OBJECTIVE: To determine the characteristics of an automated canine C-reactive protein (CRP) assay and evaluate 2 human CRP assays for use in dogs. Animals-56 client-owned dogs with pyometra and 11 healthy control dogs. PROCEDURES: Samples from 11 dogs with high (> 100 mg/L) or low (< 10 mg/L) CRP concentrations (determined by use of a canine ELISA) were evaluated by use of the automated canine CRP assay. Intra- and interassay imprecision was determined (by use of those 2 plasma pools), and assay inaccuracy was assessed by use of logistic regression analysis of results obtained via ELISA and the automated canine CRP assay. Two automated human CRP assays were used to measure plasma CRP concentration in 10 dogs. RESULTS: By use of the ELISA, mean +/- SD plasma CRP concentration was 96.1 +/- 38.5 mg/L and 10.1 +/- 23.2 mg/L in dogs with pyometra and control dogs, respectively. The automated canine assay had intra-assay coefficients of variation (CVs) of 7.8% and 7.9%, respectively, and interassay CVs of 11.1% and 13.1%, respectively. Results from the automated assay were highly correlated with results obtained via ELISA. The human assay results did not exceed 0.4 mg/L in any dog. CONCLUSIONS AND CLINICAL RELEVANCE: The automated canine CRP assay had less interassay imprecision, compared with the ELISA. The 2 human CRP assays were not suitable for analysis of canine plasma samples. The automated canine CRP assay was more precise than the ELISA for serial evaluations of plasma CRP concentration in dogs. 相似文献

Fourteen Hereford steers were used to compare carcass traits, meat quality, and fatty acid composition of beef from cattle grazing tall fescue infected with either wild-type (E+; n = 6) or novel, nil ergot alkaloid (AR542; n = 8) endophyte for 209 d. Average daily gain, live weight, and HCW were greater (P < 0.05) for AR542 cattle than for E+. No differences in LM color or pH were observed between AR542 and E+. Steaks from E+ cattle tended (P = 0.10) to have higher L* and b* than those from AR542 cattle at 0 d of display. Ground beef from E+ cattle also had higher (P < 0.05) L* than AR542 cattle, with no differences in a* or b* at 0 d of display. Color changes during display did not differ for both steaks and ground beef from E+ and AR542. Lipid oxidation levels increased (P < 0.05) during simulated retail display, but they did not differ between endophyte treatments. Adipose tissues from E+ cattle had a higher (P < 0.05) percentage of SFA, and a lower (P < 0.05) percentage of MUFA than adipose from AR542 cattle. Ground beef and i.m. fat had higher (P < 0.05) concentrations of SFA, MUFA, and cis-9, trans-11 isomer of conjugated linoleic acid, and lower (P < 0.05) concentrations of PUFA and PUFA:SFA ratio than s.c. fat. The n-6:n-3 fatty acid ratio did not differ among fat depots. Ergot-alkaloids were detected in s.c. adipose tissues, and alkaloid concentration was greater (P < 0.05) for E+ than AR542. Warner-Bratzler shear force values did not differ between endophyte types, but it decreased (P < 0.01) across the postmortem aging period. Conversely, sensory panel evaluation detected greater (P < 0.01) chewiness and lower (P < 0.05) juiciness for AR542 than for E+ steaks aged for 14 d. Although grazing cattle on tall fescue pastures infected with nil ergot alkaloid endophyte improved cattle performance, these results suggest that endophyte type has minor effects on carcass traits and meat quality of pasture-fed beef. Moreover, finishing cattle on tall fescue pastures showed the potential to enhance the fatty acid profile of beef from a human health perspective. Alkaloid concentration was greater (P < 0.05) in s.c. fat from E+ than AR542 (2.81 vs. 0.92 ppb; fresh-tissue basis). Measurement of plasma cardiac troponin I concentration ([cTnI]) is a sensitive and specific means for detecting myocardial damage in many mammalian species. Studies have shown that [cTnI] increases rapidly after cardiomyocyte injury. The molecular structure of cTnl is highly conserved across species, and current assays developed for its detection in humans have been validated in many species. In this study, [cTnI] was quantified using a 2-site sandwich assay in plasma of healthy control cats (n = 33) and cats with moderate to severe hypertrophic cardiomyopathy (HCM) (n = 20). [cTnI] was significantly higher in cats with HCM (median, 0.66 ng/mL; range, 0.05-10.93 ng/mL) as compared with normal cats (median, <0.03 ng/mL; range, <0.03-0.16 ng/mL) (P < .0001). An increase in [cTnI] was also highly sensitive (sensitivity = 85%) and specific (specificity = 97%) for differentiating cats with moderate to severe HCM from normal cats. [cTnI] was weakly correlated with diastolic thickness of the left ventricular free wall (r2 = .354; P = .009) but not with the diastolic thickness of the interventricular septum (P = .8467) or the left atrium: aorta ratio (P = .0652). Furthermore, cats with congestive heart failure at the time of cTnI analysis had a significantly higher [cTnI] than did cats that had never had heart failure and those whose heart failure was controlled at the time of analysis (P = .0095 and P = .0201, respectively). These data indicate that cats with HCM have ongoing myocardial damage. Although the origin of this damage is unknown, it most likely explains the replacement fibrosis that is consistently identified in cats with moderate to severe HCM. 相似文献

