Apyrases (E.C.3.6.1.5; NTP-NDPases) are distributed in the cytosol, nuclei, cytoskeleton, and on the surface of plant cells. Some may play an important role in signal transduction from exogenous stimuli. We previously found a protein of ca. 55-kDa (CWP-55) in an ATPase-rich fraction from the pea cell wall bound to the elicitor and supprescins (suppressors of defense) from pea pathogen Mycosphaerella pinodes. We cloned the cDNA of CWP-55 that coincided with PsAPY1, one of two NTPase clones in a pea cDNA library. An analysis with a green fluorescent protein fusion protein indicated that PsAPY1 was distributed in the cell wall, nucleus, and cytoplasm. The recombinant PsAPY1 expressed in Escherichia coli had ATP-hydrolyzing activity responsive not only to the elicitor and supprescins from the pea pathogen but also to other elicitors such as a bacterial harpin, a yeast extract, and a synthetic glycopeptide. Biotinylated fungal signal molecules were bound to the recombinant PsAPY1 specifically. Resonant mirror detection confirmed such binding characteristics of PsAPY1. Based on these results, we discuss the role of cell-wall-bound NTPases in recognizing and responding to microorganisms on the cell wall surface.  相似文献

ABSTRACT:   Relationships between otolith and somatic sizes were examined for Japanese anchovy Engraulis japonicus and sardine Sardinops melanostictus larvae collected broadly in the western North Pacific, based on a substantial data set derived from a previous paper. Allometric formulae showed close fits to the relationships between otolith radius and standard length, and the formulae differed between anchovy and sardine larvae. Despite the high correlations, the effect of somatic growth rate on the otolith and somatic size relationship (the 'growth effect') was significantly detected for both anchovy and sardine larvae. Slower growing larvae tended to have larger otoliths than faster growing conspecifics at the same somatic size. This growth effect was more obvious for sardine larvae than for anchovy larvae, probably because of their differential responses of somatic growth to temperature shifts. The growth effect could lead to the possibility of biases in the back-calculation and size estimation processes. As the growth effect is considered to be a general phenomenon and its extent to be species-specific, the relationship between otolith and somatic size and its uncoupling should be scrutinized before application of techniques based on the otolith and somatic size correlation.  相似文献

The effect of lipopolysaccharide (LPS) on humoral and cell-mediated immunity was assessed using LPS-sensitive C3H/HeN mice. A single injection of LPS significantly decreased the anti-sheep red blood cells (SRBC) antibody titers, but not the number of anti-SRBC antibody producing spleen cells. In contrast, double LPS injection did not significantly decrease the anti-SRBC titers and even increased the number of anti-SRBC antibody producing spleen cells. Similarly, single LPS injection significantly suppressed the swelling of the footpad, but double LPS injection caused milder suppression. These results suggest that a difference in the level and timing of exposure to LPS may influence the immune response to infection or vaccination.  相似文献

The present study was conducted to determine the criteria for selecting good quality embryos on Day-2 post-insemination and at the blastocyst stage. Bovine oocytes were matured, fertilized and cultured in vitro. First, Day-2 embryos were classified based on the number of blastomeres into 2-cell, 3- to 4-cell, 5- to 8-cell and >8-cell stage embryos; chromosome samples were then prepared. In the second experiment, the Day-2 embryos classified according to the number of blastomeres were cultured separately for an additional 6 days (Day 8). The resultant Day-8 blastocysts from each group of Day-2 embryos were classified into the following 3 grades based on morphology and diameter: Grade A, hatched and hatching blastocysts; Grade B, expanded blastocysts; and Grade C, unexpanded blastocysts. Chromosome samples were then prepared. The 5- to 8-cell stage Day-2 embryos had the lowest incidence of chromosomal abnormalities (13.5%, P<0.05) and the highest development rate to blastocysts (59.2%, P<0.05). Furthermore, the blastocysts derived from the 5- to 8-cell stage embryos had the largest mean number of cells (102.8+/-42.4, P<0.05), largest number of metaphases per blastocyst (9.5+/-4.8, P<0.05) and lowest incidence of chromosomal abnormalities (24.6%, P<0.05). The Grade A blastocysts had the largest mean number of cells (136.6+/-33.4, P<0.05), a large number of metaphases per blastocyst (11.9+/-5.5, P<0.05) and a low incidence of severe chromosomal abnormalities (17.3%). The results showed that, at Day 2, the 5- to 8-cell stage embryos were of better quality since they had the lowest incidence of chromosomal abnormalities and the highest blastocyst rate and the resultant blastocysts had the largest number of cells and lowest incidence of chromosomal abnormalities. In particular, selection of Grade A blastocysts can improve the development rate to term.  相似文献

