Pathology of experimentally-induced, acute toxoplasmosis in macropods   总被引:1,自引:0,他引:1  
SUMMARY Thirteen Tammar wallabies (Macropus eugenii) were dosed orally with 500, 1000 or 10 000 oocysts of Toxoplasma gondii, as part of a vaccination trial. Eleven animals died of acute toxoplasmosis 9 to 15 days after challenge. The lesions were similar in all animals, consisting of foci of necrosis and inflammation in the intestines, lymphoid tissue, adrenal cortex, heart, skeletal muscle and brain, and severe generalised pulmonary congestion and oedema. Free and intracellular tachyzoites of Toxoplasma were associated with the lesions. The remaining 2 animals had shown no signs of disease when euthanased four months after challenge. Small, focal, non-suppurative inflammatory lesions were seen in brain, heart and skeletal muscle of these animals and chronic Toxoplasma infection was confirmed by mouse inoculation.  相似文献

The effects on eyespot of carbendazim, prochloraz or a mixture of carbendazim and prochloraz, applied twice yearly and to the same plots in each year, were studied in five successive crops of winter wheat. Some plots were uninoculated (with a background population of the eyespot fungus, Pseudocercosporella herpotrichoides, which was initially mainly MBC-sensitive W-type), whereas others were inoculated artificially in the first autumn of the experiment (1984) with mainly MBC-sensitive W-type, MBC-resistant W-type, MBC-sensitive R-type or MBC-resistant R-type fungus. Samples were taken in April (1985–1987) and July (1985–1989). When all inoculation treatments are considered together, carbendazim was ineffective at all sampling times after April 1985 because of its rapid selection for MBC-resistance, and increased eyespot incidence and severity in some samples. Prochloraz applied alone decreased eyespot incidence and severity at all sample times except July 1988. Where applied with carbendazim, prochloraz always decreased eyespot incidence and severity, sometimes more so than when applied alone. The control of eyespot by the fungicides was influenced by the initial populations in the first two seasons only. Carbendazim decreased eyespot incidence in 1985 only where no artificial inoculum was added (both samples) and where mainly MBC-sensitive R-type inoculum was added (April 1985). Prochloraz applied alone was ineffective in July 1985 where mainly MBC-sensitive R-type fungus was added, and, applied with carbendazim, was ineffective in July 1986 where mainly MBC-resistant fungus was added. Subsequently, the populations were unrelated to the initial populations and were mainly a result of selection by fungicides where these were applied. Although selecting for the R-type fungus and, where no carbendazim was applied, for MBC-sensitivity, prochloraz remained generally effective after repeated applications over five seasons.  相似文献

When plants encounter salinity, growth is reduced initially by water stress and subsequently by toxic levels of ions and by interference with nutrient acquisition and translocation. Calcium (Ca2 +) in particular seems to have an important role in salt tolerance and there are reports of a beneficial effect of increasing Ca2 + availability. Higher potassium (K+) concentrations in plants may also improve salinity tolerance as sodium (Na)+/K+ ratios have been shown to be important. Previous work with a range of Acacia species has suggested that endogenous seed Ca2 + and K+ concentrations might influence salinity tolerance at germination, but this has not previously been tested with a single species. The objectives of this investigation were thus to determine whether (1) altered Ca2 + and K+ nutrition of Brassica campestris (rapa) L. plants affects the yield and ion content of their seeds, and (2) seeds with different Ca2 + and K+ contents differ in their salinity tolerance. Plants were grown in a growth room or greenhouse in (1) Gem® horticultural sand (2) Silvaperl® perlite and sand (2:1), or (3) Shamrock® Medium General Purpose Irish Sphagnum Peat and Vermiperl® vermiculite (1:1). Plants in each growth substrate were supplied with nutrient solutions based on a modified Hoagland's solution as a control, low Ca2 + and low K+ solutions containing those elements at half the control strength, but all other mineral elements as far as possible at control strength, and high Ca2 + and high K+ solutions containing those elements at double control strength but all other mineral elements, as far as possible, at control strength. An increase in substrate available Ca2 + and K+ resulted in increased Ca2 + and K+ concentration in seeds, respectively, and was accompanied by a reduction in seed K+ and Ca2 +, respectively. The Ca2 + and K+ concentrations of seeds affected their salinity tolerance. Increases in seed Ca2 +, K+ or Ca2 ++ K+ concentrations decreased salinity tolerance at germination. The results, especially in terms of Ca2 + nutrition, contradict previous results of an increased salinity tolerance with increased Ca2 + and/or K+ concentrations.  相似文献

CASE HISTORY: An adult male Birman cat was evaluated for recurrent, intermittent vomiting or regurgitation, occasionally associated with abdominal discomfort.

CLINICAL FINDINGS AND DIAGNOSIS: Radiographs, including an oesophogram, indicated an oesophageal obstruction. Prior to treatment, the cat's condition deteriorated and it was euthanised at the owner's request. Post-mortem examination revealed a gastro-oesophageal intussusception, a trichobezoar impacted into the intussusceptum, and a dilated oesophageal hiatus consistent with a chronic hiatal hernia.

