Phylogenetic relationships among natural populations of perennial buckwheat, Fagopyrum cymosum Meisn., revealed by allozyme variation 总被引:2,自引:0,他引:2
Twenty natural populations of F. cymosum, an insect-pollinated allogamous wild species of Fagopyrum, were investigated for their chromosome number and allozyme variation at 10 Ioci encoding 8 enzymes. Diploid populations were obtained in Sichuan, Yunnan and Tibet in China, whereas tetraploid populations were obtained in Tibet, the Himalayan hills, Thailand and southern China. Both diploid and tetraploid populations maintained a large amount of allozyme variation. The average heterozygosity, He, ranged from 0.045 to 0.389 (0.213 on average). Tetraploid populations showed higher He values than diploid populations. Natural populations of F. cymosum were locally well differentiated (GST = 0.322), probably due to reproductive isolation between the two ploidy levels and to distribution over wide areas from southern China to the Himalayan hills. The phylogenetic tree constructed by the neighbor-joining method based on allozyme variation clarified two distinct groups of diploid populations, the Sichuan and Yunnan groups. As for the tetraploid populations, polyploidization occurred twice independently, once in Yunnan and once in eastern Tibet. Tetraploid F. cymosum diffused to warmer areas such as southern Yunnan and Thailand as well as to cooler areas of Tibet and the high Himalayan hills and became the most widely distributed of the Fagopyrum species. 相似文献

Neospora is a cyst-forming coccidian parasite that causes abortions and neuromuscular disorders in a wide variety of mammals. Japanese bovine isolate JPA1 was inoculated intraperitoneally into BALB/c nu/ nu (athymic nude) and BALB/c (congenic wild type) female mice to examine the distribution of parasites and resistance mechanisms to Neospora infection. All the athymic nude mice died within 28 days after intraperitoneal injection of 2 x 10(5) JPA1 tachyzoites, whereas all the congenic wild type mice survived without exhibiting any clinical signs. Tachyzoites were identified in the uterus and pancreas and later spread to many other organs. Most tachyzoites identified in the necrotic foci were localized in the epithelium of the venules and capillaries. Nude mice developed high level of serum interferon-gamma and interleukin-6 as infection proceeded. Inflammatory response to Neospora infection might be mediated by Th1-type dependent cellular immunity. 相似文献

An attempt was made to induce ovulation in 19 cats during the non-breeding season by the administration of a porcine pituitary gland preparation. The dosage was 2 mg for the first day and then 1 mg for each day until estrus was induced. Studies were made to determine whether fertilization and pregnancy were possible by mating at the time of induced estrus. Estrus was induced in 89.5% (17/19) of the cats, 3-8 days after the first administration. The average number of ovulations induced by mating was 7.3 +/- 1.3 (mean +/- SE) for 16 cats (excluding one cat that failed to ovulate). The uterine horn was flushed 6-8 days after mating in 9 of the 16 ovulated cats to determine the fertilization rate. An average of 3.6 ova were recovered from 8 out of the 9 cats (including one cat from which no ovum was recovered) which gave an average recovery rate of 38.6%. Expanded blastocysts were recovered from 5 cats, while from the other 3 cats, only unfertilized ova were recovered. Pregnancy was confirmed in the other 7 cats 15-20 days after mating. In two of the 7 cats implantation was observed, but abortion occurred at 24 and 27 days of pregnancy. Plasma progesterone concentrations were lower in the estrus-induced and bred cats as compared to these in the controls, with progesterone values approaching base concentrations, approximately by day 20 in 2 cats and by day 30 in 3 cats. OBJECTIVE: To describe the use of a detachable coil for transcatheter closure (TCC) of patent ductus arteriosus (PDA) in 2 dogs. STUDY DESIGN: Clinical study. ANIMALS: Two female Pembroke Welsh Corgi dogs with PDA. METHODS: Using fluoroscopic guidance, an 8-mm-diameter coil stent with 5 loops (detachable coils for PDA closure) was inserted via catheterization of the femoral artery. The catheter was passed through the PDA into the pulmonary artery. The coil was withdrawn so that 1.5 loops remained on the pulmonary side of the orifice of the ductus. The rest of the loops were pushed out from the catheter into the ductus. After confirming the correct placement of the coil and the effectiveness of the occlusion, the delivery wire was detached from the coil. RESULTS: Insertion of the coil was easily performed, even without previous experience. Immediate and marked decrease of the cardiac murmur was auscultated. Only slight residual flow was detected by angiography conducted 3 months' postoperatively. The dogs experienced quick and uneventful recovery after coil placement and required minimal postoperative care. Follow-up evaluation of the dogs showed no functional clinical signs of PDA, and no cardiac abnormalities were detected on electrocardiographic, phonocardiographic, and echocardiographic examination. In dog 1, the residual flow had disappeared on the color-flow Doppler echocardiographic examination at 18 months' postoperatively. CONCLUSION: TCC using a detachable coil was easy, safe, and effective in 2 dogs with PDA. The minimal residual shunting observed only by echocardiography seemed hemodynamically insignificant. On single oral administration of (14)C-S-53482 [7-fluoro-6-(3,4,5, 6-tetrahydrophthalimido)-4-(2-propynyl)-2H-1,4-benzoxazin-3( 4H)-one, Flumioxazin] labeled at the 1- and 2-positions of tetrahydrophthaloyl group to rats at 1 (low dose) or 100 (high dose) mg/kg, the radiocarbon was almost completely eliminated within 7 days after administration in both groups with generally very low residual (14)C tissue levels. The predominant excretion route was via the feces. The major fecal and urinary metabolites involved reduction or sulfonic acid addition reactions at the 1,2-double bond of the 3,4,5,6-tetrahydrophthalimide moiety and hydroxylation of the cyclohexene or cyclohexane ring. One urinary and four fecal metabolites were identified using chromatographic techniques and spectroanalyses (NMR and MS). Three of five identified metabolites were unique forms, reduced at the 1,2-double bond of the 3,4,5, 6-tetrahydrophthalimide moiety. On the basis of the metabolites identified in this study, the metabolic pathways of S-53482 in rats are proposed. To specify tissues forming reduced metabolites, an in vitro study was conducted. The effects of steam sterilization (SS) on soil microbial properties including metabolic diversity of the microbial communities were examined in a greenhouse compared with those of two fumigants, methyl bromide (MB) and chloropicrin (CP). The numbers of fungi decreased in all the treatments. Nitrifiers, both ammonium-oxidizing bacteria and nitrite-oxidizing bacteria, were severely affected by the SS and CP treatments, resulting in their virtual disappearance. The decrease in the levels of microbial biomass C and N after the treatments suggested that the SS and CP treatments eradicated the microorganisms more effectively than the MB treatment and that the influence of the former persisted until the end of the experiment, 4 months after the treatments. Accumulation of NH4-N was observed after the SS and CP treatments mainly due to the partial decomposition of the dead microorganisms and the marked decrease in the number of ammonium-oxidizing bacteria. The richness and average well color development (AWCD) values in the microbial communities after the SS treatment were evaluated by the carbon substrate utilization method using Biolog ECO MicroPlates. The values decreased markedly immediately after the treatment but showed a rapid recovery, while those after the CP treatment continued to decrease until the transplanting of tomato seedlings. The effect of the MB treatment on the soil microbial communities was less pronounced. The growth of the tomato plants was promoted by the SS and CP treatments due to the increase in the N supply at the initial stage of tomato growth. 相似文献

