The investigated material consisted of the core collection of the Polish Pisum gene bank at Wiatrowo and represented a described, monohybrid variation (type lines for characters/genes and their initial lines, tester lines with markers and lines from wild taxa, 266 accessions in total). A polymorphism in 18 isozymic loci was analyzed (Lap-1, Px-1, Aat-p, Aat-m, Aat-c, Pgm-c, Pgm-p, Gpi-c, Idh, Acp-1, Skdh, Est-1, Est-2, Est-3, Aldo, 6Pgd-p, 6Pgd-c, Dia-1). The practical outcome of the analysis was a selection of lines with a high number of fast or slow isozymic alleles, useful as tester lines with markers for chromosome mapping. A polymorphism was stated in all investigated loci with 2 to 4 alleles ranging from very fast, fast, slow to very slow. Fast and slow alleles occurred in all loci. In 11 loci the incidence of fast alleles was higher than slow ones. Conversly, slow alleles occurred more frequently than fast ones in 5 loci. Additionally, there was a low incidence of a very fast allele in 7 loci and of a very slow allele in 3 loci. A slow allele was rare in 2 loci. Primitive lines and land races were a source of these rare alleles. It is suggested to consider a frequent allele of a given locus found in a group of wild lines as a so-called wild type of Pisum.  相似文献

Triterpene saponins from aerial parts of Medicago truncatula cv. Jemalong A-17, M. truncatula Gaertn.var. longispina Urb., and M. truncatula Gaertn. var. truncatula were profiled and quantified using reverse-phase liquid chromatography with on-line photodiode array detection and electrospray ionization mass spectrometry (LC-PDA/ESI/MS/MS). The determination was based on standard curves obtained for the 18 available saponin standards, previously isolated from Jemalong A-17. Aerial parts of all three subspecies contained 17 saponins previously identified and also a substantial amount of astragaloside VIII (3-GlcA-Xyl-Rha soyasapogenol B), not previously reported in M. truncatula. The compositions of saponin mixtures were very similar in the three subspecies with three dominant groups, recognized as zanhic acid, medicagenic acid, and soyasapogenol glycosides. Relative proportions of these three groups were also similar in the three subspecies: var. longispina had 49.5, 48.1, and 2.4%; var. truncatula, 41.5, 53.4, and 5.1%; and Jemalong A-17, 42.1, 56.6, and 1.3% of zanhic acid, medicagenic acid, and soyasapogenol glycosides, respectively. Jemalong A-17 had 30% lower total content of saponins as compared to M. truncatula var. longispina and M. truncatula var. truncatula; in relation to the dry matter, var. longispina contained 0.22%, var. truncatula, 0.22%, and Jemalong A-17, 0.15% dry matter of saponins. If one takes into consideration that this determination was performed on spring-collected samples, it can be concluded that the concentration of saponins in M. truncatula is similar to the concentration in alfalfa (Medicago sativa); the proportions of the three groups of saponins in these species are slightly different from those found in alfalfa, having a higher content of zanhic acid glycosides.  相似文献

We observed that diffuse interfaces sharpen rather than broaden in completely miscible ideal binary systems. This is shown in situ during heat treatments at gradually increasing temperatures by scattering of synchrotron radiation in coherent Mo/V multilayers containing initially diffuse interfaces. This effect provides a useful tool for the improvement of interfaces and offers a way to fabricate better x-ray or neutron mirrors, microelectronic devices, or multilayers with giant magnetic resistance.  相似文献

The aim of this study was to determine whether blood plasma progesterone is a reliable indicator of pregnancy in mink at an early stage of gestation. We also attempted to establish the threshold value of progesterone as a pregnancy indicator. The analysis was carried out at a production farm on 42 standard female mink aged 1 year, which were grouped both according to the observed success of mating ("mated" and "unmated") and the level of blood serum progesterone measured afterwards ("pregnant" and "nonpregnant"). It was next verified whether a particular female had been assigned to the proper group in the first place. An analysis of accuracy of mating success assessment within the group of unmated females revealed that more than one-third of decisions were wrong, since some females that had been considered unmated ultimately whelped. This suggests that mating supervision by farm workers lacks reliability. A progesterone test for verification of such management decisions should limit the risk of err,or. We suggest that progesterone tests could be carried out using the threshold values 1.9 ng/ml and 20 ng/ml in blood sampled on 25 March and 8 April, respectively, although some level of uncertainty should be taken into account.  相似文献