CLINICAL RELEVANCE: Gastro-oesophageal intussusception is a rare condition in cats. Its aetiology in relation to a pre-existing hiatal hernia and a trichobezoar is discussed.  相似文献

This study aimed to evaluate various concentrations of egg yolk (5, 10, or 20%) in combination with different concentrations of glycerol (3% or 6%) added to a Tris‐based extender on the post‐thaw characteristics of sperm obtained from Tayassu tajacu. For this purpose, semen from 10 sexually male mature collared peccaries was collected by electroejaculation and evaluated for sperm motility, vigour, viability, morphology and functional membrane integrity. The ejaculates were initially extended in Tris‐fructose plus egg yolk (5%, 10% or 20%). After cooling, the semen was added to Tris‐egg yolk plus glycerol (6% or 12%), resulting in a final concentration of 3% or 6% glycerol of the extender. Straws were frozen using liquid nitrogen and thawed in a water bath at 37°C for 30 s. The frozen–thawed semen was evaluated as reported for fresh semen. After thawing, a significant decrease was verified for sperm motility and vigour, for all the samples in comparison with fresh semen. However, no differences were evidenced among treatments for any sperm characteristics evaluated (p > 0.05), except for the combination between 10% egg yolk and 6% glycerol, which provided the worst preservation of functional membrane integrity (p < 0.05). The interactions between higher concentrations of egg yolk (20%) and glycerol (6%) and also between lower concentrations of the same substances (5% egg yolk and 3% glycerol) added to the Tris‐based extender negatively affected the preservation of the normal sperm morphology after thawing (p < 0.05). In conclusion, the use of Tris‐based extender added to 10% or 20% egg yolk plus 3% glycerol is recommended for effective sperm cryopreservation in collared peccaries.  相似文献

Western flower thrips (WFT) is one of the most economically important pest insects of many crops worldwide. Recent EU legislation has caused a dramatic shift in pest management strategies, pushing for tactics that are less reliable on chemicals. The development of alternative strategies is therefore an issue of increasing urgency. This paper reviews the main control tactics in integrated pest management (IPM) of WFT, with the focus on biological control and host plant resistance as areas of major progress. Knowledge gaps are identified and innovative approaches emphasised, highlighting the advances in ‘omics’ technologies. Successful programmes are most likely generated when preventive and therapeutic strategies with mutually beneficial, cost‐effective and environmentally sound foundations are incorporated. © 2017 The Authors. Alzheimer's disease is hypothesized to be caused by an imbalance between β-amyloid (Aβ) production and clearance that leads to Aβ accumulation in the central nervous system (CNS). Aβ production and clearance are key targets in the development of disease-modifying therapeutic agents for Alzheimer's disease. However, there has not been direct evidence of altered Aβ production or clearance in Alzheimer's disease. By using metabolic labeling, we measured Aβ42 and Aβ40 production and clearance rates in the CNS of participants with Alzheimer's disease and cognitively normal controls. Clearance rates for both Aβ42 and Aβ40 were impaired in Alzheimer's disease compared with controls. On average, there were no differences in Aβ40 or Aβ42 production rates. Thus, the common late-onset form of Alzheimer's disease is characterized by an overall impairment in Aβ clearance.  相似文献

A challenge model for pancreas disease in Atlantic salmon, Salmo salar L. fry, was developed comparing two salmonid alphavirus (SAV) subtypes: SAV1 and SAV5. Viral doses of 3 × 105 TCID50 mL−1 for SAV1 and 3 × 104 for SAV5 were tested in triplicate tanks, each containing 450 salmon fry. Cumulative mortalities of 1.2% were recorded. Titres of virus recovered from the mortalities ranged from 102 to 107 TCID50 mL−1. Fry were sampled at 3, 5 and 7.5 weeks post-challenge. Sampling after 3 weeks revealed a high prevalence of infection in the absence of clinical signs, and infectious virus was recovered from 80% and 43% of sampled fry infected with SAV1 and SAV5, respectively. After 5 weeks pancreas, heart and red skeletal muscle lesions were generally observed, whilst degeneration in white skeletal muscle was observed only in fish infected with SAV1. In situ hybridisation confirmed the presence of viral genome in infected pancreas, heart and muscle. After 7.5 weeks, infectious virus (both isolates) was recovered from 13.3% of the fish sampled, with a viral titre of 102 TCID50 mL−1. Clearly, salmon fry are susceptible to SAV infection and pancreas disease.  相似文献

OBJECTIVE: To evaluate the accuracy of point-of-care tests for the diagnosis of disseminated intravascular coagulation (DIC) in dogs and assess the correlation and agreement of results between point-of-care and laboratory tests in the evaluation of hemostatic function. DESIGN: Prospective case series. ANIMALS: 59 critically ill dogs (affected dogs) with clinical signs of diseases known to predispose to DIC and 52 clinically normal dogs. PROCEDURES: Accuracy of the point-of-care tests (activated clotting time [ACT], estimated platelet count and number of schizocytes from a blood smear, plasma total solids [TS] concentration, and the protamine sulfate test) was evaluated, using receiver operating characteristic curves and likelihood ratios. A strategy, using likelihood ratios to calculate a posttest probability of DIC, was tested with 65% used as a threshold for initiation of treatment. Results of laboratory tests (coagulogram and plasma antithrombin III activity) were used as the standard for comparison in each dog. RESULTS: ACT and estimated platelet count provided the best accuracy for detection of DIC. The plasma TS concentration, schizocyte number, and protamine sulfate test had poor accuracy. The strategy using post-test probability of DIC identified 12 of 16 affected dogs that had DIC. Estimated platelet count was correlated and had acceptable clinical agreement with automated platelet count (r = 0.70). The plasma TS (r = 0.28) concentration and serum albumin (r = 0.63) concentration were not accurate predictors of plasma antithrombin III activity. The ACT did not correlate with activated partial thromboplastin time (r = 0.28). CONCLUSIONS AND CLINICAL RELEVANCE: Strategic use of likelihood ratios from point-of-care tests can assist clinicians in making treatment decisions for dogs suspected to have DIC when immediate laboratory support is unavailable.  相似文献
