Biosynthetic and chemical datasets are the two major pillars for microbial drug discovery in the omics era. Despite the advancement of analysis tools and platforms for multi-strain metabolomics and genomics, linking these information sources remains a considerable bottleneck in strain prioritisation and natural product discovery. In this study, molecular networking of the 100 metabolite extracts derived from applying the OSMAC approach to 25 Polar bacterial strains, showed growth media specificity and potential chemical novelty was suggested. Moreover, the metabolite extracts were screened for antibacterial activity and promising selective bioactivity against drug-persistent pathogens such as Klebsiella pneumoniae and Acinetobacter baumannii was observed. Genome sequencing data were combined with metabolomics experiments in the recently developed computational approach, NPLinker, which was used to link BGC and molecular features to prioritise strains for further investigation based on biosynthetic and chemical information. Herein, we putatively identified the known metabolites ectoine and chrloramphenicol which, through NPLinker, were linked to their associated BGCs. The metabologenomics approach followed in this study can potentially be applied to any large microbial datasets for accelerating the discovery of new (bioactive) specialised metabolites. 相似文献

ABSTRACT Maize production is affected by water and nitrogen (N) deficit either separately or joined, but this fact is not completely defined. The aim was to evaluate the fate of N in maize fertilized and subjected to water stress in controlled conditions. A greenhouse experiment was carried out at the University of Buenos Aires campus. The design was a 2 × 2 factorial with four replications. The factors were N: 70 and 140 kg N ha?1 as labeled urea (15N), and water: 100% or 50% of the potential evapotranspiration. The harvest of aerial and root biomass was carried out at R1 stage. Nitrogen in plants, soils nitrate, ammonia volatilization, and 15N percentage were determined. Obtained results only partially agree with previous research. Water stress depressed aerial biomass production independently of N doses. When water was limiting, the uptake of N from fertilizer was independent of N. When water was not limiting, N uptake increased with the higher N doses. Volatilization losses were 3.7 to 7.8% of N applied as fertilizer. Plant N recovers was around 45% of the N applied, except in treatment water stressed with high N rate (19%). Nitrate-N from the fertilizer in the soil at harvest and accumulated N from the fertilizer in plant were lineally related (r2 = 0.54; p < 0.001). Important destinations of N were accumulation in plant, volatilization and incorporation into soil organic matter. - 1. Growing concern associated with threats to the marine environment has resulted in an increased demand for marine reserves that conserve representative and adequate examples of biodiversity. Often, the decisions about where to locate reserves must be made in the absence of detailed information on the patterns of distribution of the biota. Alternative approaches are required that include defining habitats using surrogates for biodiversity. Surrogate measures of biodiversity enable decisions about where to locate marine reserves to be made more reliably in the absence of detailed data on the distribution of species.
- 2. Intertidal habitat types derived using physical properties of the shoreline were used as a surrogate for intertidal biodiversity to assist with the identification of sites for inclusion in a candidate system of intertidal marine reserves for 17 463 km of the mainland coast of Queensland, Australia. This represents the first systematic approach, on essentially one‐dimensional data, using fine‐scale (tens to hundreds of metres) intertidal habitats to identify a system of marine reserves for such a large length of coast. A range of solutions would provide for the protection of a representative example of intertidal habitats in Queensland.
- 3. The design and planning of marine and terrestrial protected areas systems should not be undertaken independently of each other because it is likely to lead to inadequate representation of intertidal habitats in either system. The development of reserve systems specially designed to protect intertidal habitats should be integrated into the design of terrestrial and marine protected area systems.