A paradigm of fisheries science holds that spawning stock biomass (SSB) is directly proportional to total egg production (TEP) of fish stocks. This “SSB–TEP proportionality” paradigm has been a basic premise underlying the spawner–recruitment models for fisheries management and numerous studies on recruitment mechanisms of fish. Studies on maternal effects on reproductive potential of a stock have progressed during the last few decades, leading to doubt concerning the paradigm. Nonetheless, a direct test of the paradigm at multidecadal scales has been difficult because of data limitations in the stock assessment systems worldwide. Here, we tested the paradigm for marine fish based on a novel combination of two independent 38‐year time series: fishery‐dependent stock assessment data and fishery‐independent egg survey data. Through this approach, we show that the SSB–TEP proportionality is distorted by density dependence in total egg production per spawner individual (TEPPS) or spawner unit weight (TEPPSW) at a multidecadal scale. The TEPPS/TEPPSW exponentially declined with biomass and thus was density‐dependent for Japanese sardine, a small pelagic species exhibiting a high level of population fluctuation, in the western North Pacific. By contrast, the TEPPS/TEPPSW was sardine‐density‐dependent for Japanese anchovy, another small pelagic species exhibiting a moderate level of population fluctuation well‐known for being out of phase with sardine. Our analysis revealed intraspecific (sardine) and interspecific (anchovy) density dependence in TEPPS/TEPPSW, which was previously unaccounted for in spawner–recruitment relationships. Such density‐dependent effects at the time of spawning should be considered in fisheries management and studies on recruitment mechanisms.  相似文献

ABSTRACT:   The conventional acetate peel method was modified to analyze the shell growth pattern of juvenile Manila clam Ruditapes philippinarum as small as 2 mm in shell length (SL). In the outer shell layer along the axis of maximum growth, two types of growth increments were observed: distinct increments and indistinct increments, which, respectively, do and do not continue to the middle shell layer. The distinct increments were found to be formed every two days in intertidal and shallow subtidal zones by field enclosure experiments of juveniles with datum points marked with alizarin complexone. Growth patterns of juveniles (12 mm SL) collected from the Seaside Park of Yokohama in Tokyo Bay were analyzed to confirm the modified method. Mean daily shell growth rate from April to July 2005 ranged 120–142 μm/day, which was reasonable as compared with previous studies. It was impossible to backcalculate the growth to the settlement size (i.e. 0.2 mm SL) because of erosion of the outer shell surface, and the smallest backcalculated minimum shell length was 0.8 mm. Fluctuations in daily growth rate were high, ranging 29–315 μm/day, and did not show a clear two-weekly rhythm.  相似文献

