Dicroceliosis, a lancet fluke infection, is a frequent parasitosis of small ruminants and the anthelmintic drug albendazole (ABZ) is effective in control of this parasitosis. The aim of our project was to study the metabolism of ABZ and ABZ sulphoxide (ABZ.SO) in lancet fluke. Both invitro (subcellular fractions of fluke homogenates) and exvivo experiments (adult flukes cultivated in medium) were performed for this purpose. ABZ was metabolised invitro by lancet fluke NADPH-dependent enzymes by two oxidative steps (sulphoxidation and sulphonation). The apparent kinetic parameters of these reactions have been determined. In the exvivo experiments, only ABZ sulphoxidation was observed. The stereospecificity in ABZ sulphoxidation invitro was slight, with preferential formation of (+)-ABZ.SO enantiomer. In contrast (−)-ABZ.SO formation predominated in exvivo experiments. Sulphoreduction of ABZ.SO occurred neither invivo nor exvivo. The detection of ABZ oxidative metabolites indicates the presence of drug metabolising oxidases in lancet fluke.  相似文献

Convenience products of fish that are heat processed in-pack are often subjected to a compromise between quality and durability within the framework of required safety. The purpose of this study was to provide knowledge for optimizing pasteurization of cod muscle by quantifying changes in water holding capacity (WHC), cook loss, color, and texture within a wide range of processing times and temperatures. A heat load equivalent to 2 min at 70°C may be applied to the cod while maintaining the WHC above 66%, the cook loss below 5.6%, and keeping hardness and whiteness low, provided that the processing temperature is kept in the range 68 ± 4°C.  相似文献

ABSTRACT

Fish oil was extracted and simultaneously collected into six fractions based on molecular weight and the chain length of triglycerides in terms of fatty acid constituents without splitting of the triglycerides, using supercritical carbon dioxide (SC-CO2) at optimized conditions of 40 MPa, 65°C, and a flow rate 3 mL min?1. In each type of fractionation, the first fraction (F1) was rich in saturated fatty acids (SFA; 52.57 to 61.26%), followed by monounsaturated fatty acids (MUFA; 22.17 to 23.22%) and polyunsaturated fatty acids (PUFA; (0.54 to 20.37%); the sixth fraction (F6) was rich in PUFA (48.93%), followed by MUFA (33.59%) and SFA (13.61%). It was obvious that short-chain fatty acids were extracted at an earlier fraction; therefore, the latter fractions were dominant in long-chain fatty acids, especially MUFA and PUFA. Thus, omega-3 fish oil (last three fractions) was successfully separated to be used as a value-added health product.  相似文献

The current wheat milling process separates bran from endosperm by passing tempered wheat kernels through successive break rolls and sifters. Using hydrolytic enzymes during tempering degrades bran and aleurone layers and can improve milling efficiency and yield. This study was conducted to evaluate the effects of chemical and enzymatic treatments of wheat kernels before milling on physical and milling characteristics of the resulting wheat and flour quality. Hard wheat kernels were soaked in dilute acid or water and dried back to original moisture before being tempered with enzymes in water. Kernel physical and milling characteristics (600 g) were evaluated. Dilute acid soaking did not affect the 1,000‐kernel weight and diameter but softened treated kernels. When treated kernels were pearled, bran removal was mostly from ends; and the reducing sugar content in enzyme‐treated bran was significantly higher than the control. Compared with the control, acid‐soaked enzyme‐tempered kernels showed small but significant improvement in straight flour yield, with virtually no difference in protein content, and flour color. Chemical and enzyme treatment resulted in higher ash in flour. These differences were not seen in milling of larger batches (1,500 g) of kernels.  相似文献

