Enteric pythiosis was diagnosed in a 2-year-old Quarter Horse gelding. The horse had signs of colic, which appeared to be alleviated by administration of mineral oil and analgesics, but only temporarily. Intestinal distention developed after initial examination. At surgery, a thick stenotic area was observed in the middle portion of the jejunum. The thick intestine and associated mesentery contained multiple firm nodules of gritty caseous material. Histopathologic findings included sclerosing eosinophilic granulomatous enteritis and peritonitis. Although the lesion resembled a response to migrating parasites and lacked hyphae in initial sections, the gross appearance of the lesion prompted the acquisition of additional sections to detect the etiologic agent.  相似文献

OBJECTIVE: To evaluate the effect of intermittent oral administration of ponazuril on immunoconversion against Sarcocystis neurona in horses inoculated intragastrically with S neurona sporocysts. ANIMALS: 20 healthy horses that were seronegative for S neurona-specific IgG. PROCEDURES: 5 control horses were neither inoculated with sporocysts nor treated. Other horses (5 horses/group) each received 612,500 S neurona sporocysts via nasogastric tube (day 0) and were not treated or were administered ponazuril (20 mg/kg, PO) every 7 days (beginning on day 5) or every 14 days (beginning on day 12) for 12 weeks. Blood and CSF samples were collected on day - 1 and then every 14 days after challenge for western blot assessment of immunoconversion. Clinical signs of equine protozoal myeloencephalitis (EPM) were monitored, and tissues were examined histologically after euthanasia. Results: Sera from all challenged horses yielded positive western blot results within 56 days. Immunoconversion in CSF was detected in only 2 of 5 horses that were treated weekly; all other challenged horses immunoconverted within 84 days. Weekly administration of ponazuril significantly reduced the antibody response against the S neurona 17-kd antigen in CSF. Neurologic signs consistent with EPM did not develop in any group; likewise, histologic examination of CNS tissue did not reveal protozoa or consistent degenerative or inflammatory changes. CONCLUSIONS AND CLINICAL RELEVANCE: Administration of ponazuril every 7 days, but not every 14 days, significantly decreased intrathecal anti-S neurona antibody responses in horses inoculated with S neurona sporocysts. Protocols involving intermittent administration of ponazuril may have application in prevention of EPM.  相似文献

Arbuscular mycorrhizal fungi (AMF) are important functional components of ecosystems. Although there is accumulating knowledge about AMF diversity in different ecosystems, the effect of forest management on diversity and functional characteristics of AMF communities has not been addressed. Here, we used soil inoculum representing three different AM fungal communities (from a young forest stand, an old forest stand and an arable field) in a greenhouse experiment to investigate their effect on the growth of three plant species with contrasting local distributions - Geum rivale, Trifolium pratense and Hypericum maculatum. AM fungal communities in plant roots were analysed using the terminal restriction fragment length polymorphism (T-RFLP) method. The effect of natural AMF communities from the old and young forest on the growth of studied plant species was similar. However, the AMF community from the contrasting arable ecosystems increased H. maculatum root and shoot biomass compared with forest inocula and T. pratense root biomass compared to sterile control. According to ordination analysis AMF inocula from old and young forest resulted in similar root AMF communities whilst plants grown with AM fungi from arable field hosted a different AMF community from those grown with old forest inocula. AMF richness in plant roots was not related to the origin of AMF inoculum. G. rivale hosted a significantly different AM fungal community to that of T. pratense and H. maculatum. We conclude that although the composition of AM fungal communities in intensively managed stands differed from that of old stands, the ecosystem can still offer the ‘symbiotic service’ necessary for the restoration of a characteristic old growth understorey plant community.  相似文献

Angiotensin-converting enzyme (ACE), playing a crucial role in the renin angiotensin aldosterone system, is well-known to catalyze the conversion of the decapeptide angiotensin I into the physiologically active octapeptide angiotensin II, triggering blood pressure increasing mechanisms. To meet the demand for natural phytochemicals as antihypertensive agents in functional food development, extracts prepared from a series of vegetables were screened for their ACE-inhibitory activity by means of a LC-MS/MS-based in vitro assay. By far the highest ACE inhibition was found for a lettuce extract, in which the most active compound was located by means of activity-guided fractionation. LC-MS, NMR spectroscopy, and hydrolysis experiments followed by ion chromatography led to the unequivocal identification of the ACE inhibitor as the previously not reported (S)-malic acid 1'-O-β-gentiobioside. This glycoside represents a novel class of ACE-inhibiting phytochemicals with a low IC(50) value of 27.8 μM. First incubation experiments in saliva and aqueous hydrochloric acid demonstrated the stability of (S)-malic acid 1'-O-β-gentiobioside against salivary glycosidases and stomach acid.  相似文献

