Dirofilaria immitis is the causative agent of cardiopulmonary dirofilariasis in the Canine family. The aim of the study was to evaluate the efficacy of the ethanolic extract of Azadirachta indica leaves (EEA) against the microfilaria (mf) of D. immitis in vitro. EEA was evaluated for different compound classes through HPTLC. Relative motility, mortality and morphological alterations were observed in the mf after exposure to EEA. The effect of EEA on redox status in the treated mf was evaluated by some key enzymatic and non-enzymatic parameters. An enhanced reactive oxygen species (ROS) level in the treated mf along with altered redox status was evident. DNA fragmentation and terminal-deoxynucleotidyl-transferase dUTP nick end labeling (TUNEL) confirmed apoptosis. In addition, western blotting revealed down-regulation of anti-apoptotic protein and up-regulation of pro-apoptotic proteins. Taken together, the microfilaricidal activity of EEA can be attributed to its capacity to inflict oxidative stress culminating in apoptosis.  相似文献

Four insecticides, viz., BHC, phorate, carbofuran, and fenvalerate, were applied at the rate of 7.5, 1.5, 1.0, and 0.35 kg a.i. ha(-)(1), respectively, to investigate their effects on the growth and activities of N(2)-fixing and phosphate-solubilizing microorganisms in relation to the availability of N and P in laterite (Typic Orchragualf) soil. Insecticides in general, and BHC and phorate in particular, stimulated the proliferation of aerobic nonsymbiotic N(2)-fixing bacteria and phosphate-solubilizing microorganisms and also their biochemical activities, such as nonsymbiotic N(2)-fixing and phosphate-solubilizing capacities, which resulted in greater release of available N (NH(4)(+) and NO(3)(-)) and P in soil. All the insecticides were persistent in soil for a short period of time, and the rate of dissipation was highest for fenvalerate followed by phorate, carbofuran, and BHC, depicting the half-lives (T(1/2)) 8.8, 9.7, 16.9, and 20.6 days, respectively. The insecticides followed first-order reaction kinetics during their dissipation in soil.  相似文献

Sesamol (from Sesamum indicum) is a dietary compound, which is soluble in aqueous as well as lipid phases. Free radical scavenging reactions of sesamol, 5-hydroxy-1,3-benzodioxole, have been studied using a nanosecond pulse radiolysis technique. Sesamol efficiently scavenges hydroxyl, one-electron oxidizing, organo-haloperoxyl, lipid peroxyl, and tryptophanyl radicals. Its antioxidant activity has also been evaluated with cyclic voltammetry. In biochemical studies, it has been found to inhibit lipid peroxidation, hydroxyl radical-induced deoxyribose degradation, and DNA cleavage. These antioxidant and free radical scavenging activities of sesamol have been reported in the paper.  相似文献

A blood cell type termed crystal cell in Drosophila functions in clotting and wound healing and requires Notch for specification and maintenance. We report that crystal cells express elevated levels of Sima protein orthologous to mammalian hypoxia-inducible factor-α (Hif-α) even under conditions of normal oxygen availability. In these platelet-like crystal cells, Sima activates full-length Notch receptor signaling via a noncanonical, ligand-independent mechanism that promotes hemocyte survival during both normal hematopoietic development and hypoxic stress. This interaction initiates in early endosomes, is independent of Hif-β (Τangο in Drosophila), and does not activate hypoxia response targets. Studies in vertebrate myeloid cells have shown a similar up-regulation of Hif-α protein in well-oxygenated environments. This study provides a mechanistic paradigm for Hif-α/Notch interaction that may be conserved in mammals.  相似文献

Two diploid cytotypes of Chenopodiumalbum have earlier been proposed as the probable ancestors ofhexaploid C. album in Gangetic Plainof West Bengal on the basis of cytomorphological data. The present study withthe diploid cytotypes of C. album (onebroad leaved, another narrow leaved), one wild species, C.murale and the hexaploid C.album utilizing parameters like seed protein, isozyme,RAPD, tissue culture along with conventional cytomorphological data supports ourearlier hypothesis of allopolyploid origin of hexaploid C.album. The results indicate that all the three diploidprogenitors have contributed for the evolution of present day semi-cultivatedhexaploid C. album. This study furthersuggests that the diploid cytotypes had been evolved sympatrically.  相似文献

Sterols (sitosterol, cholesterol, stigmasterol, ergosterol, and 7-dehydrocholesterol) and sitostanol have been converted in high to near-quantitative yields to the corresponding long-chain acyl esters via esterification with fatty acids or transesterification with methyl esters of fatty acids or triacylglycerols using lipase from Candida rugosa as biocatalyst in vacuo (20-40 mbar) at 40 degrees C. Neither organic solvent nor water is added in these reactions. Under similar conditions, cholesterol has been converted to cholesteryl butyrate and steroids (5alpha-pregnan-3beta-ol-20-one or 5-pregnen-3beta-ol-20-one) have been converted to their propionic acid esters, both in moderate to high yields, via transesterification with tributyrin and tripropionin, respectively. Reaction parameters studied in esterification include the temperature and the molar ratio of the substrates as well as the amount and reuse properties of the C. rugosa lipase. Lipases from porcine pancreas, Rhizopus arrhizus, and Chromobacterium viscosum are quite ineffective as biocatalysts for the esterification of cholesterol with oleic acid under the above conditions.  相似文献
