What are the components that control the assembly of subcellular organelles in eukaryotic cells? Although membranes can clearly be distorted by cytosolic factors, very little is known about the intrinsic mechanisms that control the biogenesis, shape, and organization of organellar membranes. Here, we found that the unconventional phospholipid lysobisphosphatidic acid (LBPA) could induce the formation of multivesicular liposomes that resembled the multivesicular endosomes that exist where this lipid is found in vivo. This process depended on the same pH gradient that exists across endosome membranes in vivo and was selectively controlled by Alix. In turn, Alix regulated the organization of LBPA-containing endosomes in vivo.  相似文献

Dunaliella tertiolecta (DT) was chemically investigated to isolate molecules inhibiting cancer cell proliferation and inducing apoptosis in vitro. The potency to inhibit cell growth was used for the bio-guided fractionation and isolation of active compounds using chromatographic techniques. The DT dichloromethane extract exhibited a strong anti-proliferative activity on MCF-7 and LNCaP cells, and was further fractionated and sub-fractionated by RP-HPLC. High resolution mass spectrometry and spectrophotometric analysis unequivocally identified violaxanthin as the most antiproliferative molecule present in DT DCM extract. Violaxanthin purified from DT induced MCF-7 dose-dependent growth inhibition in continuous and discontinuous treatments, at concentrations as low as 0.1 μg·mL−1 (0.17 μM). Phosphatidylserine exposure, typical of early apoptosis, was observed after 48 h treatment at 8 μg·mL−1 (13.3 μM) but no DNA fragmentation, characteristic of late apoptosis steps, could be detected even after 72 h treatment at 40 μg·mL−1 (66.7 μM). Taken together, our results demonstrate the strong antiproliferative activity of violaxanthin on one human mammary cancer cell line, and suggest that studying the pharmacology of violaxanthin and pharmacomodulated derivatives on cancer cells may allow potent antiproliferative drugs to be obtained.  相似文献

Cocoa cultivation is generally considered to foster deforestation. Contrary to this view, in the forest–savannah interface area in Cameroon, farmers have planted cocoa agroforestry systems on Imperata cylindrica grasslands, a soil-climate zone generally considered unsuitable for cocoa cultivation. We undertook a survey to understand the agricultural and ecological bases of this innovation. Age, cropping history and marketable cocoa yield were assessed in a sample of 157 cocoa plantations established on grasslands and 182 cocoa plantations established in gallery forests. In a sub-sample of 47 grassland cocoa plantations, we inventoried tree species associated with cocoa trees and measured soil organic matter levels. Marketable cocoa yields were similar for the two types of cocoa plantations, regardless of their age: 321?kg?ha?1 in cocoa plantations on grasslands and 354?kg?ha?1 in cocoa plantations in gallery forests. Two strategies were used by farmers to eliminate I. cylindrica prior to the establishment of cocoa plantations, i.e., cropping oil palms in dense stands and planting annual crops. Farmers then planted cocoa trees and fruit tree species, while preserving specific forest trees. The fruit tree and forest tree densities respectively averaged 223 and 68 trees?ha?1 in plantations under 10?years old, and 44 and 27 trees?ha?1 in plantations over 40?years old, whereas the cocoa tree density remained stable at 1,315 trees?ha?1. The Shannon–Weaver index increased from 1.97 to 2.26 over the same period although the difference was not statistically significant. The soil organic matter level was 3.13?% in old cocoa plantations, as compared to 1.7?% in grasslands. In conclusion, our results show that the occupation of grasslands by cocoa agroforestry systems is both an important example of ecological intensification and a significant farmer innovation in the history of cocoa growing.  相似文献

Nanoindentation is a powerful tool for hardness testing on a very small scale. Since the technique was first introduced for studying wood cell wall mechanics, it has been integrated as an important tool for measuring the modulus of elasticity and hardness of wood cell walls. In this study, hardness measured with nanoindentation (nanohardness) was compared with hardness measured by the standard Brinell test method (Brinell hardness) on jack pine (Pinus banksiana Lamb.) wood. Nanoindentation was performed on both the S2 layer of the secondary cell wall and the middle lamella (ML) of early- and latewood fibers. Four annual growth rings were studied. The influence of growth ring and initial spacing on both measurements was analyzed. The relationship between Brinell hardness, nanoindentation measurements, and average ring density was also studied. Results suggest that Brinell- and nanohardness are controlled by different mechanisms and factors. The location of nanohardness measurements (i.e., S2 layer or ML) also influenced hardness differently. It was concluded that nanomeasurements are not an exact representation of wood mechanical properties conducted at the macro level because of the hierarchical structure of wood. The effect of other factors such as moisture or wood extractive content may also need consideration.  相似文献

Banana streak viruses (BSV) belong to the genus Badnavirus of the family Caulimoviridae. They cause banana streak disease on banana and plantains worldwide. The recent detection of BSV in Cuba has prompted a nationwide research effort focused on the occurrence, prevalence and diversity of BSV species on dessert type banana on the island. Indexing by multiplex immunocapture-PCR (M-IC-PCR) performed on samples collected throughout the country showed that the overall prevalence of Banana streak OL virus, Banana streak GF virus and Banana streak IM virus is low in Musa acuminata genotypes in Cuba. However, the prevalence of BSV species Mysore (BSMYV) was surprisingly high in samples of cv Yangambi km5 collected from distinct and distant locations. The presence in Cuba of an as yet unreported BSV species was also investigated, showing that Banana steak VN virus is also present in Musa acuminata genotypes.  相似文献
