Background: In routine canine medicine, anticoagulated blood is often the only sample sent to laboratories for diagnostic purposes. This hampers the interpretation of protein electrophoretic tracings because plasma contains fibrinogen, which migrates in the β–γ region. In human medicine, fibrinogen can be precipitated from plasma using ethanol. Objectives: The purpose of this study was to assess ethanol precipitation as a method for removing fibrinogen from canine plasma so as to facilitate the interpretation of electorphoresis results. Methods: Blood samples collected from 40 dogs were divided into plain tubes and tubes containing EDTA (n=20) or lithium–heparin (n=20). An aliquot of plasma from each sample was incubated with ethanol at a final concentration of 100 mL/L. Cellulose acetate electrophoresis was then performed on serum, plasma, and plasma treated with ethanol. To verify the efficiency of ethanol treatment, fibrinogen was added to 5 canine serum samples at final concentrations of 2.5, 5.0, and 10.0 g/L, and electrophoresis was performed before and after ethanol treatment. Results: Visual analysis of electrophoretograms from ethanol‐treated samples confirmed the disappearance of the fibrinogen peak from the β2‐globulin region. Treatment with ethanol caused a significant decrease in the percentage of β2‐globulins and a significant increase in the percentage of α2‐globulins. Absolute values of most electrophoretic fractions were significantly decreased in ethanol‐treated plasma compared with serum. Conclusions: Ethanol treatment successfully removed fibrinogen from canine plasma and normalized electrophoretic profiles, but probably also precipitated proteins other than fibrinogen. Ethanol treatment is recommended to facilitate visual identification of abnormal monoclonal peaks, but not for determining absolute protein concentrations in electrophoretic fractions. 相似文献

To further understand the impact of urban development on wildlife populations, we examined habitat use and selection by female bighorn sheep Ovis canadensis in two endangered subpopulations near a metropolitan area in southern California. One subpopulation, which had previously been found to have low reproductive success, selected urban environments while the other did not use urban areas. In the subpopulation that used urban areas, females had smaller core activity areas and selected lower elevations and gentler slopes. These females used urban sources of water but a clear relationship between levels of urban use and periods of increased water need was not evident. Diet quality was higher among females that selected urban areas, and this increase was correlated with the level of urban use. Thus, optimal foraging behavior may have contributed to the selection of urban areas. Urban use was lowest during peak months of parturition, suggesting that reproductive strategies may also have influenced temporal patterns of urban use. Although urban areas provided bighorn sheep with forage and water resources, the use of these areas may have substantial costs. For example, females using urban areas had internal parasites that were not found elsewhere in the Peninsular Ranges. 相似文献

Six strains of Fusarium verticillioides, two of F. oxysporum, one strain of F. proliferatum, and a strain of an unidentified species were cultured on maize patties and rice and evaluated for their ability to simultaneously produce fumonisin B (FB) and C (FC) series analogues. Fumonisins were quantified by LC-MS-MS using positive ion electrospray ionization. FC1 provided characteristic fragment ions at m/z 690, 672, 654, 532, 514, and 338 corresponding to sequential loss of H2O and tricarboxylic acid moieties from the alkyl backbone, while FC3 and FC4 provided equivalent product ions 16 and 32 amu lower than the corresponding FC1 fragments, respectively. All isolates cultured on maize produced FC4. All isolates except for that of F. proliferatum also produced FC1, and three of the six strains of F. verticillioides produced FC3. All isolates except those of F. oxysporum produced detectable amounts of FB1, FB2, and FB3. Isolates that produced fumonisin B analogues produced at least 10 fold more of the B series analogues than they did of the C series analogues. The results confirm that at least some strains of F. oxysporum produce FC, but not FB, fumonisin analogues and also suggest that the genetics and physiological regulation of fumonisin production may be more complicated than previously envisaged since some strains of F. verticillioides and F. proliferatum as well as the strain of the unidentified species can simultaneously produce both FB and FC analogues. 相似文献

In studies examining patterns of mutational damage, the primary variables of interest are expressed typically as discrete counts within defined categories of damage. The multinomial distribution is a common model for such data settings. Of interest is statistical comparison of the pattern, or spectrum, of mutation among the various mutant categories. A specific question in such instances is the identification of differences among the spectral categories, identified, e.g., via simultaneous confidence intervals for pairwise differences in or ratios of the multinomial response probabilities. Herein, a selection of possible methods for constructing such intervals is described and compared via computer simulations to determine which are most appropriate for practical use in the evaluation of mutant spectra. 相似文献