- Monitoring the abundance of mobile and wide‐ranging cetacean populations for conservation management is challenging, especially when the management is focused on static protected areas. Where abundance estimates are derived from mark–recapture data, such as photo‐identification of naturally marked individuals, unpredictable movements of animals in and out of the survey area can reduce ‘capture’ probabilities and affect the precision and accuracy of resulting estimates.
- A Bayesian hierarchical log–linear model was applied to photo‐identification data collected in summer 2014 to derive a multi‐site abundance estimate for a population of bottlenose dolphins, Tursiops truncatus, ranging widely throughout the coastal waters of western Ireland. In addition, the effects of varying levels of sampling effort on the minimum detectable decrease in population size were examined.
- The abundance (median) of dolphins was estimated as 189 (coefficient of variation (standard deviation/mean), 0.11; 95% highest‐posterior density interval, 162–232). Over 50% of the well‐marked dolphins encountered throughout the study were sighted in more than one distinct coastal area, thus displaying high mobility. In addition, it was found that it would require biennial surveys to detect a 25% decline in abundance within the six‐year reporting period of the EU’s Habitats Directive.
- Given that the Special Area of Conservation designated for these dolphins consists of two separate areas covering a substantial portion of the west coast of Ireland, the multisite approach is appropriate for monitoring this population. It produces a more precise estimate and is well suited for sparse recapture data collected opportunistically at multiple sites, when the lack of resources prevents large‐scale surveys or when concentrating surveys on smaller localized areas fails to capture the broad range and unpredictable occurrence of the animals. The Bayesian multi‐site approach could be applied to the management of other wide‐ranging marine or terrestrial taxa.

- Considering the deteriorated state of the ocean, maximizing the impact of scientists' media communication is essential for gaining public interest in, and support for, marine conservation initiatives. Unlike conventional news, that involves journalists' interpretation, the media Q&A offers scientists a way to convey insights on complex marine ecosystem issues in their own words. However, the Q&A approach for communicating marine science and conservation issues has not been empirically examined for format-related differences in impact.
- The modality theory in multimedia learning suggests that information is more effective for accelerating learning and retention when visuals are combined with narrated words, than when combined with written text. This study compared the effects of identically framed information presented in two different media Q&A formats: narrated multimedia (a video on YouTube) and written text with photos (designed to resemble an online mainstream news article). In both formats, a marine scientist explained the complex phenomenon of marine ecosystem tipping points.
- In an online experiment, 116 respondents were randomly assigned to either view the video or read the article. Effects on knowledge, awareness, and motivation were tested for format-related differences. Data related to time spent on the interventions were used to test the effects of attention retention on awareness and motivation gains. Additionally, the study gauged participants' assessment about the effectiveness of the communication features and framings used and suggestions for enhancing communication effectiveness.
- Findings showed positive effects on knowledge, awareness, and motivation irrespective of media format, suggesting that other communication aspects such as the messengers, the scientist's communication style, educative content, and message frames probably contributed to impact. Findings also suggest that multimedia formats, which tend to be more costly, may not necessarily be more effective.

- As the proportion of threatened species increases, so too does the need for effective conservation strategies. In response, captive breed-and-release and habitat mitigation programmes are two conservation actions that are increasing in use and effectiveness.
- Success of these programmes is frequently hampered by the continued presence of threatening processes. In the case of amphibian reintroductions, a key threatening process that is difficult to eliminate is the deadly fungal pathogen, Batrachochytrium dendrobatidis (Bd). This pathogen is the proximate cause of decline for the threatened green and golden bell frog, Litoria aurea, and has contributed to the failure of previous breed-and-release programmes of this amphibian.
- To investigate whether is it possible to establish a captive-bred population of L. aurea that could persist in the presence of Bd, a breed-and-release programme was conducted in a created habitat that included increased salinity to inhibit fungal growth rates, exclusion of potential reservoir hosts and source animals bred from parents ‘experienced’ with the strain of Bd present in the release environment.
- The population persisted in the created habitat for more than 4 years (the timescale of monitoring) but suffered higher infection levels compared with nearby extant populations of L. aurea. These infections significantly reduced apparent survival. Infection dynamics were linked to temperature, host density in the previous season, and size and age of the host, and these factors differed from those in extant populations and are likely to have contributed to the high levels of Bd infection at the created site.
- This article discusses how it may be possible to manipulate these factors to improve the success of future breed-and-release programmes, and recommends the construction of warm water bodies and a strategy of prioritizing the release of fewer, large propagules of high body condition and mixed-age class, over large numbers of younger, smaller animals.
