Pruritic skin diseases are common in cats and demand rigorous diagnostic workup for finding an underlying etiology. Measurement of a serum allergen-specific IgE in a pruritic cat is often used to make or confirm the diagnosis of a skin hypersensitivity disease, although current evidence suggests that elevated allergen-specific IgE do not always correlate with a clinical disease and vice versa. The aim of the study was to to assess the possible influence of age, deworming status, lifestyle, flea treatment, and gender on allergen-specific IgE levels and to evaluate the reliability of IgE testing in predicting the final diagnosis of a pruritic cat. For this purpose sera of 179 cats with pruritus of different causes and 20 healthy cats were evaluated for allergen-specific IgE against environmental, food and flea allergens using the Fc-epsilon receptor based enzyme-linked immunosorbent assay (ELISA) test. The results of the study showed positive correlation between age, outdoor life style, absence of deworming, absence of flea control measures and levels of allergen-specific IgE. Gender and living area (urban versus rural) did not seem to affect the formation of allergen-specific IgE. According to these findings, evaluating allergen-specific IgE levels, is not a reliable test to diagnose hypersensitivity to food or environmental allergens in cats. On the contrary, this test can be successfully used for diagnosing feline flea bite hypersensitivity.  相似文献

The objectives of this study were to obtain relevant blood flow indices of umbilical arteries (UmA) of porcine fetuses using a laparoscopic ultrasound probe and to relate these data with fetal size at early to mid gestation. Fetal parameters and flow indices, i.e., fetal length and area, fetal heart rate (FHR), systolic pulse duration (T1), interpulse duration (T2), T2/T1 ratio, peak systolic velocity (PSV), time averaged velocity (TAV), resistance index (RI) and pulsatility index (PI), were measured in 182 fetuses of 26 pregnant Landrace gilts on pregnancy day (PD) 36 (122 fetuses from 17 gilts), PD42 (19 fetuses from 3 gilts) and PD51 (42 fetuses from 6 gilts). Fetal heart rate was higher on PD36 than on PD42 (P<0.05). No differences (P>0.05) were obtained concerning systolic pulse duration, flow velocities and RI. On PD42, the PI was lower (P<0.05), while the interpulse duration (P=0.06) and T2/T1 ratio tended (P=0.08) to be higher on PD42 compared with PD36 and to PD51. To find differences in UmA blood flow parameters concerning fetal size, i.e., fetal length, fetuses were retrospectively grouped as follows: small (lower 25%), medium (mean 50%) and large (upper 25%), respectively. Although, fetuses differed in size (P<0.001) within and between days of pregnancy, FHR, PSV, TAV, RI and PI did not differ (P>0.05) among the size classes. Only systolic pulse duration tended to be longer (P=0.05) in large compared with small fetuses on PD36, and interpulse duration was lower in large fetuses on PD36 in comparison with PD51 (P<0.05). Though there was no link between fetal blood flow indices and fetal intrauterine growth retardation (IUGR), with further studies based on these flow indices, it might be possible to evaluate nutrient- or stress-related influences on fetal growth and development, particularly in the case of IUGR.  相似文献

The objective of this study was to assess the reproductive performance of two Nguni ecotypes (Nguni and Landim) raised in a subtropical environment to enhance strategies for livestock development and restocking programmes within the southern African region. Reproduction data collected between 1996 and 2009 from 365 cows of the Landim and Nguni ecotypes were analysed. From the results, ecotype, place of birth, year and season of birth/calving had significant effects on age at first calving (AFC) and calving interval (CI). Overall means for AFC and CI were 1,071 ± 166 days and 432 ± 85 days, respectively, while average calving rate was 88.0 ± 4.7%. Heifers born in the dry season had lower AFC than heifers born in the wet season. Heifers born at Impaputo Breeding Center were the youngest at first calving, followed by the South African born ones. Heifers of the Landim ecotype also calved younger than heifers of the Nguni ecotype. CI was shorter in wet seasons (main breeding seasons) than in dry seasons. Interaction between ecotype and year–season (p < 0.005) showed that, in wet and dry seasons, Nguni cows had shorter CI than the Landim. This study demonstrates for the first time a possible genotype-by-environment interaction between Nguni ecotypes. This might aid future cattle development and restocking programmes in southern Africa taking into consideration the adaptation of indigenous genotypes and climate change.  相似文献

Background

The genus Populus is accepted as a model system for molecular tree biology. To investigate gene functions in Populus spp. trees, generating stable transgenic lines is the common technique for functional genetic studies. However, a limited number of genes have been targeted due to the lengthy transgenic process. Transient transformation assays complementing stable transformation have significant advantages for rapid in vivo assessment of gene function. The aim of this study is to develop a simple and efficient transient transformation for hybrid aspen and to provide its potential applications for functional genomic approaches.

Results

We developed an in planta transient transformation assay for young hybrid aspen cuttings using Agrobacterium-mediated vacuum infiltration. The transformation conditions such as the infiltration medium, the presence of a surfactant, the phase of bacterial growth and bacterial density were optimized to achieve a higher transformation efficiency in young aspen leaves. The Agrobacterium infiltration assay successfully transformed various cell types in leaf tissues. Intracellular localization of four aspen genes was confirmed in homologous Populus spp. using fusion constructs with the green fluorescent protein. Protein-protein interaction was detected in transiently co-transformed cells with bimolecular fluorescence complementation technique. In vivo promoter activity was monitored over a few days in aspen cuttings that were transformed with luciferase reporter gene driven by a circadian clock promoter.

Conclusions

The Agrobacterium infiltration assay developed here is a simple and enhanced throughput method that requires minimum handling and short transgenic process. This method will facilitate functional analyses of Populus genes in a homologous plant system.  相似文献

Multifunctional forestry in Germany is characterized by long production periods and highly complex biological–technical processes. Therefore forest enterprises are complex systems, which are closely intertwined with their economic environment. To ensure their economic success, forest enterprises and forest policies need to take the economic development into consideration. The cost accounting scheme of the German Forestry Council provides a basis for management accounting in multifunctional forest enterprises and distinguishes five different production areas whereby intercompany comparisons of different kinds of ownership in forest accountancy networks (FANs) are possible. Results from such a FAN are used to illustrate the economic situation and long-term development of a panel of private forest enterprises. As well these data can support decision-making processes of individual enterprises, forest economic analyses and consultation in forest policy analyses.  相似文献
