The expression of IFN-γ and IL-4 on T lymphocytes that infiltrate in nasal polyps

AIM:To investigate the expression of Th1-typed cytokine IFN-γ and Th2-typed cytokine IL-4 on T lymphocytes that infiltrate in nasal polyps for searching the pathogenesis of nasal polyps. METHODS:Nasal polyps tissue samples and peripheral blood were obtained from 21 patients. Normal human inferior turbinate mucosa and peripheral blood were obtained as well. Flow cytometry was adopted to detect the expression of IFN-γ and IL-4 of T lymphocytes. RESULTS: Th cytokines were rarely detected in inferior turbinate from normal human. Nasal polyps tissue consisted of abundant T lymphocytes. The expression of IL-4 and IFN-γ increased in peripheral blood from patients compared with normal human (P<0.05). The expression of IL-4 increased but the expression of IFN-γ decreased in nasal polyps compared with that of peripheral blood from the same patient (P＜0.05). CONCLUSION:There were generous of T lymphocytes infiltrating in nasal polyps. There was abnormal immune status in the local nasal mucosa from the patients, and the predomination of Th cytokine secretion changed compared with peripheral blood from the same patients, which resulted in the change of microenvironment of nasal mucosa and possibly close related to the formation of nasal polyps.     

甜菜夜蛾性信息素组分的鉴定及其田间试验

采用气相色谱仪(GC)及气质联用仪(GC-MS)等技术对我国甜菜夜蛾性信息素组分的鉴定结果表明,雌蛾性信息素腺体中含有4种组分,分别为Z9,E12-14:Ac(A)、Z9-14:OH(B)、Z9-14:Ac(C)和Z9,E12-14:OH(D);田间和室内种群各组分的比例(A:B:C:D)分别为47:18:18:17和43:18:23:16,比例及滴度在两种群间未有显著差异;雄蛾田间引诱测定表明,组分A、B显示性信息素活性。几种不同配比的硅橡胶塞诱芯在田间均显示极高的诱蛾活性,以9:1的AB二元诱芯(剂量100μg)最高,其诱蛾量与黑光灯相当,两者呈显著的正相关性,表明该诱芯可替代黑光灯用于甜菜夜蛾的种群测报。利用性诱捕器进行田间种群监测显示,1999年浙江省慈溪市的甜菜夜蛾共发生6代,以第4、5代发生量最高。     

细胞骨架解聚药物对小麦与叶锈菌互作诱发的细胞过敏性反应的影响

 小麦(Triticum aestivum)品种洛夫林10和叶锈菌小种366组成不亲和组合,小麦叶片发生过敏性坏死反应(HR)是小麦抵抗叶锈菌侵染的重要因素。在接种前给小麦叶片分别预注射微管解聚药物磺草硝(oryzalin)和微丝解聚药物细胞松弛素D (cytochalasin D,CD),结果表明2种药物注射使得寄主因叶锈菌侵染诱导的细胞过敏性坏死数目明显减少,并且注射药物的浓度越大,寄主细胞发生HR的数量越少。说明肌动蛋白和微管蛋白的聚合状态是诱发小麦叶片发生HR防卫反应所必需的,细胞骨架在小麦抵抗叶锈菌侵染过程中可能起着重要作用。     

莲子草假隔链格孢的寄主范围及对空心莲子草的控制作用

 空心莲子草(Alteranthera philoxeroides)由于具有繁殖迅速、适应性强等特点,于20世纪50年代被引人我国江苏和浙江等地作为牲畜饲料植物栽培,随后相继传人全国各地并引起危害,成为我国大部分省区旱地和水域的恶性杂草之一。其微生物防治目前仍处于生物防治物的调查筛选阶段,已发现的病原真菌有假隔链格孢菌(Nimbya alternantherae)、炭疽菌(Colletotrichum gloeosporioides)、立枯丝核菌(Rhizoctonia solani)及链格孢(Alternaria sp.)等。     

‘新红星’苹果果实蔗糖合酶的活性及亚细胞定位

 在‘新红星’苹果果实发育过程中, 伴随果糖、葡萄糖和蔗糖的积累, 蔗糖合酶的分解活性逐渐下降, 而合成活性逐渐升高。苹果果实的蔗糖合酶由分子量为90 kD 的亚基组成, 其免疫信号强度随发育进程而增加。蔗糖合酶主要定位于果肉细胞的细胞质中, 发育中后期该酶的分布密度有一定增加。综合结果认为, 蔗糖合酶调控发育早期蔗糖的降解和发育后期蔗糖的积累。     

Epoxyeicosatrienoic acid: the signal transduction

Cytochrome P450 monoxygenase converts arachionic acid to four epoxyeicosatrienoic acid regiosomes: 5, 6-EET (epoxyeicosatrienoic acid); 8, 9-EET; 11, 12-EET and 14, 15-EET. Recent studies show that EETs are involved in signal transduction. EETs open Ca²⁺-sensit ive K⁺ channel and inhibit Na⁺ channel, Ca²⁺-sensitive Cl^- channel and so on. What is more, EETs have been demonstrated to activate PP60^c-src and initiate a tyrosine kinase cascade that mediates mitogenic effects.     

