Opioids are the cornerstone for the treatment of acute pain in small animal patients. This is primarily because of their remarkable safety profile, high efficacy, and benefit of reversibility. There have been some significant advances in our knowledge on opioid pharmacology and clinical usage in companion animal medicine. This review discusses the progression of opioid use in small animal practice providing current misconceptions and controversies in light of routes of administration. Potential targets for research and drug development and novel therapies are discussed in addition to the concepts of glial cell modulators, individual variability, and opioid tolerance and hyperalgesia. The future brings an interesting perspective with the application of pharmacogenetics and individualized pain management in canine and feline practice. 相似文献

In future, prolonged summer drought and heat will constitute a major risk for the cultivation of shallow‐rooting beech in Central Europe and will negatively affect the productivity of beech forests. In a pot experiment under controlled conditions, the influence of long‐term (28 d) water deprivation on nitrogen (N), carbon (C), phosphate (Pi), and ascorbate (ASC) concentrations was examined in leaves and fine roots of beech seedlings (Fagus sylvatica L.) from six provenances originating from Central Europe (Germany: Neidenstein and Illertissen, intermediate habitats), the Balkan peninsula (Croatia: Zagreb and Gospic, wet habitats), and Southeast Europe (Bulgaria: Kotel, Greece: Paikos; dry habitats). The goal of the study was to identify beech provenances well adapted to water limitation during summer drought events. Our results suggest that N might be involved in the alleviation of water scarcity, whereas Pi might become a limiting factor for forest growth during drought periods. Drought stress resulted in significant changes of ASC pools in leaves and fine roots and the ASC redox state. Under well‐watered and under drought conditions, ASC in leaves was the most important factor causing differences between the provenances examined. Finally, a link between P nutrition and the capacity of antioxidative stress defense by ascorbate could be highlighted. Based on observations from this study, beech seedlings from three origins (Paikos, Zagreb, and Neidenstein) might constitute beech provenances well adapted to water shortage in summer. Shifts in the climate sensitivity of trees throughout the twentieth century might indicate climate change effects in the boreal forest ecosystem. We here evaluated such potential changes by analyzing six tree-ring width (TRW) and maximum latewood density (MXD) chronologies from northern, central and southern boreal forests in Finland (60°N–69°N). Besides latitudinal effects, differing micro-sites (lakeshore and inland) were considered to evaluate the influence of ground water access on twentieth-century tree-ring formation and climate sensitivity. Overall, the boreal MXD chronologies appeared less affected by micro-site conditions compared to the TRW chronologies. Along the boreal transect, mean growth rates decrease with increasing latitude, but the ratio of earlywood-to-latewood (~70%/~30%) remains stable. However, latewood is slightly denser at the dry inland micro-sites. The correlations with climate data showed that TRW and MXD at all sites are positively related with summer temperature. The MXD chronologies are also negatively correlated with precipitation, a feature that is largely missing in TRW. A recent trend toward wetter and cloudier conditions in the study region coincides with a shift of the MXD signal from a distinct temperature limitation toward an additional sensitivity to precipitation, which in turn stimulated the trees’ competition for sunlight. 相似文献

Background
In Ireland, every animal is examined at slaughter for its fitness for human consumption. The aim of this study was to determine the relative effectiveness of factories in submitting and subsequently in having suspect lesions confirmed as bovine tuberculosis (TB) lesions during the years 2005-2007. This work provides an update from previously published data for years 2003-2004. During 2005-2007 data were available on 4,401,813 cattle from attested herds (i.e. herds classified free of bovine TB), from which data for potential confounding factors were available for 3,344,057 slaughtered animals at one of the 37 export-licensed factories.Findings
From these animals, 8,178 suspect lesions were submitted for laboratory confirmation. Lesions from 5,456 (66.7%) animals tested as positive, and 269 (3.2%) were inconclusive for bovine TB. Logistic regression was used to determine adjusted submission and confirmation risks for each factory while controlling for confounding factors. Factory rankings based on adjusted and crude risks were similar. The average crude submission risk for all the factories was 25 lesions per 10,000 animals slaughtered, ranging from 0 to 52. The crude confirmation risk varied between 30.3% and 91.3%.Conclusions
Substantial variation in the effectiveness of lesion submission and subsequent confirmation as bovine TB was found among the 37 factories. Compared to previous years (2003-2004), there was an increased bovine TB lesion submission and confirmation risk. Continued monitoring of the effectiveness of slaughter surveillance in Ireland is recommended; emphasis should be placed on efforts to improve bovine TB surveillance in factories with lower rankings. 相似文献946.

