Detection of infectious hypodermal and hematopoietic necrosis virus (IHHNV) in shrimp is complicated by the fact that certain virus-related sequences are integrated into the genome of Penaeus monodon in some parts of the world, which has been reported so far from Africa and Australia. In this study, we evaluated the highly specific and sensitive diagnostic primer sets for detection of infectious IHHNV and integrated virus-related sequence in 177 samples of P. monodon from India. A nested primer set, IHHNV648F/R and IHHNV309F/R was used to specifically detect infectious IHHNV and not the virus-related sequences. IHHNV was detected in 67.4% postlarvae (PL) and 34% adult samples using this primer set. The OIE recommended primers IHHNV392F/R and IHHNV389F/R gave positive reaction for 86.7% PL and 67% adult samples, while the primer pair 77012F and 77353R gave positive reaction with 46.7% PL and 20% adult samples. These primers were found to detect virus-related sequence integrated into the shrimp genome. The analysis of virus-related sequence by MG831F/R primers showed that 33.7% PL and 31.7% adult shrimp possessed Type A virus-related sequence. 22.8% PL and 10.5% adults had both IHHNV and Type A virus-related sequence. Cloning and sequencing 832 bp virus-related sequence from P. monodon from India revealed presence of five shrimp DNA markers between 439 and 825 bp. This study is the first conclusive report on the presence of Type A virus-related sequence in P. monodon from India. 相似文献

Parkinson’s disease (PD) is the second most common neurodegenerative disorder in the elderly people, currently with no cure. Its mechanisms are not well understood, thus studies targeting cause-directed therapy or prevention are needed. This study uses the transgenic Caenorhabditis elegans PD model. We demonstrated that dietary supplementation of the worms with an extract from the cultivated red seaweed Chondrus crispus decreased the accumulation of α-synulein and protected the worms from the neuronal toxin-, 6-OHDA, induced dopaminergic neurodegeneration. These effects were associated with a corrected slowness of movement. We also showed that the enhancement of oxidative stress tolerance and an up-regulation of the stress response genes, sod-3 and skn-1, may have served as the molecular mechanism for the C. crispus-extract-mediated protection against PD pathology. Altogether, apart from its potential as a functional food, the tested red seaweed, C. crispus, might find promising pharmaceutical applications for the development of potential novel anti-neurodegenerative drugs for humans. 相似文献

Codium fragile and Chondrus crispus are, respectively, green and red seaweeds which are abundant along the North Atlantic coasts. We investigated the chemical composition and antiviral activity of enzymatic extracts of C. fragile (CF) and C. crispus (CC). On a dry weight basis, CF consisted of 11% protein, 31% neutral sugars, 0.8% sulfate, 0.6% uronic acids, and 49% ash, while CC contained 27% protein, 28% neutral sugars, 17% sulfate, 1.8% uronic acids, and 25% ash. Enzyme-assisted hydrolysis improved the extraction efficiency of bioactive materials. Commercial proteases and carbohydrases significantly improved (p ≤ 0.001) biomass yield (40%–70% dry matter) as compared to aqueous extraction (20%–25% dry matter). Moreover, enzymatic hydrolysis enhanced the recovery of protein, neutral sugars, uronic acids, and sulfates. The enzymatic hydrolysates exhibited significant activity against Herpes simplex virus (HSV-1) with EC50 of 77.6–126.8 μg/mL for CC and 36.5–41.3 μg/mL for CF, at a multiplicity of infection (MOI) of 0.001 ID50/cells without cytotoxity (1–200 μg/mL). The extracts obtained from proteases (P1) and carbohydrases (C3) were also effective at higher virus MOI of 0.01 ID50/cells without cytotoxity. Taken together, these results indicate the potential application of enzymatic hydrolysates of C. fragile and C. crispus in functional food and antiviral drug discovery. 相似文献

Thirty-four adult male buffaloes with clinical signs of 'earsore' were used to determine the efficacy of ivermectin against Stephanofilaria zaheeri. On the basis of parasite counts in skin scrapings, ivermectin was effective as a microfilaricide. Adult parasite counts were reduced, but removal of adults was incomplete. Clinical improvement was recorded in more than half of the treated buffaloes, but constant reinfection meant that meaningful interpretations were not possible. 相似文献

Little is known about the specificity of Bartonella spp. immunofluorescent antibody (IFA) assays in dogs. Bacteremia in sick dogs most often has been associated with Bartonella henselae (Bh), Bartonella vinsonii subspecies berkhoffii (Bvb), and Bartonella koehlerae (Bk). Clarification of the diagnostic utility of IFA serology when testing against these organisms is needed.Objective

Serum from naïve SPF dogs shown to be negative for Bartonella bacteremia did not react with any of the 6 Bartonella antigens by IFA testing. Dogs experimentally infected with Bvb genotype I developed Abs against homologous antigens, with no cross‐reactivity to heterologous Bvb genotypes, Bh H‐1, SA2 strains, or to Bk. A study has been made of the population ecology of large mammals of Silent Valley and contiguous forests of South India. The vegetation of Silent Valley and Attapadi forests are mostly west coast tropical evergreen and semi-evergreen types. In the adjoining forests Nilambur and Nilgiris, the vegetation is southern secondary moist mixed deciduous and southern montane wet temperature, respectively. Silent Valley and its contiguous forests harbour 25 species of large and medium-sized mammals, including two endangered endemic species and many others that are endangered and protected. The lion-tailed macaque, Nilgiri tahr and tiger were sighted in and around the submergence area of the proposed Silent Valley hydroelectric project. If constructed, the impact of this dam on the populations of larger mammals in the area would be severe. 相似文献

Human–wildlife conflict is a serious challenge undermining the integrity of protected areas in developing countries. Developing effective human–wildlife conflict mitigation strategies requires an understanding of the conflict patterns, species involved and attitudes of local people living along protected area boundaries. We hypothesised that (1) there was a high level of human–wildlife conflict and (2) the local people would have less favourable attitudes towards problematic wild animals. We assessed patterns of human–wildlife conflict and attitudes of local people along the boundary of Chebera Churchura National Park, Ethiopia from 2012 to 2014. A total of 354 households were selected randomly for interview. A questionnaire survey, focus group discussions and direct field observations were carried out in the selected villages. The major types of human?wildlife conflict in the area include crop raiding, livestock predation, increased risk of livestock diseases and direct threats to human life. A majority of the respondents (68.1%) faced crop damage and domestic animal loss, 12.3% reported threat to humans and 0.3% reported that the wildlife might cause diseases. Close proximity of the villages to the park and seasons influenced livestock predation intensity with highest predation in the wet season (56.0%). To mitigate these problems, the local people utilised various traditional methods, including guarding. Most respondents had positive attitudes towards the conservation of wildlife. However, as the frequency of conflicts increased in the last five years, the attitudes of local people might change. Active measures are to be implemented to mitigate the problem and safeguard the future of the wildlife around the park. The park has enormous potential to benefit more local people by implementing a participatory management approach to conservation. 相似文献
