contralateral hind limb. Maximal flexion was increased in both tarsal joints. Conclusions and Clinical Relevance-Osteoarthritis of the hip joints led to complex changes in the gait of dogs, which involved more joints than the affected hip joint alone. Each exercise had specific effects on joint kinematics that must be considered when planning a rehabilitation program. 相似文献

Predicting the impacts of selection for decreased faecal egg count (FEC) (i.e. host resistance) in grazing ruminants is difficult, due to complex interactions between parasite epidemiology, management and host responses. A mathematical model including heritable between lamb variation in host-parasite interactions, Teladorsagia circumcincta epidemiology and anthelmintic drenching, was developed and used to (i) address such interactions and their impact on outcomes including FEC, live weight (LW, kg) and pasture larval contamination (PC, larvae/kg DM), and (ii) investigate how grazing management strategies, aimed at reducing host exposure to infective larvae via pasture moves at 40 day intervals, affect these outcomes. A population of 10,000 lambs was simulated and resultant FEC predictions used to assign the 1,000 lambs with the highest and lowest predicted FEC to 'susceptible' (S) and 'resistant', (R) groups, respectively. The predicted average FEC of the S group was ~8.5-fold higher than the R group across a grazing season. The R and S groups were then simulated to graze separate pastures (R(sep) and S(sep)); and repeated for 3 grazing seasons to allow predictions to diverge and stabilize. Further, different grazing strategies were superimposed on all groups. PC and average FEC were affected by whether lambs of different resistance genotype grazed together or separately, with differences increasing across grazing seasons. By the third grazing season the average PC of the R(sep) group was reduced by ~83%, and the S(sep) group was increased by ~240%, in comparison to the whole population average. Average FEC of the R(sep) group was reduced by ~40%, and the S(sep) group increased by ~46% in comparison to the R and S groups, respectively, whilst drenching had little impact on the proportional differences in FEC between groups. Predicted LW was similar for the R and R(sep) groups irrespective of anthelmintic treatment, whilst LW of the S(sep) group was reduced by ~14% compared to the S group for un-drenched lambs, and by ~4% for drenched lambs. The differing grazing strategies were predicted to have little impact on FEC or LW, with the exception of the S(sep) group which was predicted to have a 2kg increase in LW when drenched and moved to a clean pasture. Together, these results suggest that host genotype has a substantial impact on parasite epidemiology, however the benefits of anthelmintic treatment and grazing management should only be expected for susceptible animals. Hunting is the major driver of large mammal decline in Central African forests. In slowly reproducing species even low hunting pressure leaves spatial gradients with wildlife density increasing with distance from transport routes and human settlements. Park management can use this pattern formation to identify sources of threats, but also to discriminate between different threat scenarios, such as the impact of subsistence vs. commercial hunting. We conducted an ape survey in the mountainous Moukalaba Doudou National Park, Gabon, to evaluate whether potential population gradients would emanate from the three human population centers in the region or the villages surrounding the park. Using generalized linear modeling we found hill slope as a good predictor of ape nest occurrence probability and the distance to human population centers a better predictor of ape nest density and ape nest group size than distance to villages. In fact ape nest density was three times lower at the park borders close to the human population centers than in the park’s interior. The results indicate that Moukalaba’s ape population is more impacted by commercial than subsistence hunting and suggest that park management should focus conservation efforts on the human population centers. We conclude that in particular for slowly reproducing species geographic information on wildlife population gradients are of additional value for guiding protected area management. The hunting impact on those species might be easily underestimated, if derived only from market surveys or transport route controls, where they are only rarely found. 相似文献

