Guazatine is a fungicide used in agriculture to control a wide range of seed-borne diseases of cereals and other vegetable foods. In this work, a LC-ESI-MS method was developed for the quantitative detection of guazatine residues in maize and hard wheat. Quantitative data were determined for the residues of the main diamines, triamines, and tetramines that cover more than 87% of the total contents of the mixture. The mean recoveries from the fortified cereals at 0.050 mg/kg ranged from 81 to 86%, with the coefficients of variation (CVs) ranging from 0.9 to 5.5% (n = 5). At 0.025 mg/kg, the recoveries ranged from 78 to 87%, with the CVs ranging from 0.8 to 6.3% (n = 5). The limits of quantification have been estimated to be 0.010, 0.004, 0.002, 0.002, 0.005, and 0.002 mg/kg, respectively, for GN, GG, GNG, GGN, GGG, and GGGG in maize and hard wheat (S/N ratio >10).  相似文献

A protocol based on Co-operational PCR has been successfully applied to the detection of phytoplasmas. A triprimer reaction coupled with hybridization using general and specific probes permitted detection of ' Candidatus Phytoplasma mali', ' Ca . Phytoplasma prunorum' and ' Ca . Phytoplasma pyri', and their identification as members of 16S ribosomal quarantine group X. The sensitivity of this method was at least one hundred times greater than conventional PCR and similar to that achieved by nested PCR and real-time PCR. The method was validated by testing field samples collected from Malus , Prunus and Pyrus spp. and Olea europaea and compared with seven phytoplasmas maintained in Catharanthus roseus .  相似文献

The stability of extra virgin olive oils is mainly due to their relatively low fatty acids unsaturation and to the antioxidant activity of some of the unsaponifiable components. We studied the activity of alpha-tocopherol in extra virgin olive oil in its natural state, using new and simple oxidizing conditions in "thin layer" and "bulk phase". Oxidation time course was monitored at 37 degrees C or 75 degrees C. A storage test was also performed. Two parameters were evaluated: depletion of alpha-tocopherol and formation of PUFA hydroperoxides, measured as conjugated dienes (CD) and peroxide value. The value of complex polyphenols was also measured. alpha-Tocopherol concentration decreased in function of time and temperature and showed a strong inverse correlation with the increase of CD. When alpha-tocopherol reached a "threshold value" of 60-70 mg/kg, a significant increase of CD formation was observed, together with a good correlation between CD and peroxide value. The initial amount of alpha-tocopherol is one of the components that influences oil oxidative susceptibility.  相似文献

? Context

Prescribed burning is increasingly recommended to control encroaching shrublands in the Mediterranean area.

? Aims

The aims of this paper are to analyze the fuel structural characteristics of Spartium junceum and how they influence fire behaviour during prescribed burning.

? Methods

Two winter–spring prescribed burns were conducted in 2009 and 2011. Fuel load and structure of S. junceum shrubs were assessed using the Cube Method, and shrub 3-D models were built using the FIRE PARADOX FUEL MANAGER software. Allometric equations to estimate S. junceum fuel load were developed. During burning, thermocouples measured temperature variations, which were then analyzed relative to fuel characteristics.

? Results

Fuel load components and distribution were strictly related to shrub height; in tall shrubs, most of the fine fuel was more than 1.5 m aboveground. Due to fuel vertical discontinuity, not all shrubs were burned in the fires, but wind increased fire sustainment and fuel consumption. Maximum temperatures (over 800 °C) and residence times were positively related to fuel load.

? Conclusion

S. junceum tall shrublands represent high hazard formations due to their elevated fuel load, mostly in fine fuel fractions. Vertical discontinuity among fuel strata limits fire propagation in mild weather conditions. Winter–spring prescribed burning cannot eliminate S. junceum shrublands, but do create shrub cover discontinuity. As S. junceum has fire-adapted morphological traits, a single burn is insufficient to control it.  相似文献

A phytoplasma was detected in annual blue grass (Poa annua L. Fienardo), exhibiting white leaf symptoms, that was grown in the fields near Caserta in southern Italy. Based on restriction fragment length polymorphism analysis of PCR-amplified 16S rDNA sequences, the phytoplasma associated with annual blue grass white leaf disease was identified as a new member of phytoplasma 16S rRNA group XI (16SrXI) (type strain, rice yellow dwarf phytoplasma). The annual blue grass white leaf phytoplasma is most closely related to Bermuda grass white leaf phytoplasma found in Asia. Annul blue grass white leaf and Bermuda grass white leaf phytoplasmas were designated as the third subgroup (16SrXI-C) of group XI. This is the first report that a plant pathogenic phytoplasma belonging to group 16SrXI is present on the European continent.  相似文献

