A wild strain of Agrobacterium rhizogenes was used to regenerate twelve in vitro plant lines from different hairy roots of H. perforatum (St. John's Wort). The production of the main bioactive constituents was observed even though their yields varied in the different plant lines. Two lines were selected for the hyperoside production (4.9-4.6 mg/gdw) while nine were characterized by significant yields of chlorogenic acid (ranged from 0.47 to 1.09 mg/gdw). Furthermore, one out of twelve lines showed a 10-fold higher hypericin content (0.25 mg/gdw) than that reported for the in vitro shoots in the literature. Morphological and phytochemical features were determined in order to select H. perforatum genotypes enriched in valuable bioactive compounds. 相似文献

Interactions between Zn and Cd on the accumulation of these metals in coontail, Ceratophyllum demersum were studied at different metal concentrations. Plants were grown in nutrient solution containing Cd (0.05–0.25 mg l?1) and Zn (0.5–5 mgl?1). High concentrations of Zn caused a significant decrease in Cd accumulation. In general, adding Cd solution decreased Zn accumulation in C. demersum except at the lowest concentration of Zn in which the Zn accumulation was similar to that without Cd. C. demersum could accumulate high concentrations of both Cd and Zn. The influence of humic acid (HA) on Cd and Zn accumulation was also studied. HA had a significant effect on Zn accumulation in plants. 2 mg l?1 of HA reduced Zn accumulation at 1 mg l?1 level (from 2,167 to 803 mg kg?1). Cd uptake by plant tissue, toxicity symptoms and accumulation at 0.25 and 0.5 mg l?1, were reduced (from 515 to 154 mg kg?1 and from 816 to 305 mg kg?1, respectively) by addition of 2 mg l?1 of HA. Cd uptake reached a maximum on day 9 of treatment, while that of Zn was observed on day 15. Long-term accumulation study revealed that HA reduced toxicity and accumulation of heavy metals. 相似文献

An experiment was conducted to determine the effects of extrusion conditions on chemical composition and in vitro hydrolytic and fermentative digestion of barley grits, cornmeal, oat bran, soybean flour, soybean hulls, and wheat bran. Extrusion conditions altered crude protein, fiber, and starch concentrations of ingredients. Organic matter disappearance (OMD) increased for extruded versus unprocessed samples of barley grits, cornmeal, and soybean flour that had been hydrolytically digested. After 8 h of fermentative digestion, OMD decreased as extrusion conditions intensified for barley grits and cornmeal but increased for oat bran, soybean hulls, and wheat bran. Total short-chain fatty acid production decreased as extrusion conditions intensified for barley grits, soybean hulls, and soybean flour. These data suggest that the effects of extrusion conditions on ingredient composition and digestion are influenced by the unique chemical characteristics of individual substrates. 相似文献

Blending, shaking with a mechanical wrist-action shaker or ultrasonication were compared for removing bacteria from a sandy soil and a silt loam soil. Bacteria were counted microscopically after staining with acridine orange. There was no significant difference in the numbers of bacteria released from the sandy soil by the different treatments but ultrasonication extracted more bacteria from the silt loam. Highest recoveries of bacteria were obtained with a combination of Tris buffer as extractant, and 25% Ringer's solution as diluent. Ultrasonication of an irradiated sandy soil containing added Bacillus cereus removed 97% of the bacteria from the sand grains, but lysed some cells. Members of the indigenous soil population were more firmly held by the soil than added bacteria. Optimum conditions for recovering soil bacteria by ultrasonication were either 10.5 μm for 30s or 5 μm for 2 min. With the natural population, the numbers of bacteria and the proportion of metabolizing bacteria increased a little after ultrasonication as shown by labelling cells with 3H-glucose and detecting uptake by autoradiography. However, three isolates of bacteria tested showed varying amounts of injury after ultrasonication, with 5 μm for 2 min causing more damage than 10.5 μm for 30s. Ultrasonication was a more efficient means of extracting the natural bacterial population of soil than shaking but the time and amplitude must be adjusted to avoid injury to cultured bacterial cells. 相似文献