Estuarine and marine habitat use patterns in the halfbeak Zenarchopterus dunckeri were examined at Iriomote Island, southern Japan, by analyzing otolith Li/Ca and Sr/Ca ratios. The ranges of both Li/Ca and Sr/Ca ratios in juvenile Z. dunckeri from the maximum (30 psu) to minimum (0.5 psu) salinity levels of brackish water estimated from rearing experiments, were compared with those of wild individuals collected from upstream and downstream stations in the Urauchi River estuary. The majority of wild-caught individuals had invariable Li/Ca and Sr/Ca ratios along an otolith transect from the core to the posterior edge, which fell within the otolith Li/Ca and Sr/Ca ranges estimated for estuarine individuals in the rearing experiments, suggesting that such individuals developed within the estuary without migrating to a marine environment at any time, although some downstream-dwelling fish had higher otolith elemental ratios than the predetermined estuarine ranges in the mid transect section. The latter fish may have been accidentally flushed from the estuary into the sea by heavy flood events, subsequently returning to the estuary. The overall results suggested that Z. dunckeri is essentially an estuarine resident, completing its life cycle within an estuarine system. 相似文献

A 22-month-old, male Labrador retriever was presented with anorexia, dyspnoea, and fainting. The dog was diagnosed with a double-chambered right ventricle and tricuspid valve dysplasia using echocardiography and cardiac catheterisation. A marked bilateral pleural effusion was also present and chemical analysis of the fluid confirmed the diagnosis of chylothorax. Using echocardiography, a pressure gradient of 87.1 mmHg was found between the proximal and distal chambers of the double-chambered right ventricle. Initiation of cardiopulmonary bypass allowed the anomalous muscle bundle that divided the right ventricle into two chambers to be resected via a right ventriculotomy. The fainting completely resolved postoperatively, and this treatment seemed quite effective in the reduction of pressure overload ascribable to ejection disturbance. Because the tricuspid dysplasia was not corrected in the first operation, the postoperative chyle effusion was reduced but did not cease. A combination of thoracic duct ligation and passive pleuroperitoneal shunting was effective in the resolution of the chyle effusion. 相似文献

Waterlogging is the constraint for soybean growth and yield, because soybean is often cultivated in upland fields converted from paddy in Japan. However, efficient cultivation techniques for alleviating the adverse effects have not been developed. We have proposed the new soybean cultivation technique named crack fertilization which enables yield increase due to enhancing new root growth and N acquisition by increasing nodulation. Waterlogging induces N deficiency due to the suppression of nutrient uptake by the inhibition of root growth and nodule activity. Thus, it is hypothesized that crack fertilization would be effective to alleviate the inhibition of soybean growth and yield. The soybean cultivar of Sachiyutaka was planted in 1/5000 a Wagner pots and root boxes. Two separate waterlogging treatments were imposed to soybean plants at different growth stages, V1 and R4, and crack fertilization was done at V3. After these treatments, soybean plants were sampled at R5 in 2012 and 2013 experiments, respectively. Waterlogging at V1 and R4 inhibited the growth and yield of soybean and nodule growth, and the decreases in physiological parameters of soybean such as photosynthesis, chlorophyll content, and xylem sap exudation rate were observed. The adverse effects of waterlogging at V1 were alleviated by crack fertilization at V3, whereas crack fertilization could not alleviate the adverse effects of waterlogging at R4. Thus, crack fertilization after waterlogging at early vegetative stage would be the cultivation technique that enables to alleviate the adverse effects. 相似文献
