The single-dose disposition kinetics of cefonicid were determined in clinically normal lactating goats (n = 6) after intravenous (IV), intramuscular (IM) and subcutaneous (SC) administration of a conventional formulation, and after subcutaneous administration of a long-acting formulation (SC-LA). Cefonicid concentrations were determined by high performance liquid chromatography with ultraviolet detection. The concentration–time data were analysed by noncompartmental pharmacokinetic methods. Steady-state volume of distribution (Vss) and clearance (Cl) of cefonicid after IV administration were 0.14 ± 0.03 L/kg and 0.51 ± 0.07 L/h·kg, respectively. Following IM, SC and SC-LA administration, cefonicid achieved maximum plasma concentrations of 14.46 ± 0.82, 11.98 ± 1.92 and 17.17 ± 2.45 mg/L at 0.26 ± 0.13, 0.42 ± 0.13 and 0.83 ± 0.20 hr, respectively. The absolute bioavailabilities after IM, SC and SC-LA routes were 75.34 ± 11.28%, 71.03 ± 19.14% and 102.84 ± 15.155%, respectively. After cefonicid analysis from milk samples, no concentrations were found above LOQ at any sampling time. From these data, cefonicid administered at 20 mg/kg each 12 hr after SC-LA could be effective to treat bacterial infections in lactating animals not affected by mastitis problems. 相似文献

Common beans have a high nutritional value, but contain galactooligosaccharides (GO), which cause flatulence and intestinal discomfort in humans. The biochemical composition of ten bean cultivars was determined to select those of high protein and low GO contents. The cultivars varied in carbohydrate (47.02–60.17%), GO (3.12 – 5.71%), protein (22.17–33.50%), lipid (1.13–1.81%), moisture (11.42–12.93%) and ash contents (4.08–5.61%). ‘Mexico 222’ presented the highest α-galactosidase activity. Protein and GO contents were positively correlated. ‘Perry Marrow’ combined high protein and low GO concentrations, indicating it can be used in improvement programs aiming at high-quality cultivars for human consumption. 相似文献

Digestibility, fecal characteristics, and levels of glucose and urea in the plasma were determined in 8 dogs that received 2 different dog foods once or 3 times daily. One dog food (A) was 5 times more expensive than the other (B). Fecal pH and consistency, digestibility of dry matter (DM), organic matter (OM), crude protein (CP), and crude fiber (CF) were determined. Blood samples were taken from 30 min before to 60 min after a meal. Digestibilities of DM, OM, and CP, and fecal consistency were higher, and daily fecal excretion and fecal pH were lower when dogs were fed food A (P < 0.001). The feeding schedule had no effect on plasma glucose and urea. Neither feeding frequency nor food × frequency interactions was significant for the parameters studied. 相似文献

Purpose
Copper (Cu) contamination has been increasing in land ecosystems due to economic development activities. Excessive amount of Cu in soils is toxic to both plants and microorganisms. Biochar (BC) is known to immobilize soil Cu. The objectives of this research were to investigate the effects of chicken-manure-derived BC (CMB) on Cu immobilization, and growth of native metallophyte Oenothera picensis in a Cu-contaminated soil.Materials and methods
A Cu-contaminated sandy soil (338 mg Cu kg?1) was spiked and equilibrated with additional Cu (0, 100, and 500 mg Cu kg?1). The spiked soil was then amended with CMB (0, 5, and 10 % w/w) and incubated for 2 weeks. The metallophyte was grown on these treatments under greenhouse conditions for 3 months. Pore water solutions were collected from the plant pots every 30 days. After the harvest, soil and pore water pH, soil Cu fractions, pore water Cu concentration, soil microbial activity, plant biomass weight, and Cu concentration in plant parts were determined.Results and discussion
The CMB increased the pH of soils and soil pore water, and probably also soil major nutrients. It reduced the exchangeable fraction of Cu but increased its organic matter and residual fractions. At the same time, it decreased the Cu concentration in the soil pore water. The CMB increased basal respiration and dehydrogenase activity. The CMB application produced up to three and seven times more root and shoot biomass, respectively. In addition, shoots accumulated lesser Cu than control but roots did more. Plants survived in soil that was spiked with 500 mg Cu kg?1, only when CMB dose was 10 %.Conclusions
The CMB affected the Cu uptake in plant by altering the mobility, bioavailability, and spatial distribution of Cu in soils. The increase in available nutrients and decrease in Cu toxicity facilitated plant growth. The increased microbial activity probably also promoted the plant growth and reduced the Cu bioavailability. Therefore, CMB can be used to remediate Cu-contaminated soils.66.

