Haematopoietic necrosis virus [cyprinid herpesvirus 2 (CyHV-2)] was isolated during disease outbreaks in goldfish, Carassius auratus, at an ornamental fish retail site in southern England in 2004. Signs of disease included lethargy and inappetence and were first seen after water temperatures increased from 14-15 to 19-21 degrees C. External gross pathology included pale patches on the gills and skin and internally the spleen was enlarged, often with distinctive white nodules. The most prominent histopathological changes observed were necrotic lesions in the spleen and kidney and focal patches of necrosis in the gill lamellae. Necrotic cells often contained nuclei with marginated chromatin and pale intranuclear inclusions. Ultrastructural examination of the spleen tissue revealed typical herpesvirus-like particles measuring 100 nm in diameter. The virus was isolated from extracts of gill tissue in KF-1 cells at 20 degrees C and oligonucleotide primer sets were designed based on conserved gene sequences and used to amplify viral DNA by polymerase chain reaction (PCR). The PCR assays were then used to detect the virus in DNA extracted from tissues sampled during earlier disease investigations at the retail site owner's holding facility in 2002 and 2003 and stored at -70 degrees C since then. Polymerase gene-specific PCR amplification products obtained from tissue samples and from the virus isolated in cell culture shared 100% nucleotide sequence identity with the published sequence for CyHV-2.  相似文献

Here, we examine the effect of long-term pH differences and short-term pH change on N2O emissions from soil, and the microbial source (ammonia oxidation versus denitrification) of 15N-N2O emissions. 15N-fertiliser (20 g N m?2; 10 atom% excess 15N) was applied to (1) a silt loam soil of pH 7 held at 50% and 65% water-filled pore space (WFPS) (experiment 1) and (2) a loamy sand soil maintained at pH 4.5 and pH 7 for over 40 years (experiment 2). Soils were limed with CaCO3 or acidified with H2SO4, and comparisons were made with unadjusted soils. Ammonia oxidation was the main microbial source of 15N-N2O in soils limed to pH 7.0–8.1, unadjusted pH 7.1 (Experiment 1) and long-term pH 7 (experiment 2) soils. Eighty percent of 15N-N2O from the long-term pH 4.5 soil (experiment 2) was derived from denitrification suggesting a possible inhibition of N2O reduction. Short-term acidification to pH 5.6 or 4.3 lowered N2O emissions. Liming of the pH 4.5 soil resulted in over four times greater N2O emission (11 mg 14+15N-N2O m?2 over 41 days) than from the long-term pH 7.0 soil (experiment 2), with an associated increase in ammonia oxidiser-N2O and decrease in denitrifier-N2O production. This is the first report of a pH-induced change in microbial source of N2O. Our results highlight the importance of distinguishing between short- and long-term effects of pH management when predicting N2O emissions from soil, as they exhibit predominance of different microbial groups in N2O production, with likely adaptation of the microbial community.  相似文献

Authentic samples of commercially produced organic and conventionally grown tomatoes, lettuces, and carrots were collected and analyzed for their delta15N composition in order to assemble datasets to establish if there are any systematic differences in nitrogen isotope composition due to the method of production. The tomato and lettuce datasets suggest that the different types of fertilizer commonly used in organic and conventional systems result in differences in the nitrogen isotope composition of these crops. A mean delta15N value of 8.1 per thousand was found for the organically grown tomatoes compared with a mean value of -0.1 per thousand for those grown conventionally. The organically grown lettuces had a mean value of 7.6 per thousand compared with a mean value of 2.9 per thousand for the conventionally grown lettuces. The mean value for organic carrots was not significantly different from the mean value for those grown conventionally. Overlap between the delta15N values of the organic and conventional datasets (for both tomatoes and lettuces) means that it is necessary to employ a statistical methodology to try and classify a randomly analyzed "off the shelf" sample as organic/conventional, and such an approach is demonstrated. Overall, the study suggests that nitrogen isotope analysis could be used to provide useful "intelligence" to help detect the substitution of certain organic crop types with their conventional counterparts. However, delta15N analysis of a "test sample" will not provide unequivocal evidence as to whether synthetic fertilizers have been used on the crop but could, for example, in a situation when there is suspicion that mislabeling of conventionally grown crops as "organic" is occurring, be used to provide supporting evidence.  相似文献

