Amplification of the 16S rRNA gene from a blood sample obtained from a dog in southeastern Brazil was used to confirm a naturally acquired Ehrlichia (E.) canis infection. Following isolation and culturing of the new bacterial strain called Uberlândia, partial sequences of the dsb and p28 genes were obtained. The dsb partial sequence of the novel strain was 100% similar to dsb gene sequences of E. canis obtained from different geographic areas around the world. Conversely, the p28 partial sequence for the E. canis Uberlândia strain differed at several nucleotides from other sequences available in GenBank. To confirm the antigenic profile of the Uberlândia strain, an indirect immunofluorescence assay against E. canis antigens was performed using dog sera collected from two different areas in Brazil (Uberlândia and São Paulo). The results suggest that both antigens were able to identify animals seropositive for E. canis in Brazil since these Brazilian strains appear to be highly conserved. 相似文献

Air pollution can be a problem in industrial processes, but monitoring and controling the aerosols in the work place is not enough to estimate the occupational risk due to dust particle inhalation. The solubility in lung fluid is considered to estimate this risk. The aim of this study is to determine in vitro specific dissolution parameters for thorium (Th), uranium (U), and tantalum (Ta) associated to crystal lattice of a niobium mineral (pyrochlore). Th, U, and Ta dissolution factors in vitro were obtained using the Gamble solution (simulant lung fluid, SLF), particle induced X-ray emission, and alpha spectrometry as analytical techniques. Ta, Th, and U are present in the pyrochlore crystal lattice as oxide; however, they have shown different dissolution parameters. The rapid dissolution fraction (f r), rapid dissolution rate (λ r), slow dissolution rate (f s), and slow dissolution fraction (λ s) measured for tantalum oxide were equal to 0.1 and 0.45 and 0.00007 day?1, respectively. For uranium oxide, f r was equal to 0.05, λ r was equal to 1.1 day?1, and λ s was equal to 0.000068 day?1. For thorium oxide, f r was 0.025, λ r was 1.5 day?1, and λ s was 0.000065 day?1. These results show that chemical behavior of these three compounds in the SLF could not be represented by the same parameter. The ratio of uranium concentration in urine and feces samples from workers exposed to pyrochlore dust particle was determined. These values agree with the theoretical values of estimated uranium concentration using specific parameters for uranium oxide present in pyrochlore. 相似文献

Natural compounds have shown good potential for the discovery of new chemotherapeutics for the treatment of Chagas’ disease. Recently, our group reported the effective trypanocidal activity of (−)-elatol, extracted from the red macroalgae Laurencia dendroidea present in the Brazilian coast against Trypanosoma cruzi. However, the mechanism of action of this compound has remained unclear. There are only hypotheses concerning its action on mitochondrial function. Here, we further investigated the mechanisms of action of (−)-elatol on trypomastigotes of T. cruzi. For this, we evaluated some biochemical alterations in trypomastigotes treated with (−)-elatol. Our results show that (−)-elatol induced depolarization of the mitochondrial membrane, an increase in the formation of mitochondrial superoxide anion and loss of cell membrane and DNA integrity. Additionally, (−)-elatol induced formation of autophagic vacuoles and a decrease in cell volume. All together, these results suggest that the trypanocidal action of (−)-elatol involves multiple events and mitochondria might be the initial target organelle. Our hypothesis is that the mitochondrial dysfunction leads to an increase of ROS production through the electron transport chain, which affects cell membrane and DNA integrity leading to different types of parasite death. 相似文献

Different monosulfates of quercetin and epicatechin with metabolic interest were obtained by hemisynthesis and characterized regarding their chromatographic behavior and absorption and mass spectra. Three of these compounds were further isolated, and their structures were elucidated by mass spectrometry and (1)H and (13)C nuclear magnetic resonance using one- and two-dimensional techniques (heteronuclear single-quantum coherence and heteronuclear multiple-bond correlation). The calculation of the proton and carbon shifts caused by sulfation allowed for the assignment of the position of the sulfate group in the flavonoids, so that the compounds were identified as quercetin-3'-O-sulfate, quercetin 4'-O-sulfate, and epicatechin 4'-O-sulfate. It was found that sulfation at position 3' induced a large upfield shift in the carbon bearing the sulfate group and downfield displacements of the adjacent carbons, whereas no significant upfield or downfield shifts were observed with respect to the parent flavonoid when sulfation was produced at position 4'. 相似文献