The Finnish state forest enterprise, Metsähallitus, defines the regional harvest levels for a 10-year period in a strategic-level natural resources plan. Although this plan defines stand-level harvest schedules for all stands, in practice, it cannot be used, as the harvests need to be clustered in time and in space. It is applied by giving each subregion goals they need to fulfill in a tactical level planning process, and the harvests are manually clustered into predefined groups of adjacent stands (departments). In this study, we developed a hierarchical optimization process making use of departments for clustering the harvests. For each of the departments, 91 different stand-level harvest schedules (plans) were determined using incomes from one period and the forest value at the end as objectives. The department-level plans were then used as alternatives in a region-level goal optimization problem. The resulting hierarchic plan was compared to the stand-level solution of the strategic-level plan which served as a benchmark plan. The hierarchical plan clustered the harvests and achieved the goals set better than the benchmark plan, but the net present income was 3.3% lower. The approach turned out usable, but further developing of the approach is needed to reduce the costs of clustering.  相似文献

In boreal forests ericaceous shrubs often dominate the forest floor vegetation. Nitrogen enrichment has been shown to decrease shrub abundance and in this study we explored whether it also affects the root associated fungal communities. Fine roots of Vaccinium myrtillus were collected in a Norway spruce dominated forest and of Vaccinium vitis-idaea in a Scots pine dominated forest. In both forests, nitrogen enrichment was experimentally induced by adding 12.5 and 50 kg N ha−1 yr−1 for 12 (spruce forest) and four (pine forest) years. Based on terminal restriction fragment length polymorphisms, subcloning and sequencing analyses, the root associated fungal communities were examined. We found 93 fungal species including Asco-, Basidio- and Zygo-mycota. In general, the Rhizoscyphus ericae aggregate was the most dominant and this was followed by Herpotrichiellaceae and Sebacina. Ordination analysis revealed that nitrogen enrichment did not change species composition of the fungal communities in neither the spruce nor the pine forest, while fungal community structures were clearly discriminated between the dominant shrub species in each forest. Similarly, no fungal species showed a significant response to nitrogen enrichment. Therefore, nitrogen enrichment appears to have no effect on root associated fungi of understorey dwarf shrubs in boreal forests, while it is clear that spruce and pine forests harbor distinctive communities of these fungi.  相似文献

OBJECTIVE: The study aimed to evaluate the homocysteine and cobalamin status of German vegans and determine whether the intake of very small amounts of foods of animal origin can improve this status. DESIGN AND SETTING: Cross-sectional study, Germany. SUBJECTS: The dietary and nutritional intakes of 131 vegans (73 women, 58 men; age range: 20.2-82.1 years) were evaluated using a general questionnaire and two food-frequency questionnaires. RESULTS: The prevalence of inadequate cobalamin status in volunteers of the German Vegan Study was 28.2%, and that of hyperhomocysteinaemia, 38.1%. Moderate vegans were affected to a lesser extent than were strict vegans. Duration of veganism and cobalamin concentration were inversely correlated (Spearman's r=-0.175, P=0.047). Folate concentration and erythrocyte aspartic acid aminotransferase activity were not correlated with plasma homocysteine concentration, but duration of veganism correlated positively with homocysteine concentration (Spearman's r=0.319, P<0.001). Cobalamin and homocysteine concentrations were inversely correlated (when controlling for duration of veganism; r=-0.602, P<0.001). CONCLUSION: Cobalamin status needs to be improved in order to minimise the risk of hyperhomocysteinaemia.  相似文献

Pectin gels consist of polysaccharide networks surrounded by water. The gel networks can prevent release of aroma molecules from the gel to the gas phase above. In this study static headspace measurements were performed to correlate aroma concentration in the gas phase above pectin gels to different amounts of the gel ingredients. As a consequence, aroma concentration in the headspace in relation to gel texture, as characterized by rheology measurements, was also studied. Aroma concentration in the headspace above strong gels was low, due to entrapment of aroma molecules within the gel structure. Viscous solutions generally gave a high aroma concentration in the headspace, but owing to a complex matrix, this was lowered when large amounts of the gel ingredients were added. However, a high correlation between interaction terms and square terms of design variables and rheology parameters with aroma compounds indicated nonlinear and complex relationships.  相似文献

Insoluble fiber fractions from 11 fruits and vegetables were investigated for their lignin composition using the derivatization followed by reductive cleavage (DFRC) methodology. To enrich lignin contents and to minimize polysaccharide excess that led to nonanalyzable DFRC chromatograms, the insoluble fibers were degraded by a carbohydrolases mixture. The residues that were found to be representative for the insoluble fiber lignins were analyzed. The investigated fibers differ considerably in their lignin contents and also in their lignin compositions. With the exception of radish fiber, only trace amounts (or none) of the products resulting from p-hydroxyphenyl units were detected. Lignins noticeably differed in the ratio of the DFRC products resulting from syringyl units (S) and guaiacyl (G) units (G/S ratios ranged from approximately 39 to 0.2). The insoluble fiber lignins were classified as G-rich lignins (G/S ratio > 3; carrot, spinach, kiwi, curly kale, radish, and asparagus), S-rich lignins (S/G ratio > 3; rhubarb), or balanced lignins (0.3 < G/S ratio < 3; pear, apple, small radish, and kohlrabi). Information about further structural characteristics, for example, cinnamyl endgroups, was obtained from the analysis of DFRC minor products.  相似文献