Stable zeolites that have larger pore apertures and a three-dimensional pore topology are of interest because they could be used to adsorb larger molecules, particularly for application in oil refining. Several large-pore zeolitic materials with channels formed by openings of more than 12 rings are known, but all of them have a one-dimensional channel system that limits their use in catalysis. We report the synthesis and some characterizations of IM-12, a thermally stable germanium-containing zeolite that contains the first two-dimensional channel system with extra-large pores formed by 14- and 12-ring channels.  相似文献

The faeces of large herbivores include viable seeds of many plant species. With dung decomposition, some seeds migrate into the soil and influence seed bank build-up. However, only a few papers report evidence of this process, and only from grasslands. In forest studies, this subject has so far been neglected. Therefore, I ask in this paper whether seeds present in the dung of European bison migrate to the forest soil seed bank and influence its build-up. I sampled soil from underneath European bison dung pats and from surrounding areas (control samples). Samples were divided into shallow (0–5 cm) and deep (5–10 cm) series. Soil seed bank content was studied in the greenhouse for 2 years. The shallow seed bank under dung pats was characterized by the highest species richness (49 species) and seed density (>2,000 m?2). Urtica dioica, Juncus effusus and Carex remota showed a demonstrable increase in seed density in the shallow seed bank underneath dung. Three years after dung deposition, seeds of endozoochoric origin influenced the soil seed bank of coniferous forest. U. dioica, J. effusus, C. remota and other species of deciduous forests and grasslands (Agrostis capillaris, Milium effusum, Polygonum hydropiper, Polygonum mite, Scrophularia nodosa, Stachys sylvatica, Veronica chamaedrys) caused an increase in seed density (by 61 %) and species richness (by 33 %) of the shallow soil seed bank underneath dung in comparison with the control soil seed bank.  相似文献

The increasing interest in coral culture for reef restoration, biotechnological applications, and to supply the marine aquarium trade has prompted researchers to optimize coral culture protocols, with emphasis on ex situ production. However, the diversity of experimental systems employed to investigate ex situ coral production may be a bottleneck to the advance of the state of the art, as it impairs reliable comparisons between experiments, as well as the replication and optimization of culture protocols. This study presents a versatile modular culture system for experimental coral production ex situ, assembled using materials and equipments available from suppliers all over the world and thus allowing researchers worldwide to truly replicate experimental setups. The validation of the modular culture system was performed using the soft coral, Sarcophyton cf. glaucum, as a model organism. The validation experiment tested the effect of different light spectra on the photosynthetic performance, symbiont density, chlorophyll and carotenoid pigments concentration, survival, and growth of coral fragments. The validation experiment confirmed the potential of this modular culture system, which ultimately enables researchers to perform direct comparisons among experiments, and more efficiently contribute to advance ex situ coral aquaculture.  相似文献

An experiment was performed to study the immunogenicity of the dermal antigen of Marek's disease virus, extracted from the skin of 30-day-old chickens, infected with Marek's disease virus on the first day of life. Three kinds of samples were tested: (1) dermal antigen centrifuged at 10 000 g per 0.5 h, (2) dermal antigen centrifugated at 10 000 g per 0.5 h and 100 000 g per 1 h, (3) dermal antigen treated like sample (2) and partly purified by DEAE-cellulose chromatography. Samples (1) and (2) were inoculated to two-day-old chickens and the vaccination was repeated, using complete Freund's adjuvant, 21 days later. Sample (3) was inoculated to two-day-old chickens with DEAE-dextran. All the three groups were challenged together with the controls (non-vaccinated chickens) on the seventh day after the first vaccination. A reduction of mortality was observed in the chickens vaccinated with and re-vaccinated with sample (1) (23.07%) and in the chickens vaccinated with sample (3) (30.76%). The chickens of the latter group were the last to start dying from Marek's disease--only after the 10th week of life. In the chickens which had been vaccinated and revaccinated with sample (2) the mortality was not reduced. The study is continued, with particular emphasis on the relationship of DEAE-dextran to protection against Marek's disease.  相似文献
