The virulence of 57 Australian isolates of Pyrenophora tritici-repentis (Ptr), a necrotrophic fungal pathogen responsible for the major wheat disease tan spot, was assessed through plant infection assays. Isolates collected from the northern, southern, and western wheat-cropping regions of Australia were evaluated against 16 Australian bread wheat cultivars under controlled growth conditions. Following infection, the wheat panel displayed varying disease symptoms ranging from tiny necrotic specks to spreading chlorotic and necrotic lesions. Analysis of variance indicated that the wheat cultivar exhibited a greater effect on the disease response, explaining 62.7% of the variation, in comparison to the isolate (10.4%). The interaction between the cultivar and the isolate was statistically significant and was attributed to 9.8% of the total variation. All Ptr isolates examined were able to cause disease, but did not display a clear distinction in virulence on the wheat panel investigated, instead showing subtle differences in aggressiveness. Based on the disease responses, there was no obvious pattern between isolate aggressiveness and cropping region. Some cultivars, such as Hydra, exhibited an effective level of resistance in relation to the panel of isolates tested. All 57 Ptr isolates were found to possess the ToxA effector gene and lack the ToxB effector gene. The gene expression level of ToxA was up-regulated at 3 days postinfection in both ToxA-sensitive and -insensitive cultivars, independent of ToxA–Tsn1 recognition.  相似文献

Due to its high vapor pressure and low boiling point, desflurane requires a specially designed, electronically controlled, temperature and pressure compensated vaporizer to regulate agent delivery to the anesthetic circuit. However, if the vapor pressure and boiling point were decreased, desflurane could be used in any conventional variable bypass vaporizer. Raoult's Law states that the vapor pressure of a liquid is proportional to its molar fraction in a solution. Accordingly, propylene glycol was used as a solvent for desflurane, and the physical characteristics of this mixture were evaluated at various molar concentrations and temperatures. Desflurane boiling point increased and vapor pressure decreased as a nonlinear function of dilution, but these changes were less than predicted by Raoult's Law. Using a circle system with a breathing bag attached at the patient end and a mechanical ventilator to simulate respiration, an in‐circuit, nonprecision vaporizer containing 40% desflurane and 60% propylene glycol achieved a 11.5 ± 1.0% (mean ± SD) circuit desflurane concentration with a 5.2 ± 0.4 (0 = off, 10 = maximum) vaporizer setting. This experiment was repeated with a dog attached to the breathing circuit under spontaneous ventilation with a fresh gas flow of 0.5 L min–1. Anesthesia was maintained for over two hours at a mean vaporizer setting of 6.2 ± 0.4, yielding mean inspired and end‐tidal desflurane concentrations of 8.7 ± 0.5% and 7.9 ± 0.7%, respectively. Within 5 minutes after cessation of anesthesia, the dog was awake, extubated and standing. In clinical practice, propylene glycol may not prove an ideal solvent for desflurane due to its instability in solution and substantial positive deviation from Raoult's Law. However, rather than alter the vaporizer to suit physical properties of anesthetic agents, this study demonstrates that it may also be possible to alter anesthetic agents to suit physical properties of the vaporizer.  相似文献

Fungicides mixed with low concentrations of surfactant proved as effective for the eradication of apple mildew as high concentrations of surfactant alone. The fungicide-containing mixtures had the advantage of being less phytotoxic than the full strength surfactants. Fungicides without extra surfactant failed to control mildew when applied during the winter.  相似文献

Several species of wheat stem sawflies (WSS) are pests of wheat, including Cephus cinctus Norton in North America. Larvae feed inside the stems and cut the stem near plant maturity. The primary means of control is resistance due to solid stems, largely controlled by a locus on chromosome 3B (Qss.msub‐3BL). Cultivars that differed for WSS resistance, but with similar stem solidness, were crossed to determine the genetic basis for the differences. The cultivar ‘Scholar’ is susceptible, while ‘Conan’ shows resistance. Scholar and Conan possessed different alleles at Qss.msub‐3BL. Both alleles conferred solidness, yet the Conan allele conferred higher WSS resistance. An allele from Conan on chromosome 4A also decreased infestation and stem cutting. The 3B and 4A alleles from Conan acted in an additive fashion to provide increased WSS resistance without increasing stem solidness. Stem solidness has long been used by breeders as a proxy for WSS resistance because of its simplicity. Our results suggest that other resistance mechanisms may complement solid stems.  相似文献