A shortage of marine raw ingredients, such as fish oil, is predicted in the near future. The use of suitable alternative lipid sources, such as vegetable oils, is crucial for sustainable growth of the aquaculture sector. Three isonitrogenous (50% Crude Protein) and isoenergetic (19 kJ g−1) diets, in which fish oil was gradually replaced by soybean oil (0, 25 and 50% of the oil fraction) were tested for 12 weeks, using European sea bass (Dicentrarchus labrax) and rainbow trout (Oncorhynchus mykiss) juveniles. No dietary effects (p>0.05) on growth performance, feed conversion, digestibility or body composition were observed with either species. The results obtained suggest the inclusion of soybean oil up to 50% of the dietary lipid is possible, in diets for sea bass and/or rainbow trout juveniles. 相似文献

Winter hardiness is a major-limiting factor for St. Augustinegrass [Stenotaphrum secundatum (Walt.) Kuntze] grown in the transitional climatic region of the United States. Lab-based freeze tests that mimic the range of field winter survivability in St. Augustinegrass can contribute to the selection of cold hardy genotypes. This study used a whole container method, four freezing temperatures, and two data collection systems to evaluate the freezing response of nine St. Augustinegrass genotypes ranging in their winter hardiness. Results indicated ?3 and ?4 °C with average regrowth ratings of 33.6 and 17.8% respectively, were more suitable temperatures for evaluating freeze survival in St. Augustinegrass than ?5 and ?6 °C with average regrowth ratings of 0.4 and 0%, respectively. Visual ratings of surviving green tissue and regrowth were generally well correlated when evaluated over a six week period post-freeze with Pearson correlation coefficients ranging of 0.17–0.62 for ?3 °C freeze tests and 0.79–0.93 for ?4 °C freeze tests. Additionally, measurement of percent green cover using digital imaging techniques commonly utilized in turfgrass field studies were significantly correlated (0.66) with visual ratings averaged across weekly post-freeze evaluation measurements for both ?3 and ?4 °C freezing temperatures. These results provide evidence that digital imaging analyses are useful in estimating surviving green tissue and regrowth in lab-based freeze tests. This study provides additional information regarding freezing temperatures, genotype responses, and data collection methods in St. Augustinegrass, which should aid breeders in the improvement of freeze tolerance in the species. 相似文献

As the quest for marine-derived compounds with pharmacological and biotechnological potential upsurges, the importance of following regulations and applying Responsible Research and Innovation (RRI) also increases. This article aims at: (1) presenting an overview of regulations and policies at the international and EU level, while demonstrating a variability in their implementation; (2) highlighting the importance of RRI in biodiscovery; and (3) identifying gaps and providing recommendations on how to improve the market acceptability and compliance of novel Blue Biotechnology compounds. This article is the result of the work of the Working Group 4 “Legal aspects, IPR and Ethics” of the COST Action CA18238 Ocean4Biotech, a network of more than 130 Marine Biotechnology scientists and practitioners from 37 countries. Three qualitative surveys (“Understanding of the Responsible Research and Innovation concept”, “Application of the Nagoya Protocol in Your Research”, and “Brief Survey about the experiences regarding the Nagoya Protocol”) indicate awareness and application gaps of RRI, the Nagoya Protocol, and the current status of EU policies relating to Blue Biotechnology. The article categorises the identified gaps into five main categories (awareness, understanding, education, implementation, and enforcement of the Nagoya Protocol) and provides recommendations for mitigating them at the European, national, and organisational level. 相似文献

This is the first report on the effect of light intensity and plant growth‐promoting rhizobacteria (PGPR) on the growth of a tropical forage grass, being a relevant study to improve pasture management in conventional farming and integrated crop‐livestock‐forestry systems. In this study, our aim was to evaluate the effects of light intensity and Burkholderia pyrrocinia and Pseudomonas fluorescens inoculation on Brachiaria brizantha cv. BRS Piatã growth, and phenotypic plasticity to shade. The experiment was conducted in a semi‐controlled environment. Seedlings of B. brizantha were allocated to full sun and shade. P. fluorescens and B. pyrrocinia were inoculated individually or co‐inoculated by soil drench, 14 days after seedling emergence. We evaluated morphogenesis, structural and growth parameters. Irrespective of the light regime, co‐inoculated plants had greater leaf area and SPAD index (chlorophyll content). Increase in total biomass production in co‐inoculated plants was over 100% and 300%, under full sun and shade respectively. Co‐inoculated P. fluorescens and B. pyrrocinia increased shade tolerance in B. brizantha, improving plant performance. Co‐inoculation promoted growth in B. brizantha under both sun and shade, indicating its potential as a bio‐fertilizer in conventional and integrated systems, especially in silvopastoral systems, where light availability to pasture growth may be limited. 相似文献