Equine protozoal myeloencephalitis is a common neurologic disease of horses in the Americas usually caused by Sarcocystis neurona. To date, the disease has not been induced in horses using characterized sporocysts from Didelphis virginiana, the definitive host. S. neurona sporocysts from 15 naturally infected opossums were fed to horses seronegative for antibodies against S. neurona. Eight horses were given 5x10(5) sporocysts daily for 7 days. Horses were examined for abnormal clinical signs, and blood and cerebrospinal fluid were harvested at intervals for 90 days after the first day of challenge and analyzed both qualitatively (western blot) and quantitatively (anti-17kDa) for anti-S. neurona IgG. Four of the challenged horses were given dexamethasone (0.1mg/kg orally once daily) for the duration of the experiment. All challenged horses immunoconverted against S. neurona in blood within 32 days of challenge and in CSF within 61 days. There was a trend (P = 0.057) for horses given dexamethasone to immunoconvert earlier than horses that were not immunosuppressed. Anti-17kDa was detected in the CSF of all challenged horses by day 61. This response was statistically greater at day 32 in horses given dexamethasone. Control horses remained seronegative throughout the period in which all challenged horses converted. One control horse immunoconverted in blood at day 75 and in CSF at day 89. Signs of neurologic disease were mild to equivocal in challenged horses. Horses given dexamethasone had more severe signs of limb weakness than did horses not given dexamethasone; however, we could not determine whether these signs were due to spinal cord disease or to effects of systemic illness. At necropsy, mild-moderate multifocal gliosis and neurophagia were found histologically in the spinal cords of 7/8 challenged horses. No organisms were seen either in routinely processed sections or by immunohistochemistry. Although neurologic disease comparable to naturally occurring equine protozoal myeloencephalitis (EPM) was not produced, we had clear evidence of an immune response to challenge both systemically and in the CNS. Broad immunosuppression with dexamethasone did not increase the severity of histologic changes in the CNS of challenged horses. Context
Conservation corridors must facilitate long-distance dispersal movements to promote gene flow, prevent inbreeding, and allow animals to shift ranges with climate change. Least-cost models are used to identify areas that support long-distance movement. These models rely on estimates of landscape resistance, which are typically derived from habitat suitability.Objectives
We examine two key steps in estimating resistance from habitat suitability: choosing a procedure to estimate habitat suitability, and choosing a transformation function to translate habitat suitability into resistance.Methods
We used linear and nonlinear functions to convert three types of habitat suitability estimates (from expert opinion, resource selection functions, and step selection functions) into resistances for elk (Cervus canadensis) and desert bighorn sheep (Ovis canadensis nelsoni). We evaluated the resulting resistance maps on an independent set of observed long-distance, prospecting movements.Results
A negative exponential function best described the relationship between resistance values and habitat suitability for desert bighorn sheep indicating long-distance movers readily travel through moderately-suitable areas and avoid only the least suitable habitat. For desert bighorn sheep, all three suitability estimates performed better than chance, and resource and step selection functions outperformed expert opinion. For elk, all three suitability estimates performed the same as chance.Conclusions
When designing corridors to facilitate long-distance movements of mobile animals, we recommend transforming habitat suitability into resistance with a negative exponential function. Use of an exponential transformation means that larger fractions of the landscape offer low resistance, allowing greater flexibility in where a corridor is located.57.

In Germany and Europe exotic fruits from distant places are becoming more and more important. In the tropics and subtropics an enormous variety of edible fruits exists. In 2008 the dry fruit pulp of the baobab (Adansonia digitata L.) was approved as novel food for the EU and thus entered the formal food market. Hence, an increasing number of products became available in Europe. The baobab is an impressive tree in the savannas of Africa. Due to its dense and wide spreading root system and its enormous water storing capacity in the stem, the tree can grow under very dry environmental conditions. In many regions of Africa it is a multipurpose indigenous fruit tree species which offers shelter and provides food, fodder and medicine as well as raw material for many purposes. However, even in Europe a wide range of products is now available as shown in the present study, which identified more than 300 baobab products or products with baobab as an ingredient. In Germany, not only the natural baobab pulp but also ready-made foodstuffs such as soft drinks, sandwich spreads, cereal bars, sweets and chocolates are available. Furthermore, a diverse range of pharmaceutical and cosmetic products with extracts from the baobab is available in the market. The products range from after shave and perfume to shampoo and shower gel to leg lotion and foot spray. In Germany baobab saplings and seeds are offered for planting and cultivation as an indoor pot plant. Commercially marketed baobab bonsai seedlings for the European market are produced in Senegal. In Africa the baobab fruits are mainly harvested from wild stands. However, the commercial production of baobab fruits in orchards could be an important future opportunity for income generation to many local households. More research on baobab, including the domestication of this wild fruit tree species, and the conservation of its natural stands as important genetic resource are prerequisites for sustainably increased fruit production. 相似文献

