Golden hamsters were selected for large (high selection direction: H) and small (low selection direction: L) testis size, as measured in live males at age 13 weeks over six generations. The selection response and correlated responses in female reproductive traits were evaluated in terms of the divergence between H and L lines for testis size. In males, the differences in testis size at 13 weeks, testis weight at 19 weeks, and body weight at 13 and 19 weeks were significant at the 1% level from generation 1 onward. The realized heritability of testis size, as estimated from regression of the selection responses on effective selection differentials, was 0.29. This was similar to the heritability estimated by using the multiple‐trait animal model restricted maximum likelihood (0.30). In females, significant differences in ovulation rate at the 5% level were detected in generations 5 and 6. Litter size 1 day after birth also diverged between the H and L lines, but the difference was not consistently significant. The realized genetic correlation between testis size and ovulation rate was 0.67.  相似文献

To investigate the effects of amla on renal dysfunction involved in oxidative stress during the aging process, we employed young (2 months old) and aged (13 months old) male rats and administered SunAmla (Taiyo Kagaku Co., Ltd., Japan) or an ethyl acetate (EtOAc) extract of amla, a polyphenol-rich fraction, at a dose of 40 or 10 mg/kg body weight/day for 100 days. The administration of SunAmla or EtOAc extract of amla reduced the elevated levels of serum creatinine and urea nitrogen in the aged rats. In addition, the tail arterial blood pressure was markedly elevated in aged control rats as compared with young rats, while the systolic blood pressure was significantly decreased by the administration of SunAmla or EtOAc extract of amla. Furthermore, the oral administration of SunAmla or EtOAc extract of amla significantly reduced thiobarbituric acid-reactive substance levels of serum, renal homogenate, and mitochondria in aged rats, suggesting that amla would ameliorate oxidative stress under aging. The increases of inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 expression in the aorta of aging rats were also significantly suppressed by SunAmla extract or EtOAc extract of amla, respectively. Moreover, the elevated expression level of bax, a proapoptotic protein, was significantly decreased after oral administration of SunAmla or EtOAc extract of amla. However, the level of bcl-2, an antiapoptotic protein, did not show any difference among the groups. The expressions of renal nuclear factor-kappaB (NF-kappaB), inhibitory kappaB in cytoplasm, iNOS, and COX-2 protein levels were also increased with aging. However, SunAmla or EtOAc extract of amla reduced the iNOS and COX-2 expression levels by inhibiting NF-kappaB activation in the aged rats. These results indicate that amla would be a very useful antioxidant for the prevention of age-related renal disease.  相似文献

Fresh and decomposed dissolved organic matter (DOM) derived from 13 plant biomass and animal manure sources was characterized using multidimensional fluorescence spectroscopy with parallel factor analysis (PARAFAC), high-performance size-exclusion chromatography, and UV-vis spectroscopy. The PARAFAC analysis modeled seven fluorescence components: tryptophan-like, tyrosine-like, and five humic substance-like components. For most of the plant-derived DOM solutions, decomposition significantly affected the concentration of three humic substance-like-associated components, increasing two and decreasing one. The effect of decomposition upon DOM derived from animal manures was dependent on the manure source. For a majority of the DOM extracts, the ratio of fluorescence intensity to absorptivity at 254 nm increased following decomposition, indicating that fluorescing DOM compounds were generally more resistant to biodegradation than nonfluorescing UV-absorbing compounds. Molar absorptivity, humification index (HIX), and apparent molecular weight (MWAP) increased by 38.0, 38.8, and 370%, respectively, following decomposition. Spearman correlation analysis showed a strong positive relationship between the humic substance-like components and the DOM MWAP, absorptivity, and HIX. The results of this study support the use of multidimensional fluorescence spectroscopy with PARAFAC as a method to monitor the decomposition of carbon-rich soil amendments such as crop residues, green manures, and animal manures.  相似文献

In the present study, 42 multiparous Holstein cows were used to investigate the relationship between fat accumulation in the liver and dry matter intake, milk yield and blood metabolites. Based on the percentage of fat in the liver cell at 2 weeks post‐parturition, the cows were classified into three groups. These groups were: (i) less than 10% of fat (normal group, n = 29); (ii) 10–20% of fat (mild group, n = 6); and (iii) more than 20% of fat (moderate group, n = 7). The bodyweight of the moderate group was high (771 kg) before calving. The sufficiency rates of total digestible nutrients (TDN) were remarkably decreased (approximately 65%) in early lactation. The milk fat yield and milk fat composition of the moderate group were higher (P < 0.05) than the other groups at 1 and 2 weeks post‐parturition. It was suggested that non‐esterified fatty acids (NEFA) mobilized from adipose tissues was directly used by the mammary gland for synthesis of milk fat. The percentage of bromsulfalein (BSP) retention of the moderate group was high (21.1%) at 30 min, and it showed that the BSP clearance function was significantly decreased. The concentrations of NEFA, β‐hydroxybutyric acid and glucose were appropriate indicators of energy status; however, aspartate aminotransferase, γ‐glutamyl transpeptidase and total bilirubin were not sensitive indicators of a moderate fatty liver. Thus, high‐yielding cows that calve in an overweight condition are more likely to develop excessive fat accumulation in the liver because of great mobilization from adipose tissues post‐parturition. In cows with a moderately fatty liver, a decrease in TDN sufficiency rates, an increase of milk fat yield and a reduction of liver function were observed in early lactation. The increase of serum NEFA and milk fat composition resulting from mobilization of adipose tissues helped to diagnose moderate fatty liver.  相似文献