Recent studies of veterinary practice have suggested a correlation between well-developed communication skills and job satisfaction, career retention, customer satisfaction, decreased lawsuits, and financial remuneration for veterinarians. Veterinary educators are under growing pressure to teach functional communication skills to veterinary students; however, the methods employed have not been well evaluated. In this study we have evaluated veterinary student's attitudes to learning communication skills by participating in role play. The study indicates that experiential learning modalities such as role play are perceived as effective by students, despite reluctance to participate and some discomfort surrounding participation.  相似文献

OBJECTIVE: To determine if a constant rate local anesthetic delivery system is more effective than continuous intravenous (IV) morphine infusion for postoperative analgesia. ANIMALS: Twenty client-owned dogs undergoing total ear canal ablation. METHODS: Dogs were randomly assigned to the lidocaine group (LID) or the morphine group (MOR). The LID group received a constant rate infusion of lidocaine locally and a continuous IV infusion of saline, while the MOR group received a constant rate infusion of saline locally and a continuous IV infusion of morphine. The primary investigator evaluated each patient and determined a hospital behavior score, anesthesia recovery score, preoperative pain score, and serial postoperative pain and sedation scores over 38 hours. Pain and sedation observations were videotaped and scored by three additional evaluators. Evaluators were blinded to treatment assignments. RESULTS: There were no significant differences in age, weight, hospital behavior scores or anesthesia recovery scores. The primary investigator's pain scores were not significantly different, but sedation scores were significantly lower for the LID group. Sedation and pain scores by the video evaluators were not significantly different between groups. Kappa agreement between observers was poor, but better agreement was noted between sedation scores than pain scores. Drug-related complications were significantly lower in the LID group (n = 0) compared with the MOR group (n = 5). Wound complications were not significantly different (LID = 4, MOR = 4). Intravenous delivery complications occurred in 12 (60%) patients. Local delivery complications occurred in five (25%) dogs. Delivery complications were not significantly different between groups. CONCLUSIONS AND CLINICAL RELEVANCE: Continuous incisional lidocaine delivery was an equipotent and viable method of providing postoperative analgesia compared with IV morphine. Lidocaine delivery resulted in a trend toward lower pain scores, significantly lower sedation scores, and no dogs requiring analgesic rescue. Wound complications secondary to local infusion were minor and self-limiting. Drug-related complications occurred only in the MOR group.  相似文献

Mercury (Hg) contamination from a variety of point and non-point sources, including atmospheric inputs, is currently considered to be the most serious environmental threat to the well being of fish and wildlife resources in the southeastern United States. Fish consumption advisories have been issued in all ten states comprising the U.S. Fish and Wildlife Service's Southeast Region. Both freshwater and marine species have been affected with levels ranging as high as 7.0 ppm in some individuals. Many other species, including various species of reptiles, birds and mammals (including humans) are also contaminated. Impacts noted range from reproductive impairment to mortality.  相似文献

The Sierra Nevada batholith is localized in the axial region of a complex faulted synclinorium that coincides with a downfold in the Mohorovicic discontinuity and in P-wave velocity boundaries within the crust. Observed P-wave velocities are compatible with downward increase in the proportion of diorite, quartz diorite, and calcic granodiorite relative to quartz monzonite and granite in the upper crust, with amphibolite or gabbro-basalt in the lower crust, and with periodotite in the upper mantle. The synclinorium was formed in Paleozoic and Mesozoic strata during early and middle Mesozoic time in a geosyncline marginal to the continent. Granitic magmas are believed to have formed in the lower half of the crust at depths of 25 to 45 kilometers or more, primarily as a result of high radiogenic heat production in the thickened prism of crustal rocks. Magma was generated at different times in different places as the locus of down-folding shifted. It rose into the upper crust because it was less dense than rock of the same composition or residual refractory rocks. Refractory rocks and crystals that were not melted and early crystallized mafic minerals that settled from the rising magma thickened the lower crust. Wall and roof rocks settled around, and perhaps through, the rising magma and provided space for its continued rise. Erosion followed each magmatic episode, and 10 to 12 kilometers of rock may have been eroded away since the Jurassic and 7 to 10 kilometers since the early Late Cretaceous.  相似文献

We have examined the architecture of a protein complex in the absence of bulk water. By determining collision cross sections of assemblies of the trp RNA binding protein, TRAP, we established that the 11-membered ring topology of the complex can be maintained within a mass spectrometer. We also found that the binding of tryptophan enhances the stability of the ring structure and that addition of a specific RNA molecule increases the size of the complex and prevents structural collapse. These results provide definitive evidence that protein quaternary structure can be maintained in the absence of bulk water and highlight the potential of ion mobility separation for defining shapes of heterogeneous macromolecular assemblies.  相似文献