OBJECTIVE: To characterize the bacteria of the genital tract in adult cats; assess the effect of estrus, mating, and administration of progestins on those microorganisms in females; and evaluate whether results of bacteriologic culture of vaginal swabs are affected by cleansing of the vulva prior to sampling or by repeated sampling. ANIMALS: 66 female and 29 male cats undergoing routine ovariohysterectomy or castration. PROCEDURE: Specimens were obtained from vaginal and uterine or preputial mucosae with swabs moistened with sterile saline (0.9% NaCl) solution. In 9 cats, vaginal specimens were obtained before and after cleansing of the vulva with ethanol; in 7 female cats, 2 vaginal speci mens were obtained in immediate succession. RESULTS: Aerobic bacteria were most commonly isolated from cats' vaginas and prepuces; anaerobic bacteria were isolated frequently from males (41%) but rarely from females (5%). Generally, culture results were not affected by cleansing of the vulva or repeated vaginal sampling. The bacterial population of the vaginas of cats was influenced by stage of the estrous cycle but not by mating or administration of progestins. Bacteria were not isolated from the uterus of any cat. CONCLUSIONS AND CLINICAL RELEVANCE: In cats, bacteria of the genital tract in females are predominantly aerobic; in males, aerobic and anaerobic bacteria are found. The bacterial population of the vagina is affected by stage of the estrous cycle. Pure growth of bacteria in culture of genital tract specimens is a normal finding; antimicrobials should only be administered if clinical signs of genital infection are present. 相似文献

Six Hereford steers (295 kg) cannulated in the proximal duodenum were used to evaluate the effects of forage and sunflower oil level on ruminal biohydrogenation (BH) and conjugated linoleic acid (CLA) outflow. Steers were fed one of six treatment diets in a 3 x 2 factorial arrangement of treatments (grass hay level: 12, 24, or 36% of DM; and sunflower oil level: 2 or 4% of DM) in a 6 x 6 Latin square design. The remainder of the diet was made up of steam rolled corn and protein/mineral supplement. Duodenal samples were collected for 4 d following 10-d diet adaptation periods. Data were analyzed with animal, period, forage level, sunflower oil level, and two-way interaction between forage and sunflower oil level in the model. Dry matter intake showed a quadratic response (P < 0.04), with an increase in DMI as forage level increased from 12 to 24% followed by a decrease in DMI when 36% forage was fed. Flow of fatty acids at the duodenum was higher (P < 0.03) for 4 vs. 2% sunflower oil diets, and similar among forage levels. Apparent ruminal digestibility of NDF increased in a linear manner (P < 0.04) as dietary forage level increased. Ruminal BH of dietary unsaturated 18-C fatty acids, oleic acid, and linoleic acid increased linearly (P < 0.05) as dietary forage level increased. Linoleic acid BH tended (P < 0.07) to be greater for 4 than 2% sunflower oil level. Duodenal flow of pentadecyclic, stearic, linolenic, and arachidic acids increased linearly (P < 0.05) as dietary forage level increased from 12 to 36%. Duodenal flow of linoleic acid decreased in a linear manner (P < 0.03) with increasing dietary forage level. Flow of trans-10 octadecenoate decreased linearly (P < 0.03) as dietary forage level increased, whereas trans-11 vaccenic acid flow to the duodenum increased (P < 0.01) linearly with increased dietary forage. Dietary forage or sunflower oil levels did not alter the outflow of cis-9, trans-11 CLA. Flows of cis-11, trans-13, and cis-9, cis-11 CLA increased linearly (P < 0.05) with increased dietary forage. Flows of cis-11, cis-13, and trans-11, trans-13 CLA decreased linearly (P < 0.05) with increased dietary forage. Increasing dietary forage levels from 12 to 36% in beef cattle finishing diets increased BH of unsaturated 18-C fatty acid and outflow of trans-11 vaccenic acid to duodenum without altering cis-9, trans-11 CLA outflow. 相似文献

OBJECTIVES: To estimate the incidence and breed predilection for canine dystocia using data from insurance claims. The risk factors for cesarean section (CS) were assessed for bitches with dystocia. STUDY DESIGN: Retrospective, longitudinal study. SAMPLE POPULATION: Insurance claims records (1995-2002) from a Swedish animal insurance database (Agria), including approximately 200,000 bitches. METHODS: The overall incidence rate of dystocia in insured bitches was calculated by dividing the number of reimbursed dystocia claims with the number of dog years at risk. Subsequently, incidence rates were stratified by breed, region, and habitat. The proportion of bitches with a dystocia claim that had CS were calculated, and risk factors for CS were assessed using a logistic regression model. RESULTS: Between 1995 and 2002, 3894 (2%) of 195,931 Swedish bitches included in the study had a reimbursed insurance claim for dystocia. The overall incidence rate of dystocia was 5.7 cases/ 1000 dog years at risk. Some breeds like the Scottish terrier were at increased risk of dystocia. Among bitches with dystocia, 63.8% were treated by CS. CONCLUSION: Dystocia in the bitch is more common than reported earlier. The risk of developing dystocia varies by breed, and a high percentage (63.8%) of affected bitches undergo CS.