Inhibitory effect of berberine on the activation and proliferation of T lymphocytes

AIM: To investigate the effect of berberine (Ber) on the activation and proliferation of T lymphocytes and its mechanism of action. METHODS: Whole peripheral blood from normal subjects was stimulated with phytohemagglutinin (PHA) or phorbol ester (PDB) plus ionomycin (Ion) and the expression levels of CD69 and CD25 were evaluated with flow cytometry after the staining with appropriate fluorescent monoclonal antibody. The distribution of cell cycles was analyzed by propidium iodide staining and dead cells by 7-aminoactinomycin live staining. RESULTS: 100 μmol/L and 50 μmol/L of Ber had significant inhibition of the expression of CD69 on T cells stimulated with PDB plus Ion or PHA, while effect of 25 μmol/L Ber was not significant. And as time of action extended, the extent of inhibition decreased. For the expression of CD25, Ber at the concentrations as above all exerted significant inhibitory effect in a dose-dependent manner. Moreover, Ber could block lymphocytes cell cycle progression from G₀/G₁ phase to S and G₂/M phase without phase specificity. Besides, live staining analysis revealed that Ber did not have significant cytotoxicity on lymphocytes. CONCLUSIONS: Ber significantly inhibits the expression of early and mid activation antigens of T cells and also blocks the progression of lymphocytes cell cycles. These results suggest that Ber exerts immunosuppression effect through inhibiting the activation and proliferation of T cells.     

Study on the electrophysiological characteristics of ion channels of stem cell derived cardiomyocytes of mouse

AIM: To study the electrophysiological characteristics of ion channels of stem cell derived cardiomyocytes(SCDC) of mouse. METHODS: Embryonic stem cells of D₃ line(ES-D₃) were cultured on the MEF feeder layer with BRL conditioned medium, and fetal mouse heart cells(FMHC)were cultured in vitro. Then ES-D₃ cells were induced to differentiate into many kinds of cells. SCDC were harvested on day 12 after differentiation initiating and identified by electro-microscope and immunocytochemistry. SCDC and FMHC were prepared for the patch-clamp research. Sodium and calcium currents together were elicited and compared between SCDC and FMHC. RESULTS: The current characteristics of sodium and calcium channels of SCDC were very similar to FMHC. CONCLUSION: The functional expression of ion channels occurred during ES-D₃ cells differentiation and the electrophysiological characteristics of sodium and calcium channels of SCDC are very similar to FMHC.     

Protein expression of cyclin D2 and p16 in proliferation and differentiation of cultured cardiac myocytes

AIM:To investigate the protein expression of cyclin D₂ and p16 in proliferation and differentiation of cultured cardiac myocytes.METHODS:One-day-old Sparague-Dawley rats were used. Cardiac myocytes(CM) were collected by a trypsin-dispersal method and cultured. Cell growth line and fluorescence activated cell sorting (FACS) were used to investigate the proliferation of CM. Ultra-thin sections were made to observe the ultrastructure of CM under transmission electron microscope. The expression of cyclin D₂ and p16 in CM were measured using immunocytochemistry and image analysis.RESULTS:①Results of cell growth line and FACS analysis showed that cultured CM could proliferate in the first 3 cultured days, but the ability decreased quickly, concomitant with differentiation. CM was obseved quiescent in cell cycle three days later. The ultrastructure of CM showed the large amount of myofilaments and mitochondrion. ②The protein expression of cyclin D₂ in 3,4,5 day CM group was 0.89 times(P＜0.05),0.80 times (P＜0.05) and 0.56 times (P＜0.01) of that in 1 day group, respectively. The expression of p16 in CM was increased during the culture process, 2,3,4,5 day group were 1.63 times, 1.72 times, 1.99 times and 2.84 times (P＜0.01) of that in 1 day group, respectively.CONCLUSION:Cultured neonatal rat cardiac myocytes could proliferate during the first 3 days after incubation, but the ability of proliferation decreased, from the fourth day, concomitant with differentiation. Cyclin D₂ and p16 play the key roles in CM postnatal development. Downregulation of cyclin D₂ and upregulation of p16 may induce CM differentiation.     

达县分葱田甜菜夜蛾发生规律初步研究

采用系统调查方法研究了达县地区分葱田甜菜夜蛾的生活史、发生与环境的关系。甜菜夜蛾在达县分葱田1a发生6代,以三至五代为害最重,世代重叠。其数量消长与虫源、夏秋季气候、天敌、栽培管理等因素密切相关。通过调查分析提出本县初始虫源非本地虫源。