Qualitative/quantitative phytochemical variations were observed in dried flowering tops of cultivated Hypericum perforatum L. cv. Zorzi infected by phytoplasmas of the "ash yellows" class, identified by direct and nested polymerase chain reaction (PCR); this is the first report of ribosomial group 16SrVII phytoplasmas in St. John's Wort. Methanolic extracts of healthy and infected plants were separated by reversed phase high-performance liquid chromatography to quantify naphthodianthrones and flavonoids, while essential oils were analyzed by means of gas chromatography (GC)-GC/MS. The affected plants exhibited decreased amounts of rutin (1.96 +/- 0.23 vs 4.96 +/- 0.02 mg/g), hyperoside (2.38 +/- 0.21 vs 3.04 +/- 0.05 mg/g), isoquercitrin (1.47 +/- 0.04 vs 3.50 +/- 0.08 mg/g), amentoflavone (0.12 +/- 0.01 vs 0.39 +/- 0.02 mg/g), and pseudohypericin (1.41 +/- 0.23 vs 2.29 +/- 0.07 mg/g), whereas the chlorogenic acid content was doubled (1.56 +/- 0.11 vs 0.77 +/- 0.02 mg/g). Hypericin, quercitrin, and quercetin contents were not severely affected. The essential oil yield was drastically reduced in infected material (0.11 vs 0.75% in healthy material) and revealed an increased abundance of sesquiterpenes (beta-caryophyllene, delta-elemene, and germacrene D, in particular) and a matching decrease in monoterpene hydrocarbons and aliphatics. The consequences that the phytopathological condition of cultivated H. perforatum plants has on the commercial quality, market value, and therapeutic efficacy are outlined.  相似文献

Hot peppers are a good source of dietary antioxidants, encompassing, apart from widespread compounds (flavonoids, phenolic acids, carotenoids, vitamin A, ascorbic acid, tocopherols), also specific constituents such as the pungent capsaicinoids (capsaicin, dihydrocapsaicin, and related analogues). We have shown that capsinoids also show remarkable antioxidant activity. These benign analogues of capsaicin could protect linoleic acid against free radical attack in simple in vitro systems, inhibiting both its autoxidation and its iron- or EDTA-mediated oxidation. These properties were retained in some simple synthetic analogues (vanillyl nonanoate and its dimerization products). Capsiate, dihydrocapsiate, and their analogues were devoid of pro-oxidant activity and showed a highly significant antioxidant activity in all systems investigated. Vanillyl nonanoate, a simple capsinoid mimic, was also tested on cell cultures for cytotoxic activity and the capacity to inhibit FeCl(3)-induced oxidation.  相似文献

An experiment was carried out in a young high-density olive grove (556 plants ha−1Olea europaea L., cv Coratina) located in Southern Italy to evaluate the effect of different soil water availability on the vegetative and productive performances of olive trees also looking into the quality of the resulting oils. Trials were carried out over a 3-year period on trees subjected to irrigation and grown under rainfed conditions. Vegetative tree response, as shoot elongation and trunk diameter, was evaluated. Yield per plant, fruit characteristics, oil quality indices (free fatty acid content, peroxide value, UV adsorption at 232 and 270 nm, total phenols, α-tocopherol content) were determined for both irrigated and non-irrigated treatments in the ‘on’ years 1997 and 1999 (6th and 8th year after planting, respectively).  相似文献

The importance of n-3 polyunsaturated fatty acid (n-3 PUFA) intake has long been recognized in human nutrition. Although health benefits, n-3 PUFA are subject to rapid and/or extensive oxidation during processing and storage, resulting in potential alteration in nutritional composition and quality of food. Bottarga, a salted and semi-dried mullet ( Mugil cephalus ) ovary product, is proposed as an important source of n-3 PUFA, having high levels of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). In this work, we investigated the extent of lipid oxidation of grated bottarga samples during 7 months of storage at -20 °C and room temperature under light exposure. Cell viability, lipid composition, and lipid peroxidation were measured in intestinal differentiated Caco-2 cell monolayers after 6-48 h of incubation with lipid and hydrophilic extracts obtained from bottarga samples at different storage conditions. The storage of bottarga did not affect the n-3 PUFA level, but differences were observed in hydroperoxide levels in samples from different storage conditions. All tested bottarga extracts did not show a toxic effect on cell viability of differentiated Caco-2 cells. Epithelial cells incubated with bottarga oil had significant changes in fatty acid composition but not in cholesterol levels with an accumulation of EPA, DHA, and 22:5. Cell hydroperoxides were higher in treated cells, in relation to the oxidative status of bottarga oil. Moreover, the bottarga lipid extract showed an in vitro inhibitory effect on the growth of a colon cancer cell line (undifferentiated Caco-2 cells).  相似文献

The presence of phytoplasma inFragaria ananassa x Duch cv Senga Sengana showing strawberry green petals symptoms was observed by electron microscopy of phloem tissue. No phytoplasmas were found in asymptomatic strawberry plants used as controls. Nucleic acids extracted from these plants were used in nested-PCR assays with primers amplifying 16S rRNA sequences specifie for phytoplasmas. Bands of 1.2 kb were obtained and the subsequent nested-PCR with specific primers and RFLP analyses allowed to classify the detected phytoplasmas in the aster yellows group (16SrI). They belonged to the subgroup I-C of which type strain is clover phyllody phytoplasma.  相似文献
