Among grow-to-finish pigs from 10 herds in Alberta and Saskatchewan, 23 (16%) of 144 fecal samples were culture-positive and 40 (28%) of 144 pigs were seropositive for Salmonella. With a Bayesian model specifying dependence between the 2 tests, the sensitivity (Se) of culture and real-time polymerase chain reaction (RT-PCR) was 79% to 86%, depending on the cut-off value for the enzyme-linked immunosorbent assay (ELISA). Culture specificity (Sp) was assumed to be 100%; RT-PCR Sp was found to be 94%. The ELISA Se was 76% and 51% at optical density cut-off values ≥ 20% and ≥ 40%, respectively; the Sp was 94% at each cut-off value. The model showed some sensitivity to ELISA prior information, the ELISA Se being approximately 8% lower when informative prior information was specified in the model. When there was no adjustment for dependence between culture and RT-PCR, the posterior estimates for both culture and RT-PCR Se were 11% higher than with the conditional-dependence model and had considerably narrower probability intervals, which suggests that correlation between culture and PCR is important and should be adjusted for in future studies. 相似文献

Theileria parva antigens recognized by cytotoxic T lymphocytes (CTLs) are prime vaccine candidates against East Coast fever in cattle. A strategy for enhancing induction of parasite-specific T cell responses by increasing recruitment and activation of dendritic cells (DCs) at the immunization site by administration of bovine Flt3L and GM-CSF prior to inoculation with DNA vaccine constructs and MVA boost was evaluated. Analysis of immune responses showed induction of significant T. parva-specific proliferation, and IFN-γ-secreting CD4(+) and CD8(+) T cell responses in immunized cattle. However, antigen-specific CTLs were not detected. Following lethal challenge, 5/12 immunized cattle survived by day 21, whereas all the negative controls had to be euthanized due to severe disease, indicating a protective effect of the vaccine (p<0.05). The study demonstrated the potential of this technology to elicit significant MHC class II and class I restricted IFN-γ-secreting CD4(+) and CD8(+) T cells to defined vaccine candidate antigens in a natural host, but also underscores the need to improve strategies for eliciting protective CTL responses. 相似文献

Different degrees of dependency on the activity of arbuscular mycorrhizal fungi (AMF) exist between native maize landraces and hybrids. In Los Tuxtlas, Mexico, the Popoluca people maintain a traditional polycultural land management with more than 15 native landraces of maize; however, it is not known whether the recent substitution of local maize for improved hybrids and fertilization has affected the integrity of the mycorrhizal symbiosis in these naturally phosphorus-poor systems. A greenhouse experiment was conducted to evaluate the response of four Popoluca maize landraces and the hybrid Texcoco to the presence of native AMF in conditions of low and medium P input (5 and 65 mg kg?1, respectively). After 120 days in both P treatments, the native landraces Black and Yellow presented higher colonization and had acquired more P in their shoot biomass than the hybrid. The moderate fertilization did not appear to have affected the integrity of the mycorrhizal symbiosis, since all of the maize types presented a positive mycorrhizal dependency (2–14 %). Under low P conditions, the Texcoco hybrid maize presented one of the highest mycorrhizal dependencies; however, unlike the local landraces, this was not reflected in a higher tissue P concentration. The results obtained indicate that the native maize Black was the best at capturing symbiotic and direct P, which makes this landrace an important genetic and cultural heritage for the Popoluca and for the world. 相似文献

Small-scale toxicity testing with microbiotests is a rapidly-expanding component of the field of aquatic toxicology which contributes diverse contamination assessment tools and approaches for a variety of environmental (liquid and solid) media. In this short review on microbiotesting, some of the recent developments conducted under the second St. Lawrence River Action Plan (1993–1998) at the St. Lawrence Centre (Environment Canada, Quebec Region, Montreal) are recalled. These include 1) employing the SOS Chromotest to determine the genotoxic status of major industrial effluents discharging to the St. Lawrence River and their potential impact on downstream biota, 2) developing an algal solid phase assay to predict the toxic potential of freshwater sediments, 3) developing a microplate-based cnidarian assay to screen for toxicity of chemicals and environmental samples, 4) developing an alternative assay to whole fish acute (sub)lethal toxicity testing with the help of rainbow trout primary hepatocytes, 5) developing a microplate-based phagocytosis assay to check for immunocompetence of feral bivalve shellfish and 6) conducting a major investigation to develop a cost-effective multitrophic bioanalytical battery to assess the (geno)toxicity of freshwater sediments. In addition, integrative tools with specific microbiotests were respectively constructed to determine the toxic potential of industrial effluents (PEEP: Potential Ecotoxic Effects Probe) and that of sediments (SED-TOX). Such examples illustrate the diversity of on-going endeavors in the field of small-scale toxicity testing internationally, as further corroborated by recent books entirely dedicated to the subject. Wild sablefish, Anoplopoma fimbria, are a valuable commercial species whose populations are declining. Fortunately, sablefish are excellent species for commercial aquaculture. Sablefish raised under high‐density conditions in the marine environment require the use of efficacious vaccines to control disease. Sablefish impacted by disease in net pens may have poor flesh quality and high mortality during grow‐out. As a result, disease can cause financial hardship for sablefish aquaculture operators. The efficacy of a multivalent vaccine preparation for sablefish, administered either by intraperitoneal (i.p.) injection or by immersion, against atypical and typical Aeromonas salmonicida, the causative agents of atypical and typical furunculosis, respectively, was examined. A. salmonicida can affect sablefish at any age and size. Consequently, an efficacious vaccine that can be appropriately and optimally administered to all life stages is desirable. Sablefish vaccinated by immersion at ~1.5 or ~4.5 g with a whole‐cell multivalent vaccine were not protected against either typical or atypical A. salmonicida. Factors that may have contributed to the ineffectiveness of the immersion vaccine are discussed. By contrast, the relative per cent survival (RPS) or potency of the whole‐cell multivalent vaccine injected i.p. in juvenile sablefish at ~50 g against typical and atypical A. salmonicida was 94.3% and 81.7% respectively. The success of various grass-legume mixtures in controlling competing vegetation, and their effect on subsequent survival and growth of Sitka spruce seedlings was studied on a coastal alluvial site in northwestern British Columbia. Mechanically scarified (bladed) strips were hand seeded to pure and combined mixtures of legumes, bunchgrasses, and sodforming grasses. An unseeded control (bladed but not seeded) was also established.Alsike clover (Trifolium hybridum L.) was the most successful legume species. Orchardgrass (Dactylis glomerata L.), big bluegrass (Poa ampla Merr.), and creeping red fescue (Festuca rubra L.) were the most successful grass species. Red alder (Alnus rubra Bong.) density and height were lower in legume-seeded treatments. Red elderberry (Sambucus racemosa L.) and salmonberry (Rubus spectabilis Pursh.) were effectively reduced by blading and have reestablished slowly. A combination of blading and early establishment of either sodforming grasses or bunchgrasses effectively decreased reinvasion by thimbleberry (Rubus parviflorus Nutt.).Growth of Sitka spruce was best in the unseeded control treatment or in the treatments with legumes but no sodforming grasses. Presence of sodforming grasses decreased both diameter and height growth. Sitka spruce diameter decreased with increasing red alder density. After 5 growing seasons, spruce has outgrown all competitors except red alder. 相似文献