Landspraying while drilling (LWD) is an approved disposal method for water-based drilling mud (WBM) systems in western Canada. The mud is applied either on cultivated land, where it is incorporated by cultivation, or on vegetated land where it is not incorporated. This study examined the effects of summer WBM application (0, 15, 20, 40, and 80 m3 · ha?1) on native vegetation properties. Our results indicated that LWD increased bare ground but decreased lichen cover at the 80 m3 · ha?1 rate relative to the untreated control. Nitrogen (N), sulfur (S), and magnesium (Mg) concentrations in aboveground plant tissue increased with increasing LWD rate in samples taken 45 d after WBM application, but these differences disappeared 1 yr after treatment. Increase in tissue concentration of phosphorus (P) with LWD rate, however, was only detected 3 yr after LWD. Nonetheless, these changes in tissue chemistry were not associated with significant changes in biomass yield or species composition. Overall, our results suggest that single WBM applications at rates (≤ 20 m3 · ha?1) commonly used in western Canada, if properly managed, are unlikely to adversely affect native prairie vegetation. 相似文献

The relationship between the soil parameters measured during soil testing using the bevameter system and horizontal forces acting on a simple tillage tool were investigated. Field experiments were conducted on untilled, compacted and recently tilled soil. On both soils, five sites were randomly chosen where bevameter and draft measurements were performed. The parameters measured were modulus of soil deformation, wet and dry bulk density, soil moisture content, tool operating depth, tool operating speed and horizontal draft.

Dietary supplements that promote healthy aging are mostly warranted in an aging society. Because of age-related risks, anti-inflammatory and anti-oxidative agents such as microalgae are potential candidates for intervention. In a randomized controlled trial, we tested Phaeodactylum tricornutum (PT), a microalgae rich in eicosapentaenoic acid (EPA), carotenoids, vitamins, and β-glucans, cultured in bioreactors. In this pilot trial, 19 healthy elderly received supplements for two weeks based on either the whole PT (A), the β-1,3-glucan-rich PT supernatant (SupB), the combination thereof (A+SupB), or a Comparator product (Comp). The primary outcome variable plasma interleukin-6 was reduced after treatment with A+SupB compared to the Comp group (p = 0.04). The mobility parameters 5 s sit-to-stand test (p = 0.04 in the A group) and by trend gait speed (p = 0.08 in the A+SupB diet) were improved compared to Comp. No treatment effects were observed for fatty acids, compared to Comp but omega-6 to -3 fatty acid ratio (p = 0.006) and arachidonic acid/EPA ratio (p = 0.006) were reduced within group A+SupB. Further, the SupB study product reduced faecal zonulin (p = 0.03) compared to the Comp. The data revealed an anti-inflammatory and potentially anti-oxidative effect of particular PT preparations, suggesting that they might be suitable for effects in healthy elderly. 相似文献

A retrospective analysis of 619 upper and lower cheek teeth from 62 horses was performed. Based on clinical findings, as well as radiographic and magnetic resonance (MR) imaging findings, the teeth were classified into five groups. There were 20 teeth with abnormal MR imaging signals as well as clinical alterations and 599 healthy teeth. Using MR imaging, the appearance of pulp in diseased and disease‐free teeth was compared, and the appearance of abnormal pulp was studied. Subsequently, the ability of MR imaging to diagnose pulpitis and pulp necrosis in teeth with normal external appearance was investigated. In horses with clinically verified dental disease, abnormal MR imaging findings were confirmed in the pulp of all affected teeth. An enlarged blurred pulp image with a lower signal intensity was observed only in clinically diseased teeth and was a reliable criterion for diagnosing dental disease on MR imaging. On the other hand, partial or complete absence of pulp in all MR imaging sequences was observed in both diseased and nondiseased teeth. The leaching of aldicarb and thiofanox in soils (sandy loam, silt loam and sandy clay loam), and their uptake by sugarbeet plants were studied. Three irrigation levels were maintained: half, normal and double dose. The residues were determined as the sum of the insecticidal metabolites (parent compound + sulphoxide+ sulphone) for both pesticides. Leaching was greatly influenced by the amount of water added and the soil type. Under normal conditions, leaching seemed to proceed very slowly, keeping the chemicals available for uptake by the root systems for a long time. The concentration of insecticide in the leaves was highest in beets grown on sandy loam and lowest in those grown on sandy clay loam. The quantity of irrigation did not influence the residue concentration in the leaves greatly, although its influence was obvious on the total residue present (μg per plant). Increasing the water dose always resulted in a higher total residue, and a greater plant weight. The breakdown in the soils was directly related to the water dose. The experiments show that thiofanox was more stable than aldicarb and was taken up by sugarbeet to a greater extent. 相似文献