This study reports 14th-year response of a boreal mixedwood stand to different harvest intensities (uncut, 50% partial cut with and without removal of residuals after 3 years, and clearcut), spot site preparation treatments (none and scalped), and chemical weeding frequencies (none, single, and multiple) in northeastern Ontario. The response variables include the survival and growth of planted white spruce (Picea glauca [Moench] Voss) and jack pine (Pinus banksiana Lamb.), height and density of natural regeneration and shrubs, and cover of shrubs and non-woody vegetation. Harvesting and weeding generally improved survival and growth of planted trees, although white spruce survival did not significantly differ among the three weeding frequencies. Harvesting tended to increase heights of hardwood (mostly trembling aspen (Populus tremuloides Michx.)) and conifer (largely balsam fir (Abies balsamea (L.) Mill.).) natural regeneration, cover and density of shrubs, and cover of herbs, lichens, and ferns. Chemical weeding reduced height, density and cover of shrubs, height and density of hardwood regeneration, and fern cover, but increased moss and lichen cover. Spot scalping did not significantly affect planted seedling, natural regeneration, or the vegetation.Maximum survival and growth of planted white spruce and jack pine were achieved using a combination of clearcutting and multiple weeding. However, partial cutting followed by a single weeding produced acceptable survival and reasonable growth of planted trees, particularly for white spruce. Partial canopy removal alone substantially reduced the amount of hardwood regeneration, relative to clearcutting, but did not adequately suppress understory shrubs. Significant improvement in seedling growth following multiple weedings was evident primarily in the complete canopy removal treatments: 50% partial cut with removal of residuals after 3 years and clearcut. While the effects of harvesting and weeding on planted crop trees found in the 5th-year assessments generally persisted at year 14, survival decreased, likely due to light competition from developing hardwood and shrubs.  相似文献

Cultivated barley is the major livestock feed grain in the Northern Plains and Northwestern United States due to the fact that its short growing season and limited rainfall limit the planting and production of corn. Starch and fiber content play a significant role in feedlot performance of animals raised on barley feed. To study the underlying genetic locations and mechanisms for these traits, a recombinant inbred line population was derived from a cross between the hulled barley cultivar Valier and a hull-less Swiss landrace line, PI370970. Valier has a high acid detergent fiber content (ADF) and low starch and protein while PI370970 contains low ADF and high starch and protein content. To detect associated QTLs, data were collected and analyzed from irrigated and rain-fed environments. A total of 30 main effect QTLs and four epistatic QTLs were identified which conditioned ADF, starch and protein content under rain-fed, irrigated and combined analyses. These QTLs were located on chromosomes 2H, 3H, 5H, 6H and 7H. Major ADF and starch QTL were identified on chromosome 7H near the nud locus (the locus controlling hulled vs. hull-less caryopsis). High heritability estimates for both ADF and starch content suggest that early selection for these traits during breeding would be productive. Low ADF-QTL were independently verified in a second population in a different genetic background.  相似文献

More than half of the barley grown in the USA is used for livestock feed, with the remaining stocks diverted for human food and malting purposes. The use of barley grain as a major source of cattle feed has been criticized because of its rapid digestion in the rumen, which can result in digestive disorders in cattle. In sacco dry matter digestibility (ISDMD) and particle size (PS) after dry rolling have been found to play a role in the feedlot performance of barley as a feed grain. Reducing the rate of ISDMD is predicted to result in significantly improved animal health and average daily gain. A recombinant inbred line population derived from a cross between a high ISDMD, two-rowed barley cultivar (Valier) and a six-rowed Swiss landrace line (PI370970) exhibiting far slower ISDMD has been developed for studying the underlying genetic locations and mechanisms of these traits. To detect associated quantitative trait loci (QTLs), we collected and analyzed data from irrigated and rain-fed environments. A significant negative correlation was observed between ISDMD and PS. High heritability estimates for ISDMD and PS suggest that early selection for these traits during breeding would be achievable. Four QTLs were identified on chromosomes 2H, 6H, and 7H, explaining 73–85% of ISDMD phenotypic variation, while three QTLs on 2H and 7H were associated with variation in PS and explained 58–77% of its variation. A major QTL on chromosome 2H tightly linked to the morphology-modifying gene vrs1 was found to dramatically control 35–62% of the phenotypic variation of ISDMD and 26–53% of that of PS. The impact of the vrs1 locus on ISDMD was validated in two populations representing different genetic backgrounds. Our results suggest that it may also be advantageous to simultaneously overlap these QTLs around the vrs1 locus.  相似文献
