Plant functional types (PFT) have been used to describe the response of native vegetation to environmental factors (i.e., fertility) and to livestock disturbance, but rarely under conditions of continuous grazing. In this work we investigate whether the long-term response of grassland communities submitted to a gradient of continuous grazing pressure can be described with such an approach. After 15 yr of differentiation of the grazing pressure applied to native grasslands we measured leaf dry-matter content (LDMC) and specific leaf area (SLA) of Poaceae populations of the communities. A grazing pressure gradient was created by levels of daily forage allowance: 4, 8, 12, and 16 kg of dry matter per day per 100 kg of animal live weight, monitored monthly. PFTs were defined by numerical analysis, where an algorithm finds the optimal trait subset based on the agreement between matrices of species × traits, paddocks × grass biomass, and environmental variables (levels of forage allowance and soil characteristics). The results show that it is possible to describe a gradient of grazing pressure by means of LDMC and/or SLA measured only on the Poacea contributing at least 80% of the total Poaceae biomass. Four PFTs were differentiated by these leaf traits. PFTs having low LDMC and high SLA are characteristic of high intensity of use and are made up largely of stoloniferous C4 species typical of rapid resource capture strategies. Conversely, PFTs characterized by high LDMC and low SLA include species that are representative of low grazing pressure. Variations in the aggregate value of traits are due to changes in the species proportions and not to leaf-size adaptation as hypothesized. We conclude than in the absence of a gradient of fertility, plants with strategies of resource capture tend to be more represented under high grazing pressures. This situation results in a loss of functional diversity, but in particular a reduction in forage availability, which is incompatible with high animal production.  相似文献

We investigated color preference, seasonal abundance, spatial distribution and species composition of thrips in northern highbush blueberries, Vaccinium corymbosum L., in New Jersey (USA). White sticky traps were more attractive to thrips compared with yellow or blue traps. Thrips captures using white sticky traps showed that their flight activity begins 20–30 d after the onset of flowering, with 10, 50 and 90% of trap captures observed at 383, 647 and 1231 degree-day accumulations, respectively (10 °C base temperature). Two methods were used to study thrips distribution within a blueberry bush. First, white sticky traps were placed within the bush canopy at three different heights. The highest numbers of thrips were caught on traps in the middle and top one-third of the canopy while the lowest numbers were caught in the bottom one-third. A second method determined the distribution of thrips on the blueberry plant at different heights and phenological stages. The highest numbers of thrips were found on young leaves at lower parts of the canopy, whereas flowers and fruit had fewer thrips and none were found on buds; these thrips were identified as, Scirtothrips ruthveni (88% of adults) and Frankliniella tritici (12%). The distribution of thrips within a blueberry planting was investigated using an evenly-spaced grid of white sticky traps in combination with on bush beating-tray samples. Thrips counts from traps correlated with direct counts on the bush across the entire blueberry field (macro-scale level); however, within the field (micro-scale level), there was no correlation between the number of thrips on traps and on individual bushes near traps. Early in the season, trap counts were higher on bushes closer to the forest, indicative of movement of thrips from wild hosts into blueberry fields. However, this was not the case for direct on bush counts or trap counts for the later part of the season, where there was no clear forest “edge” effect. Percent fruit injury due to thrips feeding was low, and it correlated with thrips counts on bushes but not from counts on traps. Overall, our data show that thrips counts on sticky traps need to be interpreted with care because these numbers weakly correlated with the numbers of thrips on bushes at the micro-scale level and percent fruit injury; however, they can be useful predictors of thrips activity across entire blueberry fields (macro-scale).  相似文献

Efficient selection of potato varieties with enhanced nutritional quality requires simple, rapid, accurate, and cost-effective assays to obtain tuber chemical composition information. Our objective was to develop simple protocols to determine phenolics, anthocyanins, and antioxidant capacity in polyphenolic extracts of potatoes using Fourier transform infrared spectroscopy combined with multivariate techniques. Lyophilized potato samples (23) were analyzed. Polyphenolic compounds were extracted from potatoes and applied directly applied onto a three-bounce ZnSe crystal for attenuated total reflectance measurements in the infrared region of 4000 to 700 cm (-1). Robust models were generated (r > or = 0.99) with standard error of cross-validation values of 4.17 mg gallic acid equivalent/100 g (total phenolics), 0.87 mg pelargonidin-3-glucoside/100 g (monomeric anthocyanins), and 130.8 mumol Trolox equivalent/100 g (antioxidant capacity) potato powder. In addition, classification models discriminated potato samples at the species and variety level. Application of a simple infrared spectroscopic protocol allowed simultaneous rapid quantification of specific nutritional components in potatoes and efficient selection of value-added potato varieties.  相似文献

