The nucleotide sequence of the region of Gardner-Rasheed feline sarcoma virus (GR-FeSV) encoding its primary translation product, p70gag-fgr, has been determined. From the nucleotide sequence, the amino acid sequence of this transforming protein was deduced. Computer analysis indicates that a portion of P70gag-fgr has extensive amino acid sequence homology with actin, a eukaryotic cytoskeletal protein. A second region of P70gag-fgr is closely related to the tyrosine-specific kinase gene family. Thus, the v-fgr oncogene appears to have arisen as a result of recombinational events involving two distinct cellular genes, one coding for a structural protein and the other for a protein kinase.  相似文献

Structural modification of photosynthetic reaction centers is an important approach for understanding their charge-separation processes. An unprecedented persistent structural transformation of the special pair (dimer) of bacteriochlorophyll molecules can be produced by light absorption alone. The nonphotochemical hole-burned spectra for the reaction center of Rhodopseudomonas viridis show that the phototransformation leads to a red shift of 150 wave numbers for the special pair's lowest energy absorption band, P960, and a comparable blue shift for a state at 850 nanometers, which can now be definitively assigned as being most closely associated with the upper dimer component. Additional insights on excited-state electronic structure include the identification of a new state.  相似文献

To examine the combined effects of phosphorus (P) nutrition and CO2 on photosynthesis, chlorophyll fluorescence (CF), and nutrient utilization and uptake, two controlled‐environment experiments were conducted using 0.01, 0.05 and 0.20 mM external phosphate each at ambient and elevated CO2 (aCO2: 400 and eCO2: 800 µmol mol?1, respectively). The CF parameters were affected more by P nutrition than by CO2 treatment. Photoinhibition of photosystem II (PSII) was due to increased minimal CF (Fo′) and decreased maximal CF (Fm′), and efficiency of energy harvesting (Fv′/Fm′). In addition, reduced electron transport rate (ETR), the quantum yield of PSII (ΦPSII) and CO2 assimilation ( ), and overall photochemical quenching in the P‐deficient leaves led to reduction in the efficiency of energy transfer to the PSII reaction center. Stimulation in the ΦPSII/ and photorespiration (ETR/Pnet) was found under P deficiency, whereas the opposite was the case under CO2 enrichment. On average, photosynthetic rate (Pnet) and stomatal conductance declined by 50–53% at 0.05 mM P and by 70–72% at 0.01 mM P as compared to the 0.20 mM P treatment. However, P deficiency, especially at eCO2, tended to increase the intrinsic water‐use efficiency. In the P‐deficient plants, the decline in the P and N utilization efficiency (up to 91%) of biomass production was mainly associated with greater reduction in the biomass relative to the tissue P concentration as the P supply was reduced. However, it was significantly stimulated by eCO2 especially at higher P supply. The CO2 × P interaction was observed for some parameters such as Fo′, Fm′, P utilization efficiencies of photosynthesis and biomass production that might be attributed to the irresponsiveness of these parameters to eCO2 under low P treatment. Thus, P deficiency limited the beneficial effect of eCO2. A close relationship between total biomass and photosynthesis with the P and N utilization or uptake efficiencies was found. The P utilization efficiency of Pnet appeared to be stable across a range of leaf P concentrations, whereas the N‐utilization efficiency markedly increased with leaf P and differed between CO2 levels. An apparent effect of both the treatments (P and CO2) on N‐uptake and utilization efficiency also indicated the alteration in N acquisition and assimilation in cotton plants.  相似文献

This study was designed to investigate the cancer preventive activities of wheat bran (WB) oil. We studied the colon cancer preventive effects of WB oil and its subfractions in the Apc(min/+) mouse model, a recognized mouse model for human colorectal cancer, and used human colon cancer cell lines (HCT-116 and HT-29) to identify possible active fractions in WB oil. Our results showed that the oil fraction of WB was more active than the water fraction against the growth of human colon cancer cell lines and that 2% WB oil significantly inhibited the overall tumorigenesis by 35.7% (p < 0.0001) in the Apc(min/+) mouse model. The WB oil was further fractioned into nonpolar lipids and phytochemicals and the phytochemical fraction was fractionated into phytosterols and phytosterol ferulates, 5-alk(en)ylresorcinols, and unidentified constituents by normal phase silica gel column chromatography. Results on cell culture showed that the phytochemical fraction had a higher inhibitory effect on HCT-116 human colon cancer cells than that of WB oil, whereas the nonpolar lipid fraction had less growth inhibitory effectiveness. However, neither fractions showed a stronger inhibition than WB oil in the Apc(min/+) mouse model. The current results demonstrate, for the first time, the intestinal cancer preventive activity of WB oil. The active ingredients, however, remain to be identified.  相似文献

