- 1. Sponges are an important component of coral reef ecosystems, but even though they are widespread with the ability to significantly influence other benthic community members they rarely feature to any great extent in current monitoring or biodiversity assessment programmes conducted by volunteer and professional groups. This exclusion is usually because of the taxonomic problems associated with sponge identification.
- 2. A potential alternative to monitoring temporal or spatial change in sponge assemblages and assessing biodiversity levels is to characterize sponges using morphologies present rather than collecting species data. Quantifying sponge biodiversity (for monitoring and biodiversity assessments) at the morphological level is less time and resource consuming than collecting species data and more suited to groups with little training and experience of sponge taxonomy or in regions where detailed taxonomic information on sponges is sparse.
- 3. This paper considers whether the same differences and similarities in sponge richness and assemblage composition can be identified using species and morphological data in response to environmental gradients at two coral reef ecosystems in south‐east Sulawesi, Indonesia, and whether volunteers can be used to reliably collect morphological information. Sponge morphologies were classified into 14 groups and different morphological assemblages were found by the author at the two sites and between depth intervals. Comparisons of sponge species and morphological composition data showed that common patterns in assemblage structuring and richness could be identified irrespective of whether morphological or species data were used. In addition, a positive linear relationship was found between sponge species and morphological richness.
- 4. The morphological data recorded by volunteer divers (n=10) were compared with that collected by the author. Although volunteers recorded fewer sponges than the author (approximately 15% less), missing mainly small encrusting specimens, similar assemblage structure could be identified from both the volunteers' and the author's data.
- 5. The results showed that the same differences in sponge assemblages between sites and depths could be identified from both species and morphological data. In addition, these morphological data could be reliably collected by volunteer divers.

We examined the potential for producing the large numbers of sandfish (Holothuria scabra) needed for restocking programmes by co-culturing juveniles with the shrimp Litopenaeus stylirostris in earthen ponds. Our experiments in hapas within shrimp ponds were designed to detect any deleterious effects of sandfish on shrimp, and vice versa. These experiments showed that a high stocking density of juvenile sandfish had no significant effects on growth and survival of shrimp. However, survival and growth of sandfish reared with shrimp for 3 weeks were significantly lower than for sandfish reared alone. Increased stocking density of shrimp also had a significant negative effect on survival and/or growth of sandfish. A grow-out trial of juvenile sandfish in 0.2-ha earthen ponds stocked with 20 shrimp post-larvae m− 2, and densities of sandfish between 0.8 and 1.6 individuals m− 2, confirmed that co-culture is not viable. All sandfish reared in co-culture were dead or moribund after a month. However, sandfish stocked alone into 0.2-ha earthen ponds survived well and grew to mean weights of ∼ 400 g within 12 months without addition of food. The grow-out trial demonstrated that there is potential for profitable pond farming of sandfish in monoculture. Further research is now needed to identify the optimal size of juveniles, stocking densities and pond management regimes. 相似文献

The objective of the present study was to compare the effectiveness of dietary marine phospholipids (MPL) obtained from krill and soybean lecithin (SBL) on the rearing performance and development of seabream (Sparus aurata) larvae. Larvae were fed from 16 to 44 day posthatching (dph) five formulated microdiets with three different levels (50, 70 and 90 g kg–1) of phospholipids (PL) obtained either from an MPL or from a SBL source. Larvae‐fed MPL show a higher survival, stress resistance and growth than those‐fed SBL, regardless the dietary PL level. Overall, the increase in MPL up to 70 g kg–1 total PL in diet was enough to improve larval gilthead seabream performance, whereas even the highest SBL inclusion level (90 g kg–1 PL) was not able to provide a similar success in larval growth or survival. Inclusion of SBL markedly increased the peroxidation risk as denoted by the higher TBARs in larvae, as well as a higher expression of CAT, GPX and SOD genes. Moreover, SBL tends to produce larvae with a lower number of mineralized vertebrae and a lower expression of osteocalcin, osteopontin and BMP4 genes. Finally, increasing dietary MPL or SBL lead to a better assimilation of polyunsaturated fatty acids in the larvae, n‐3HUFA (especially 20:5n‐3) or n‐6 fatty acids (especially 18:2n‐6), respectively. In conclusion, MPL had a higher effectiveness in promoting survival, growth and skeletal mineralization of gilthead seabream larvae in comparison with SBL. 相似文献

Carp (Cyprinus carpio L.) is a pest species in Australian waterways, and cyprinid herpesvirus 3 (CyHV‐3) is being considered as a potential biological control (biocontrol) agent. An important consideration for any such agent is its target specificity. In this study, the susceptibility to CyHV‐3 of a range of non‐target species (NTS) was tested. The NTS were as follows: 13 native Australian, and one introduced, fish species; a lamprey species; a crustacean; two native amphibian species (tadpole and mature stages); two native reptilian species; chickens; and laboratory mice. Animals were exposed to 100–1000 times the approximate minimum amount of CyHV‐3 required to cause disease in carp by intraperitoneal and/or bath challenge, and then examined clinically each day over the course of 28 days post‐challenge. There were no clinical signs, mortalities or histological evidence consistent with a viral infection in a wide taxonomic range of NTS. Furthermore, there was no molecular evidence of infection with CyHV‐3, and, in particular, all RT‐PCRs for viral mRNA were negative. As a consequence, the results encourage further investigation of CyHV‐3 as a potential biocontrol agent that is specific for carp. 相似文献