This study aims to describe macronutrient and boron deficiencies in pepper grown in a hydroponic system in the presence of a substrate during the vegetative and fruiting phases. The experiments were carried out into a greenhouse, and consisted of a randomized block design with eight treatments: complete solution (control) and nitrogen (-N), phosphorus (-P), potassium (-K), calcium (-Ca), magnesium (-Mg), sulfur (-S) and boron (-B) deficient solutions, with four repetitions for each assay. Visual diagnoses of certain macronutrient and boron deficiencies can be applied by farmers to aid in soil management, since they are observed in leaves, while N, P, Ca, S and B deficiencies led to roots symptoms. Pepper fruits showed nutritional deficiency symptoms when grown under N, K, Ca, Mg, S and B deficits. The appearance of nutrient deficiency symptoms in pepper plants occurs in the following order: N, K, P, Mg, S, Ca and B.  相似文献

Nowadays, the therapeutic efficiency of small interfering RNAs (siRNA) is still limited by the efficiency of gene therapy vectors capable of carrying them inside the target cells. In this study, siRNA nanocarriers based on low molecular weight chitosan grafted with increasing proportions (5 to 55%) of diisopropylethylamine (DIPEA) groups were developed, which allowed precise control of the degree of ionization of the polycations at pH 7.4. This approach made obtaining siRNA nanocarriers with small sizes (100–200 nm), positive surface charge and enhanced colloidal stability (up to 24 h) at physiological conditions of pH (7.4) and ionic strength (150 mmol L−1) possible. Moreover, the PEGylation improved the stability of the nanoparticles, which maintained their colloidal stability and nanometric sizes even in an albumin-containing medium. The chitosan-derivatives displayed non-cytotoxic effects in both fibroblasts (NIH/3T3) and macrophages (RAW 264.7) at high N/P ratios and polymer concentrations (up to 0.5 g L−1). Confocal microscopy showed a successful uptake of nanocarriers by RAW 264.7 macrophages and a promising ability to silence green fluorescent protein (GFP) in HeLa cells. These results were confirmed by a high level of tumor necrosis factor-α (TNFα) knockdown (higher than 60%) in LPS-stimulated macrophages treated with the siRNA-loaded nanoparticles even in the FBS-containing medium, findings that reveal a good correlation between the degree of ionization of the polycations and the physicochemical properties of nanocarriers. Overall, this study provides an approach to enhance siRNA condensation by chitosan-based carriers and highlights the potential of these nanocarriers for in vivo studies.  相似文献

The effects of prescribed burning on nutrient cycling were studied in a stand of oocarpa pine (Pinus oocarpa Schiede) in central Honduras. Small paired runoff plots were subjected to a low-intensity prescribed burn in one of two seasons and nutrient losses in runoff water and sediment were monitored together with nutrient inputs in bulk precipitation.Burning increased the loss of nutrients and net losses of calcium, potassium and magnesium were recorded during the first 1612 months after burning. Increased rates of sediment loss rather than increased losses in surface runoff were responsible for the greater part of the increase in total nutrient losses from the burned plots. Greater quantities of nutrients were lost from the plots burned just prior to the dry season than from those burned after the start of the wet season because of slow recovery of the ground vegetation during the dry season and exposure to the intense rains of May and early June. Recovery from burning was rapid and runoff and sediment loss from the burned plots approached the levels of controls 2 years after burning. As nutrient losses are a reflection of total runoff and sediment losses, it is probable that no net nutrient losses would have been detected during the 2nd year after burning had sampling been continued. The net nutrient losses recorded were small and this is attributed to the low initial fuel loadings, low fire intensities and the rapid rate of recovery of the ground vegetation. The significance of other potential sources of losses and gains of nutrients is discussed and it is concluded that low-intensity prescribed burning for hazard reduction, with burning cycles ranging from three to seven or more years, will reduce the rate of nutrient accumulation on the site rather than deplete the nutrient capital.  相似文献

Juveniles of Pinctada mazatlanica and Pteria sterna were collected with artificial collectors from Falsa Bay, Baja California Sur, Mexico during a 12 month experimental period. Filament substrate artificial collectors contained in 2 mm plastic mesh bags (25 ± 25 cm) were deployed from the surface to 17 m of depth, and replaced monthly. Pinctada mazatlanica had the greatest recruitment (12 juveniles/cm2) in late summer and P. sterna (5 juveniles/cm2) in winter. Maximum spat collection from September 1986 to April 1987 occurred at 10 m depth for P. mazatlanica and at 4–7 m depth for P. sterna . However, peak spatfall under 11m depth occurred from late July to August 1987. The mean prodissoconch shell height was 254 μ m (SE = 0.003; N = 50) for P. mazatlanica and 313 μm (SE = 0.004; N = 50) for P. sterna . The growth equations for the first five weeks of both species, based on the prodissoconch shell height and the maximum spat height (measured from the umbo to distal edge) are presented.  相似文献