High levels of metals impede plant growth by affecting physiological processes. Siderophores are microbial Fe-chelators that, however, bind other metals. This study evaluated plant growth in a soil containing elevated levels of metals, including Al, Cu, Fe, Mn, Ni, and U, using Streptomyces-derived cell-free supernatant containing siderophores and auxins. Cowpea plants in the soil were treated with the culture filtrate. Growth was measured and biochemical analyses such as chlorophyll contents, RNA and protein quantification, lipid membrane peroxidation, and anti-oxidative responses were conducted to evaluate oxidative stress in the plants. Liquid chromatography-mass spectrometry was used to simulate competition for siderophore binding, and metal content of plants was determined spectroscopically. Whereas the metals inhibited plant growth, addition of siderophores improved growth. There was evidence of lipid peroxidation, an enhanced superoxide dismutase activity, and lowered chlorophyll, RNA, protein, carotenoid and residual indole acetic acid contents, especially in control plants. Siderophore competition assays between Al and Fe, and Fe and Cu suggested that trivalent metals are more competitive for siderophore binding than divalent ones. Compared to control plants, higher amounts of metals were obtained in siderophore-treated plants. Siderophores were able to supply plants with Fe in the presence of levels of metals, mainly Al, Cu, Mn, Ni and U that otherwise inhibit Fe acquisition. This led to enhanced chlorophyll content, circumventing lipid peroxidation effects on leaves. Siderophores lowered the formation of free radicals, thereby protecting microbial auxins from degradation and enabling them to enhance plant growth which in turn resulted in augmented metal uptake. 相似文献

Sampling was conducted at seven sites on, and at varying distances adjacent to, the Island of Montréal (Québec, Canada), and as far as 1,100 km away in Northern Québec, to explore the hazard potential of snowpacks in remote, rural, and urban environments. Ecotoxic effects of melted snow were ascertained with a suite of small-scale bioassays representing several aquatic taxonomic groups (bacteria, micro-algae, micro-invertebrates, fish liver cells) as well as with biomarker measurements determined with a rainbow trout primary hepatocyte (RTPH) assay. Bioassays undertaken with the cnidarian Hydra attenuata and RTPH cell assays, and to a lesser extent with the micro-alga Pseudokirchneriella subcapitata, proved particularly sensitive to infer the presence of bio-available pollutants in snow samples collected from all sites, thereby suggesting their contamination (at least) via atmospheric sources. Furthermore, biomarker responses indicated that snow samples presumably included metals (free Zn biomarker), organics (CYP 1A1 biomarker), estrogens (alkali-labile phosphate biomarker) as well as chemicals capable of causing oxidative stress (LPO biomarker), depending on the site being considered. Overall, effects data acquired during this preliminary investigation on the ecotoxicity of snowpacks submit that adverse impact toward aquatic biota is conceivable at some sites during spring meltdown. Because snow has a recognized affinity for sequestering solids and contaminants of atmospheric origin, future studies aimed at identifying sources and chemicals implicated in observed effects are legitimate endeavors. 相似文献

It is often claimed that agroforestry could increase the total productivity per land unit compared to monocropping systems. The aim of this study was to evaluate, in a sub-humid Mediterranean climate, the behavior of the yield components, phenology, LAI and NDVI of durum wheat in an alley-cropping system. Our hypothesis was that the microclimate changes in agroforestry could change the devlelopment and yield of cereals. Two different experiments were carried out: in 2015 under 16-year old poplars in East–West lines and in 2016 under 21-year-old ash trees in North–South lines. In each experiment, 12 genotypes of durum wheat were sown. The grain yield was not significantly different in agroforestry and full sun conditions in 2015; however, both systems in this experiment had a particularly low yield (≈ 10% of the historical average yield of the plot). In 2016, the grain yield was significantly lower in agroforestry in comparison with full sun conditions. In both experiments, the most impacted yield component by agroforestry was the number of grains per spike. Similarly, in both experiments, the number of grains per spike was the only yield component impacted by the position within the alley inside agroforestry. Surprisingly, in 2016 the grain yield was higher in the West than in the center position of the alley. In both experiments, agroforestry delayed the maturity of the crop. The use of standard growing degree days was not sufficient to explain the difference in phenology between agroforestry and full sun conditions. 相似文献