European eel, Anguilla anguilla L., migrating to the sea encounter many man‐made structures that can hamper and delay migration or induce mortality. Studying small‐scale behavioural movements in front of these man‐made structures could provide insight in further mitigating adverse effects. The behaviour of eel approaching a trash rack in front of a large pumping station was investigated using a dual‐frequency identification sonar (DIDSON). Eels approaching the trash rack swam through the rack (40.5%) but also showed turning behaviour at (44.7%) or in front of the rack (14.7%). Eels approaching the rack had varying body positions, predominantly head or tail first, but also curled up into a ball or drifted sideways. After turning in front or at the trash rack, eels showed upstream and downwards swimming towards the canal bottom. The results suggest a stepwise response to potential cues, when firstly the body position is changed in such a way that secondly, later on, enhances eventual fast upstream escapement when perceived necessary. Implications for management of these behavioural observations are discussed.  相似文献

Objective This study investigated whether the bispectral index (BIS monitor) corresponded with the clinical assessment of anaesthetic depth in dogs. Study design Prospective clinical study. Animals Sixty‐five dogs undergoing anaesthesia for surgery. Methods Dogs were assigned to one of three different anaesthetic techniques. A three point scale was devised to determine the clinical depth of anaesthesia (CDA); CDA 1 represented light, CDA 2 surgical and CDA 3 excessive depth of anaesthesia. BIS values were recorded and CDA assessed at specific times and points throughout surgery. Data were statistically analysed using mixed model regression. Results Clinical depth of anaesthesia was assessed as CDA 1 on 68, 2 on 748 and 3 on four occasions. The BIS recorded for CDA 1 differed significantly from that for CDA 2 (p < 0.001). However, individual BIS values measured at light and surgical levels of anaesthesia overlapped considerably. The sensitivities and specificities calculated for BIS to diagnose CDA 1 compared to CDA 2 in the three anaesthetic protocols were 28–86% and 55–85%. The accompanying positive predictive value was 0.08–0.29 and the negative predictive value was 0.95–0.97. End‐tidal isoflurane concentrations (anaesthetic techniques 1 and 3) and propofol infusion (technique 2) at CDA 1 was significantly lower than those at CDA 2 (p = 0.001). Conclusions Although BIS values overall distinguished between CDA scores, the calculated specificities, sensitivities and predictive values were low, and there were anomalous results in individual cases. Clinical relevance The use of the BIS as the sole method to determine anaesthetic depth in dogs is imprudent.  相似文献

The aim of the present study was to clarify the effects of hypothalamic dopamine (DA) on the secretion of growth hormone (GH) in goats. The GH‐releasing response to an intravenous (i.v.) injection of GH‐releasing hormone (GHRH, 0.25 μg/kg body weight (BW)) was examined after treatments to augment central DA using carbidopa (carbi, 1 mg/kg BW) and L‐dopa (1 mg/kg BW) in male and female goats under a 16‐h photoperiod (16 h light, 8 h dark) condition. GHRH significantly and rapidly stimulated the release of GH after its i.v. administration to goats (P < 0.05). The carbi and L‐dopa treatments completely suppressed GH‐releasing responses to GHRH in both male and female goats (P < 0.05). The prolactin (PRL)‐releasing response to an i.v. injection of thyrotropin‐releasing hormone (TRH, 1 μg/kg BW) was additionally examined in male goats in this study to confirm modifications to central DA concentrations. The treatments with carbi and L‐dopa significantly reduced TRH‐induced PRL release in goats (P < 0.05). These results demonstrated that hypothalamic DA was involved in the regulatory mechanisms of GH, as well as PRL secretion in goats.  相似文献

During surveillance for bovine diarrhea of unknown causes in Japanese black cattle in Kagoshima Prefecture, Japan, we found two types of novel kobu-like viruses in fecal samples of calves. Sequence analyses revealed that they had L protein and 2A protein with H-box/NC sequence motif, which are present in kobuviruses. Phylogenetic analysis revealed that they were related to kobuviruses; however, they clustered apart from other kobuviruses. In the prevalence study of two types of novel kobu-like viruses, 16.9% and 10.4% prevalence of these viruses were observed in the feces of diarrheal calves in this area.  相似文献

Mannose-binding rice lectin (MRL), which is almost identical to the salt-induced protein SalT, binds to mannose and glucose residues. Expression of the MRL gene in response to infection with Magnaporthe oryzae, the rice blast fungus, was stronger in the incompatible interaction than in the compatible. Transgenic rice plants that constitutively over-expressed MRL strongly suppressed the growth of invasive hyphae of the fungus on leaf sheaths and the development of typical susceptible-type lesions on leaf blades, but did not affect penetration by the fungus in comparison with the wild-type. On a polycarbonate plate, purified recombinant MRL inhibited conidial attachment and appressorium formation but not conidial germination. These results suggest that MRL may play an essential role in disease resistance by suppressing development of M. oryzae in situ.  相似文献