Evaluating phenotypic diversity makes it possible to identify discrepancies in aptitudes among animals of different genetic bases, which is an indicator of adaptive or selective differences between populations. The objective of this work was to evaluate the morphofunctional diversity of 452 male and female adult equines (Arabian, Quarter Mile, Pantaneiro, and Criollo breeds, and undefined crossbreeds of horses and mules) raised in the Pantanal biome (Brazil). Linear measurements were performed to estimate conformation indexes. Initially, a discriminant analysis was performed, regardless of the animal’s size, followed by factor analysis. The factors were characterized and used as new variables. The diversity among equines and their relationship with the factors were evaluated using multivariate analysis. The factors were classified according to their decreasing importance: balance, rusticity, and robustness for the measurement factors; and load, ability, conformation, and equilibrium for the index factors. The genetic groups of equines have well-defined morphofunctional characteristics. The main differences are based on the rusticity and ability typologies in relation to those based on performance. Equines introduced to the Pantanal biome presented a more robust and compact body with good conformation. As a result, these horses may have superior athletic performance during equestrian activities when compared to the Pantaneiro local breed. However, this biotype may represent less rusticity (less adaptive capacity). Therefore, the regional breed can be equal or better in equestrian activities than breeds introduced to the Pantanal biome. Thus, breeders may cross horses from local breeds as an alternative to those introduced. Undefined crossbred male equines presented a different profile from the Pantaneiro breed, which may indicate little use of crossbreeds in breeding.  相似文献

Nine horses received 20 mg/kg of intravenous (LEVIV ); 30 mg/kg of intragastric, crushed immediate release (LEVCIR ); and 30 mg/kg of intragastric, crushed extended release (LEVCER ) levetiracetam, in a three‐way randomized crossover design. Crushed tablets were dissolved in water and administered by nasogastric tube. Serum samples were collected over 48 hr, and levetiracetam concentrations were determined by immunoassay. Mean ± SD peak concentrations for LEVCIR and LEVCER were 50.72 ± 10.60 and 53.58 ± 15.94 μg/ml, respectively. The y ‐intercept for IV administration was 64.54 ± 24.99 μg/ml. The terminal half‐life was 6.38 ± 1.97, 7.07 ± 1.93 and 6.22 ± 1.35 hr for LEVCIR , LEVCER , and LEVIV , respectively. Volume of distribution at steady‐state was 630 ± 73.4 ml/kg. Total body clearance after IV administration was 74.40 ± 19.20 ml kg?1 hr?1. Bioavailability was 96 ± 10, and 98 ± 13% for LEVCIR and LEVCER , respectively. A single dose of Levetiracetam (LEV ) was well tolerated. Based on this study, a recommended dosing regimen of intravenous or oral LEV of 32 mg/kg every 12 hr is likely to achieve and maintain plasma concentrations within the therapeutic range suggested for humans, with optimal kinetics throughout the dosing interval in healthy adult horses. Repeated dosing and pharmacodynamic studies are warranted.  相似文献

The acquisition of equine oocyte developmental capacity is ensured by the follicular environment, such as granulosa cells, which could reflect the meiotic development potential of immature oocytes. This study evaluated the relationship between DNA fragmentation of granulosa cells, using the chromatin dispersion test, and equine oocyte meiotic development after in vitro maturation. Granulosa cells and cumulus–oocytes complexes (n = 50) were recovered from slaughterhouse‐derived ovaries. Oocytes were in vitro matured, stained and evaluated under fluorescence microscopy. Maturation rates were classified into outstanding, medium and poor levels of maturation using 25th and 75th percentiles as thresholds. For DNA assessment, each sample was processed with the Ovoselect® kit (Halotech DNA). High, low and total DNA fragmentation percentages were compared among levels of maturation rates by ANOVA, followed by Duncan test. Results were expressed as mean ± SE. Total and high DNA fragmentation rates of granulosa cells were significantly higher (p < 0.05) in follicles whose oocytes had reached outstanding maturation level than those originating from follicles whose oocytes had reached poor maturation level. In conclusion, the DNA fragmentation analysis of equine granulosa cells can be a valuable test to identify equine oocytes showing the best meiotic competence after in vitro maturation.  相似文献