Swine influenza virus (SIV) and Mycoplasma hyopneumoniae (Mhp) are widespread in farms and are major pathogens involved in the porcine respiratory disease complex (PRDC). The aim of this experiment was to compare the pathogenicity of European avian-like swine H1N1 and European human-like reassortant swine H1N2 viruses in na?ve pigs and in pigs previously infected with Mhp. Six groups of SPF pigs were inoculated intra-tracheally with either Mhp, or H1N1, or H1N2 or Mhp+H1N1 or Mhp+H1N2, both pathogens being inoculated at 21 days intervals in these two last groups. A mock-infected group was included. Although both SIV strains induced clinical signs when singly inoculated, results indicated that the H1N2 SIV was more pathogenic than the H1N1 virus, with an earlier shedding and a greater spread in lungs. Initial infection with Mhp before SIV inoculation increased flu clinical signs and pathogenesis (hyperthermia, loss of appetite, pneumonia lesions) due to the H1N1 virus but did not modify significantly outcomes of H1N2 infection. Thus, Mhp and SIV H1N1 appeared to act synergistically, whereas Mhp and SIV H1N2 would compete, as H1N2 infection led to the elimination of Mhp in lung diaphragmatic lobes. In conclusion, SIV would be a risk factor for the severity of respiratory disorders when associated with Mhp, depending on the viral subtype involved. This experimental model of coinfection with Mhp and avian-like swine H1N1 is a relevant tool for studying the pathogenesis of SIV-associated PRDC and testing intervention strategies for the control of the disease. 相似文献

Southern Hemisphere plantation forestry has grown substantially over the past few decades and will play an increasing role in fibre production and carbon sequestration in future. The sustainability of these plantations is, however, increasingly under pressure from introduced pests. This pressure requires an urgent and matching increase in the speed and efficiency at which tools are developed to monitor and control these pests.Aim
To consider the potential role of semiochemicals to address the need for more efficient pest control in Southern Hemisphere plantations, particularly by drawing from research in other parts of the world.Results
Semiochemical research in forestry has grown exponentially over the last 40?years but has been almost exclusively focussed on Northern Hemisphere forests. In these forests, semiochemicals have played an important role to enhance the efficiency of integrated pest management programmes. An analysis of semiochemical research from 1970 to 2010 showed a rapid increase over time. It also indicated that pheromones have been the most extensively studied type of semiochemical in forestry, contributing to 92% of the semiochemical literature over this period, compared with research on plant kairomones. This research has led to numerous applications in detection of new invasions, monitoring population levels and spread, in addition to controlling pests by mass trapping or disrupting of aggregation and mating signals.Conclusion
The value of semiochemicals as an environmentally benign and efficient approach to managing forest plantation pests in the Southern Hemisphere seems obvious. There is, however, a lack of research capacity and focus to optimally capture this opportunity. Understanding the mechanisms that maintain protective antibody levels after immunisation is important for vaccine design. In this study, we have determined the kinetics of plasma and memory B cells detectable in the blood of cattle immunised with model T-dependent or T-independent antigens. Immunisation with the T-D antigen resulted in an expansion of TNP-specific plasma cells post-TNP primary and booster immunisations, which was associated with increased titres of TNP-specific IgG antibodies. Although no TNP-specific memory B cells were detected in the T-D group following the primary immunisation, we detected an increase in the number of TNP-specific memory B cells post-TNP boost. In contrast, no TNP-specific plasma or memory B cells were detected after primary or secondary immunisation with the T-I antigen. We then investigated if immunisation with a third party antigen (tetanus toxin fragment C, TTC) would result in a bystander stimulation and increase the number of TNP-specific plasma and memory B cells in the T-D and/or T-I group. TTC immunisation in the T-D group resulted in a small increase in the number of TNP-specific plasma cells post-TTC primary immunisation and boost, and in an increase in the number of TNP-specific memory B cells post-TTC boost. This bystander effect was not observed in the animals previously immunised with the T-I antigen. In conclusion, the present study characterised for the first time the B cell response in cattle to immunisation with T-D and T-I antigens and showed that bystander stimulation of an established T-D B cell memory response may occur in cattle. 相似文献

γδ T cells represent an unconventional subset of T lymphocytes that are abundant in epithelial tissues and serve as an early immune defense against microbes. We have, for the first time, identified γδ T cells in steady-state thoracic duct lymph (TDL) from rats. The lymph contains γδ T cells expressing CD8 but not CD4, CD25, MHC-II or CD103. The percentage of TDL γδ T cells in rats does not change when the mesenteric lymph nodes (MLN) are surgically removed. Cystotomy is a common surgical procedure in small animal veterinary medicine, yet common pre-, intra-, and post-operative practices have not been described. This survey evaluated cystotomy practices of 106 veterinarians in Ontario. The majority of respondents reported practices consistent with standard recommendations, but some deficiencies in antimicrobial and analgesic use, as well as intra- and post-operative practices, were identified. Some factors associated with the likelihood that practitioners reported recurrent urolithiasis or urinary tract infection are contrary to typical recommendations, such as the association of the use of absorbable, multifilament suture or a dorsal (versus ventral) incision and a lesser likelihood of reporting post-operative urinary tract infections. While care must be taken interpreting these statistical associations, the results suggest that objective assessment of common cystotomy recommendations (use of monofilament, absorbable suture) is required. Re-assessment of certain peri-operative practices, such as analgesic and antimicrobial administration, and post-operative testing, is required for a minority of practitioners. 相似文献
