Burrowing nematodes (Radopholus similis) are among the most serious nematode pests affecting banana and plantain (Musa spp.). In Uganda, bananas, which are known locally as “matooke”, are the main staple. Radopholus similis populations collected in four banana-growing locations (Namulonge, Mbarara, Ikulwe and Mukono) were cultured monoxenically on carrot discs, and we compared the variability in reproductive fitness and virulence (as a function of time and inoculum level) of different populations of R. similis from Uganda. Their level of pathogenicity was determined by assessing the nematode reproductive ratio; that is, final population divided by the initial population. These in vitro experiments showed that the R. similis population from Mbarara had the highest reproduction ratio, while the population from Mukono had the lowest reproduction ratio. This assessment along with pathogenicity experiments on host Musa plants provides a means for defining pathogenicity groups among R. similis populations. 相似文献

Ceramide engagement in apoptotic pathways has been a topic of controversy. To address this controversy, we tested loss-of-function (lf) mutants of conserved genes of sphingolipid metabolism in Caenorhabditis elegans. Although somatic (developmental) apoptosis was unaffected, ionizing radiation-induced apoptosis of germ cells was obliterated upon inactivation of ceramide synthase and restored upon microinjection of long-chain natural ceramide. Radiation-induced increase in the concentration of ceramide localized to mitochondria and was required for BH3-domain protein EGL-1-mediated displacement of CED-4 (an APAF-1-like protein) from the CED-9 (a Bcl-2 family member)/CED-4 complex, an obligate step in activation of the CED-3 caspase. These studies define CEP-1 (the worm homolog of the tumor suppressor p53)-mediated accumulation of EGL-1 and ceramide synthase-mediated generation of ceramide through parallel pathways that integrate at mitochondrial membranes to regulate stress-induced apoptosis. 相似文献
Nematode pathogenicity experiments on Musa host plants were carried out in pot trials. Both the final nematode population densities and percentages root necrosis on different host plants were higher for the Radopholus similis population from Mbarara than for the populations from Namulonge, Ikulwe and Mukono (Uganda). The R. similis population from Mbarara managed to break the resistance of the diploid banana Pisang Jari Buaya, a known worldwide source of resistance to R. similis. The diploid banana hybrid TMB2× 9128-3 and the triploid dessert banana Yangambi km5 showed resistance against the four R. similis populations. These results indicate that differences in pathogenicity among different R. similis populations exist and should be taken into consideration for plantain and banana germplasm enhancement. 相似文献
Two new 5-methylcoumarin glycosides named diosfeboside A (1) and B (2) and five known compounds namely kaempferol 3-O-α-l-rhamnopyranosyl-(1→2)-β-d-glucopyranoside (3), ursolic acid (4), betulinic acid (5), stigmasterol (6) and stigmasterol 3-O-β-d-glucopyranoside (7) were isolated from the leaves of Diospyros crassiflora (Hiern). Their structures were established through interpretation of 1 and 2D NMR, mass spectra analysis and comparison with reported data. In vitro cytotoxic activity of the new compounds against human carcinoma cell lines (HL-60, Bel-7402, BGC-823, and KB) was evaluated and no cytotoxicity was observed for each of them. 相似文献
The prevention of Ungulate protoparvovirus 1 (UPV1) infection and consequently the reproductive losses is based on vaccination of all pigs intended for breeding. As maternally derived antibodies (MDA) can interfere with the development of immunity following vaccination, it is important to know the duration of anti-UPV1 MDA to determine the optimal age for the best vaccination efficacy. To elucidate the association between dam and piglet antibody levels against UPV1 and to estimate the decrease rate of MDA, sera and colostrum of 127 gilts (before the first vaccination against UPV1; 15 days after the second vaccine dose; at farrowing; and during the second pregnancy) and sera of 276 piglets (prior to initial colostrum intake; at 7, 21, 57, 87, and 128 days-old) were tested by ELISA. Most gilts (85.8%) had anti-UPV1 antibodies before vaccination and after vaccination all were positive. At 7 days old, the majority of the piglets had anti-UPV1 antibodies, but around 57 days old, only 35.3% were positive and at 87 days old, all were negative. The estimated average half-life of MDA was 29.8 (28.8–30.9) days. A strong correlation was determined between piglet serum at 7 days old with gilt serum at farrowing time (r = 0.77, n = 248, P < 0.001) and with piglet serum at 7 days old with colostrum (r = 0.73, n = 248, P < 0.001). The MDA decreased earlier and the antibody half-life was a little longer than previously reported. Based on these findings, UPV1 vaccination can be performed earlier than usual.