The surface runoff potential of trifloxysulfuron {N-[(4,6-dimethoxy-2-pyrimidinyl)carbamoyl]-3-(2,2,2-trifluoroethoy)-pyridin-2-sulfonamide sodium salt} in cotton (Gossypium hirsutum L.) production systems has not been evaluated. The objectives of this study were to (i) determine sorption/desorption coefficients for trifloxysulfuron; (ii) quantify foliar washoff of trifloxysulfuron when applied to cotton at the five-leaf stage; and (iii) determine the surface runoff potential of trifloxysulfuron when applied to cotton at the five-leaf stage and to bare soil. Freundlich sorption and desorption coefficients were 1.15 and 1.22, respectively. Sorption data indicated that trifloxysulfuron was moderately sorbed to soil and that it will be transported primarily in the dissolved phase of surface runoff. Foliar washoff studies revealed that approximately 91% of trifloxysulfuron applied to cotton at the five-leaf stage was available for washoff 72 h after application. Simulated rainfall (7.5 cm h-1) applied 1 day after herbicide application (7.9 g ha-1) resulted in average concentrations of trifloxysulfuron in surface runoff water of 0.8 microg L-1 for bare plots and 1.3 microg L-1 for cotton plots. Cumulative trifloxysulfuron losses in surface runoff from bare plots and cotton plots were 0.13 and 0.21 g ha-1, respectively. These values correspond to fractional losses of 1.7% for bare plots and 2.7% for cotton plots. Greater runoff losses of trifloxysulfuron from cotton plots were attributed to foliar washoff. Trifloxysulfuron runoff losses may be curtailed if the herbicide is applied early postemergence when canopy coverage is minimal, thereby reducing the potential for foliar washoff.  相似文献

Seeds are vulnerable to a variety of threats resulting in extinction. Collection of seeds and saving them in secure facilities—seedbanks—to be shared for breeding disease-resistant varieties is the best solution. Working in the spirit of the International Treaty on Plant Genetic Resources for Food and Agriculture, the Global Crop Diversity Trust, the Consultative Group for International Agricultural Research, and the Food and Agriculture Organization played a vital role in establishing over 1,700 seedbanks worldwide. Svalbard Global Seed Vault, known as Doomsday Vault, is an example of a global backup system for the planet's plant genetic resources.  相似文献

Harvest time (HT) and nutrient management may have an effect on switchgrass (Panicum virgatum L.) yield and nutrient cycling. Experimental objectives were to quantify HT and nitrogen (N) rate effects on dry matter yield (DMY), nutrient concentration, and N use. “Alamo” switchgrass was grown under four N rates (0, 40, 80, and 120 kg ha?1) with six monthly HT (May to October) in a randomized complete block design experiment with a split-plot arrangement. Yield increased cubically and quadratically with HT in Years 1 and 2. Aftermath yield decreased linearly with HT in both years. N rate increased yield in Year 2 only with no yield benefit at rates >40 kg N ha?1. Nutrient concentration decreased from May to October, while its removal was determined by DMY. N use and recovery were greatest at 40 kg N ha?1 and declined with additional N inputs. Results indicate that harvesting post-frost-kill produced greater DMY with less nutrient concentration and removal.  相似文献

Balanced autosomal translocations are a known cause for repeated early embryonic loss (REEL) in horses. In most cases, carriers of such translocations are phenotypically normal, but the chromosomal aberration negatively affects gametogenesis giving rise to both genetically balanced and unbalanced gametes. The latter, if involved in fertilization, result in REEL, whereas gametes with the balanced form of translocation will pass the defect into next generation. Therefore, in order to reduce the incidence of REEL, identification of translocation carriers is critical. Here, we report about a phenotypically normal 3‐year‐old Arabian mare that had repeated resorption of conceptuses prior to day 45 of gestation and was diagnosed with REEL. Conventional and molecular cytogenetic analyses revealed that the mare had normal chromosome number 64,XX but carried a non‐mosaic and non‐reciprocal autosomal translocation t(4;10)(q21;p15). This is a novel translocation described in horses with REEL and the first such report in Arabians. Previous cases of REEL due to autosomal translocations have exclusively involved Thoroughbreds. The findings underscore the importance of routine cytogenetic screening of breeding animals.  相似文献