Changes in fatty acid metabolism in Atlantic salmon (Salmo salar) induced by vegetable oil (VO) replacement of fish oil (FO) and high dietary oil in aquaculture diets can have negative impacts
on the nutritional quality of the product for the human consumer, including altered flesh fatty acid composition and lipid
content. A dietary trial was designed to investigate the twin problems of FO replacement and high energy diets in salmon throughout
the entire production cycle. Salmon were grown from first feeding to around 2 kg on diets in which FO was completely replaced
by a 1:1 blend of linseed and rapeseed oils at low (14–17%) and high (25–35%) dietary oil levels. This paper reports specifically
on the influence of diet on various aspects of fatty acid metabolism. Fatty acid compositions of liver, intestinal tissue
and gill were altered by the diets with increased proportions of C18 polyunsaturated fatty acids and decreased proportions of n-3 highly unsaturated fatty acids (HUFA) in fish fed VO compared
to fish fed FO. HUFA synthesis in hepatocytes and enterocytes was significantly higher in fish fed VO, whereas β-oxidation
was unaltered by either dietary oil content or type. Over the entire production cycle, HUFA synthesis in hepatocytes showed
a decreasing trend with age interrupted by a large peak in activity at seawater transfer. Gill cell prostaglandin (PG) production
showed a possible seasonal trend, with peak activities in winter and low activities in summer and at seawater transfer. PG
production in seawater was lower in fish fed the high oil diets with the lowest PG production generally observed in fish fed
high VO. The changes in fatty acid metabolism induced by high dietary oil and VO replacement contribute to altered flesh lipid
content and fatty acid compositions, and so merit continued investigation to minimize any negative impacts that sustainable,
environmentally-friendly and cost-effective aquaculture diets could have in the future.

Freshly isolated rainbow trout hepatocytes were exposed to tert-butyl hydroperoxide (BuOOH), a substrate for glutathione peroxidase.
BuOOH at a concentration approximately equimolar (1 mM) with intracellular reduced glutathione (GSH) caused a reversible increase
in intracellular glutathione disulphide (GSSG) but did not compromise cell viability or damage membrane lipids. BuOOH at 10
mM caused a large irreversible increase in intracellular GSSG followed by efflux into the medium. Considerable leakage of
lactate dehydrogenase and loss of highly unsaturated fatty acids, particularly docosahexaenoic acid also occurred. Dependence
of hydroperoxide removal on flux through the hexose monophosphate pathway was suggested by the increased release of 14CO2 from [1-14C] glucose from hepatocytes incubated with BuOOH. 相似文献

Leaching of sulfur (S) on sandy soils may limit the effectiveness of S fertilizers especially when applied at sowing. The effectiveness of S sources for canola (oil seed rape, Brassica napus L.) grown in sandy low S soils of south-western Australia is not known. This study was completed to determine the relative effectiveness of gypsum and a gypsum-based by-product from synthetic rutile processing called Canola Blue for canola grown in low S soils of the region. Canola Blue is a mixture of gypsum and elemental S, and is granulated so its effectiveness may vary from gypsum. We measured the effectiveness of the two S sources in the glasshouse for young seedling growth and for minimizing S leaching. In the four field experiments, the two S sources were evaluated for relative effects on canola seed yield and the concentration of oil in seed. Canola Blue applied at sowing was as effective as gypsum for canola growth in the glasshouse and when applied at 35 days after sowing (DAS) was as effective as gypsum for seed yield in the field. For the glasshouse study, Canola Blue when applied to the soil surface (topdressed) at 35 DAS was as effective as gypsum in achieving a rapid recovery of leaves from S deficiency symptoms and of shoot growth. Where S deficiency appears during the growing season, topdressing with Canola Blue appears to be as effective under the conditions of this experiment as was gypsum. The goal of this study was to define the role for p38 mitogen-activated kinase (MAPK) in the signaling mechanism regulating pro-inflammatory cyclooxygenase (COX) gene expression in lipopolysaccharide (LPS)-activated equine leukocytes for the purposes of identifying novel targets for anti-inflammatory therapy in endotoxemic horses. The p38 MAPK has been shown to positively regulate inflammatory gene expression in human leukocytes and can be activated by a variety of stimuli including LPS, TNF-alpha, and IL-1. Activation-associated phosphorylated p38 MAPK has been implicated in the up-regulation of several inflammatory genes, including COX-2 which ultimately results in the production of prostanoids that are responsible for the pathophysiology associated with endotoxemia. Our hypothesis is that activation of p38 MAPK is essential for LPS-induced COX-2 expression in equine peripheral blood leukocytes. We tested our hypothesis by investigating the effects of the specific p38 MAPK inhibitors SB203580 and SB202190 on LPS-induced COX-2 protein expression and PGE(2) production in equine leukocytes. LPS stimulation activated p38 MAPK and increased COX-2 expression in a dose-dependent manner with maximal activation observed after 30min and 4h, respectively, at a concentration of 10 ng/ml LPS. In contrast, LPS stimulation did not affect COX-1 protein expression. Pretreatment with SB203580 or SB202190 significantly inhibited LPS-induced activation-associated p38 MAPK phosphorylation, COX-2 mRNA and protein levels, and PGE(2) production in equine leukocytes. Maximal inhibition of LPS-induced COX-2 protein expression was achieved at a concentration of 10 microM SB203580. We concluded that p38 MAPK is essential for LPS-induced COX-2 expression suggesting that p38 MAPK is a potential target for anti-inflammatory therapy during equine endotoxemia. 相似文献