Heritage apple (Malus domestica Borkh. hybrids) and pear (Pyrus communis L. hybrid) trees grow in villages throughout Terceira Island, Azores, Portugal. Some of these pears have different names but similar morphology. The objective of this study was to determine synonymy, homology, and phylogeny of apples and pears from Terceira and to examine potential relationships of the island pears with standard apples and pears of Portuguese or American descent. Nine apple microsatellite markers were used to determine genetic relationships. Distance- and parsimony-based cluster analysis grouped these genotypes into separate apple and pear clades. The Terceira apples were divided into two clades: the maçā and the reineta-reinette. Among the 17 heritage apple genotypes, seven unique accessions were identified and four groups of synonyms, or possibly clones, were detected including: ‘Reineta Agosto’ and ‘Reineta Verde’ from Altares; ‘Reineta Castanha’ and ‘Reineta Verde Miuda’; ‘Maçā Pêra,’ ‘Maçā Calhau’, ‘Pêro Branco’ from Salga and from Terra-Chā and ‘Maçā Marmelo’; and the five genotypes ‘Maçā Sao Joao’, ‘Malápio Rosa’, ‘Maçā Gaspar’, ‘Maçā Branca’ and ‘Maçā Pato’. In addition, two homonyms were detected. ‘Pêro Vermelho’ from Terra Chā was a separate genotype from a tree from Doze Ribeiras of the same name, but Pêro Branco from Terra Chā appears to be a clone that can be distinguished by an additional allele at CH1F07a from a tree with that name from Salga. One pair of apple clones, ‘Reineta Agosto’ and ‘Reineta Verde’ from Altares appear to be derived from an unreduced gamete of ‘Golden Delicious.’ Another apple genotype ‘Maçā Acida’ could be a sibling of the ‘Maçā Pêra’ clonal group. Other tested standard apples from the US genebank were unrelated to Terceira genotypes. Of the seven heritage pears, five unique genotypes and one pair of synonyms were detected. ‘Pêra Papo Pintassilgo’ from Raminho and ‘Pêra Vermelha’ from the nursery of Serviço de Desenvolvimento Agario da Terceira (SDAT) were synonyms. ‘Passans du Portugal’ was related to ‘Pêra Cabaca’ but other standard pears from the US genebank were unrelated to Terceira genotypes. Future studies will include additional apple and pear cultivars from other Islands of the Azores and continental Portugal, and wild Asian species to further explore genetic relationships. 相似文献

The effects of dietary flax oil and antioxidants on ascites and pulmonary hypertension in broilers using a low temperature model 总被引:3,自引:1,他引:3
1. Three experiments were conducted using a low temperature model to induce pulmonary hypertension (PH) and ascites in broiler chickens. Diets containing 25 g or 50 g flax oil/kg food and control diets with an equivalent amount of animal/vegetable (A/V) blend oil, with and without supplemental antioxidants (vitamin C and vitamin E) were used. The amount of PH was assessed by the ratio of right ventricle weight to total ventricle weight (RV/TV ratio). Birds were considered to suffer from pulmonary hypertension syndrome (PHS) if the RV/TV ratio was greater than 0.299. 2. In experiment 1, the test diets contained 50 g oil/kg food and were given during the grower period only. Birds fed on the flax oil diet tended to have a lower incidence of PHS, ascites and lower RV/TV ratios than birds fed on the control diet. However, when the flax oil diet was supplemented with antioxidants, the incidence of ascites, PHS, haematocrit and whole blood and plasma viscosity increased compared with birds fed on the flax oil diet without antioxidants. These effects were not seen in experiment 2, when the test diets containing 30 g oil/kg food (25 g flax oil plus 5 g A/V blend oil/kg food compared to 30 g A/V blend oil/kg food) were given during the grower period. However, in experiment 3, when the test diets containing 30 g oil/kg food were given from day 1 to week 8, birds fed on the control diet supplemented with antioxidants had a higher incidence of PHS than those fed on the control diet alone. 3. In all 3 experiments, there was no significant effect of dietary fat source or supplemental antioxidants on total food intake or food conversion. 4. We conclude that diets containing 50 g flax oil/kg food tend to reduce the incidence of PHS and ascites in broilers using a low temperature model but the results were not statistically significant. Since 1992 the European Union helps member states to reverse the loss of biodiversity in agricultural landscapes by the financial support of agri-environment schemes. Long-term studies investigating the effects of these schemes are an essential prerequisite for the development of an effective policy to restore biodiversity on farmland. In Dutch meadow landscapes almost all agri-environment schemes focus on the restoration of meadow bird populations by postponement of the mowing date. Between 1990 and 2002 we measured long-term changes in meadow bird densities in areas with and without agri-environment schemes in the Netherlands, both before and after the start of the contract. During these years bird territories were surveyed during five field visits between 15 March and 15 June. Densities of black-tailed godwit (Limosa limosa), and redshank (Tringa totanus) were higher in the areas with management agreements, but these differences were already present before the start of the contracts. After the start of the management contracts densities of black-tailed godwit and oystercatcher (Haematopus ostralegus) did not increase, while those of lapwing (Vanellus vanellus) and redshank even declined relative to the control areas. It is concluded that the current agri-environment schemes are not sufficient to restore meadow bird populations in Dutch agricultural landscapes. In addition to the prescribed postponement of the mowing date, it is probably necessary to raise groundwater levels and to reduce fertilization to allow for the development of an open vegetation structure that will increase chick survival to sufficiently high levels. 相似文献