The complete cDNA sequence of the Nile tilapia T-cell receptor (TCR) β chain was cloned using 5' RACE. The full-length, 1263-bp cDNA contained a 942-bp open reading frame (ORF) encoding a 314-amino-acid protein. Sequence analyses revealed that the Nile tilapia TCR β chain contains four conserved cysteine residues involved in the formation of disulphide bridges and a conserved amino acid motif believed to be important for assembly and signalling of the TCR αβ/CD3 complex, both of which are normally found in the TCR β chain of other vertebrates. As detected using semi-quantitative and quantitative RT-PCR, the highest expression level of TCR β was detected in the thymus. Interestingly, Streptococcus agalactiae significantly induced the up-regulation of the TCR β chain, and the strongest up-regulation was detected in the brain and peripheral blood leucocytes (PBLs). In in vitro experiments, concanavalin A and Aeromonas hydrophila were found to significantly increase the expression of the TCR β chain in PBLs after 48 h (P < 0.01) and 72 h (P < 0.05), respectively. Furthermore, real-time PCR analysis showed that intraperitoneal injection (IP) of 10(7) cfu mL(-1) of S. agalactiae could induce TCR β expression that was greater than the expression observed following administration of 10(9) cfu mL(-1). The presence of the TCR β chain in fish detected in this study suggests the presence of T-cell populations that have been found in higher vertebrates, which may play a crucial functional role in the response to fish pathogens.  相似文献

The production performances of giant freshwater prawn Macrobrachium rosenbergii and Nile tilapia Oreochromis niloticus in C/N‐controlled periphyton‐based polyculture systems were evaluated in triplicate. Three different management practices were compared: the traditional practice without addition of periphyton substrates and carbohydrate (Control), addition of maize flour to maintain a carbon: nitrogen rate of 20:1 (treatment CN) and addition of both maize flour and periphyton substrates (treatment CN+P). This experiment used a pre‐optimized stocking density of tilapia and freshwater prawn by Asaduzzaman et al. Aquaculture [286 (2009) 72]. All ponds were stocked with prawn (3 m2) and monosex Nile tilapia (1 m?2). Bamboo side shoots were posted vertically into the pond bottoms as periphyton substrate covering an additional area of 171 m2 for periphyton development. A locally formulated and prepared feed containing 17% crude protein with C/N ratio close to 15:1 was applied twice daily in all ponds considering the body weight of freshwater prawn only. Water quality parameters, except total alkalinity did not vary significantly (> 0.05) among treatments. Both, organic matter and total heterotrophic bacterial loads (THB) in the sediment were significantly (P < 0.05) higher in treatment CN+P followed by treatment CN and control. Periphyton biomass in terms of dry matter and chlorophyll a values constantly decreased during the culture period. Substrates contributed 66% and 102% higher net yield of freshwater prawn than CN and control treatment respectively.  相似文献

Short-term (three to four years) effects of forest harvesting on soil solution chemistry were investigated at two Norway spruce sites in southern Norway, differing in precipitation amount and topography. Experimental plots were either harvested conventionally (stem-only harvesting, SOH) or whole trees, including crowns, twigs and branches were removed (whole-tree harvesting, WTH), leaving residue piles on the ground for some months before removal. The WTH treatment had two sub-treatments: WTH-pile where there had been piles and WTH-removal, from where residues had been removed to make piles. Increased soil solution concentrations of NO3–N, total N, Ca, Mg and K at 30?cm depth, shown by peaks in concentrations in the years after harvesting, were found at the drier, less steep site in eastern Norway after SOH and WTH-pile, but less so after WTH-removal. At the wetter, steeper site in western Norway, peaks were often observed also at WTH-removal plots, which might reflect within-site differences in water pathways due largely to site topography.  相似文献

The released excess anthropogenic nitrogen and carbon produces habitat enrichment, as exemplified by the modification of one-storied pine stands by introducing deciduous species into the understory or second story. In this study, we discuss the validity of pine stand modification by pitfall-trapping epigeic carabid beetles. Two hypotheses were formulated: (1) proportion of late-successional species is higher in assemblages inhabiting pine stands with understory or a second story than in one-storied pine stands; (2) plant litter composition affects carabid beetle assemblages more than other environmental variables. Additionally, characteristic carabid species of the respective pine stand types were identified. GLMM analysis revealed a higher proportion of late-successional species in pine stands with understory or a second story than in one-storied ones. NMDS separated those pine stand types. RDA analysis indicated that pine litter and humus had the strongest effect on carabid beetle assemblage structure in one-storied stands, being drier and thicker in this stand type than in the others. Indicator value analysis identified two characteristic non-forest species in one-storied stands The study revealed that the introduction of understory and particularly a second story into pine stands increased carabid beetle diversity and the proportion of late-successional species, confirming the validity of pine stand modification.  相似文献