Abstract:   The aim of the present study was for individual variation in behavior to be characterized and related to differences in physiological stress responses in juvenile tiger pufferfish (fugu) Takifugu rubripes . A set of siblings ( n  = 330) from wild parents was subjected to behavior tests, and some were classified into active and inactive groups ( n  = 42 each group). The active animals consisted of those individuals that kept active swimming during 6 min after transfer from one tank to another, and the inactive animals were those that stopped swimming within 6 min. Time-course acute stress responses, which were elicited by anesthesia and subsequent removal of a pectoral fin, were compared between the active and inactive fish. The stress indicators used were free and total plasma cortisol, and plasma glucose. Although no significant difference was observed at each sampling point between groups, the inactive fish required shorter recovery time from acute stress in all three indicators than the active fish. The percentage of free to total plasma cortisol in the pre-stress condition was significantly lower in the inactive group than in the active group. Our findings indicate that fugu has behavioral variation that is associated with stress responsiveness. The inactive fish are expected to be more tolerant of stress and suitable for aquaculture.  相似文献

When an elicitor is applied to plants to induce resistance, one of the first detectable events is the efflux of ions from the treated tissue. Here we are the first to demonstrate that an elicitor from Mycosphaerella pinodes evokes leakage of Na+ and K+ ions from isolated cell walls of pea and cowpea in vitro, as observed for epicotyl tissues. Pharmacological experiments showed that this elicitor-stimulated leakage was sensitive to vanadate and N-(3-methylphenyl)biphenyl-4-sulfonamide (NGXT-191), that inhibit a cell wall-associated ATPase (apyrase). Vanadate or NGXT-191 suppressed elicitor-induced superoxide generation and expression of defense genes in vivo. On the basis of these results, we assume that the leakage of these ions, probably associated with an ATP-dependent process(es) in the cell wall, is likely associated with induced defenses of pea and cowpea.  相似文献

One of the biological functions of bovine lactoferrin (LF) is modulation of the host defense system, including cytokine production and immune response. The aim of the present study was to investigate the effect of oral administration of LF in calves on lipopolysaccharide (LPS)‐induced metabolic and hormonal changes in inflammatory response. Thirty Holstein calves at 4 day of age were given one of three oral doses of LF (0, 1, 3 g/day) for 10 days (?10 day to ?1 day). They were injected i.v. with LPS (50 ng/kg bodyweight) the day (day 0) after the end of LF treatment. Plasma samples were obtained on ?10, 0 day (immediately before LPS injection), and at 2, 6, 12, 24, 48, 72, and 96 h after LPS injection. Plasma tumor necrosis factor‐α concentrations at 2 h after LPS treatment were lower (P < 0.05) in LF 1 g/day‐fed claves compared with LF 0 g/day (control) calves. On day 0 there were no significant group differences in plasma LF concentration. Plasma concentration of haptoglobin in control calves was elevated by LPS injection. In LF groups, plasma haptoglobin concentrations slightly increased after LPS injection, but those levels at 6–24 h were lower (P < 0.05) than in the control group. The LF treatment inhibited (P < 0.05) the reduction of plasma ferrin concentration in calves following LPS challenge. The concentration of plasma aspartate aminotransferase in calves treated with LF was lower (P < 0.05) than in control calves at 24–96 h after LPS treatment. The concentration of plasma insulin‐like growth factor‐1 (IGF‐1) in all groups was decreased by LPS treatment, while in the LF groups the IGF‐1 level was higher (P < 0.05) than in the control group. Plasma adrenocorticotropic hormone and insulin concentrations in LF groups were lower (P < 0.05) than in control calves at 2 h after LPS injection. These data suggest that LF has a substantial anti‐inflammatory effect on the modulation of the host defense system in preruminant calves.  相似文献

Yellowish lesions with downy, gray growth developed on the leaves of carnations (Dianthus caryophyllus) in a greenhouse in Hokkaido Prefecture, Japan, in March 2011. We identified the fungus on diseased plants as Peronospora dianthicola on the basis of the morphologies of the conidia, conidiophores, and oospores. To confirm pathogenicity of the fungus, we inoculated healthy plants with a conidial suspension of the fungus, and the plants developed the same signs and symptoms as the naturally infected plants. We then reidentified P. dianthicola from the diseased lesions. This is the first report of downy mildew of carnation in Japan.  相似文献