A controlled field trial was conducted to assess the potential influence of practitioner inexperience during early pregnancy diagnosis with ultrasound (PD‐US) on the risk of pregnancy loss. A veterinarian with more than 10 years’ experience in PD‐US (Vet‐A) and a veterinarian with fewer than 12 months’ experience at the start of the study (Vet‐B) visited the same dairy farm once a week for 33 and 26 weeks, respectively. The two veterinarians did not interact with each other at any time during the study, nor did they know that their data would later be used in this study. Using the same farm scanner, they performed PD‐US at 28–34 day after breeding, together diagnosing 915 pregnancies. All cows were re‐checked at 49–56 day after artificial insemination, and cows no longer pregnant were recorded as having suffered pregnancy loss. Although Vet‐A and Vet‐B diagnosed a similar proportion of pregnancies (58.44 ± 16% vs 56.96 ± 18%, p > .05), the rate of pregnancy loss was significantly higher among cows diagnosed by Vet‐B (10.41 ± 11.2% vs 4.87 ± 9.0, p = .029). In addition, among cows diagnosed by Vet‐B, the rate of pregnancy loss was significantly higher among cows diagnosed, while he had fewer than 12 months’ PD‐US experience (11.17 ± 12.14%) than among cows that he diagnosed later (7.14 ± 11.01%, p = .038); in fact, this latter loss rate was comparable to that among cows diagnosed by Vet‐A during the same period (3.51 ± 9.83%, p = .620). These results suggest that inexperience with PD‐US during the late embryonic period can increase risk of early pregnancy loss, supporting the need for proper training. 相似文献

Computer‐assisted systems for the assessment of sperm morphometry (ASMA systems) have been used successfully with several mammalian species. Unfortunately, they have so far been of little use for assessing bird semen, a consequence of the filiform shape of avian spermatozoa. This study compares two staining techniques (Hemacolor® and aniline blue staining) for the morphometric analysis of rooster and red‐legged partridge spermatozoa as part of a computer‐assisted light microscopy method. For both species, Hemacolor® staining provided a significantly higher percentage of measurable cells (93.7 ± 11.7% in roosters and 71.9 ± 15.3% in red‐legged partridges). Hemacolor® also showed greater repeatability (lower coefficients of variation) for length and area in roosters' sperm and for width in the case of red‐legged partridge's sperm. In the roosters, the Hemacolor® technique returned significantly (p < 0.05) larger sperm head width and area values than did the aniline blue technique, while the latter resulted in greater sperm head length values (p < 0.05). In the red‐legged partridge, no differences were seen in the results for sperm head width and area provided by the two techniques, but aniline blue staining was associated with longer length measurements. In conclusion, the morphometric values recorded differed depending on the staining method and species. However, the Hemacolor® technique might be deemed the more appropriate for computerized sperm assessment systems as it provides larger percentages of measureable cells and shows greater repeatability. 相似文献

This work examines the effect of the interaction between different concentrations of two cryoprotectants – glycerol (GLY) and dimethylacetamide (DMA) – and two methods of cryopreservation – pellets produced by plunging into liquid nitrogen and gradual in‐straw freezing – on frozen/thawed chicken sperm variables. Sperm was cryopreserved using: (i) 6% DMA, following the in‐straw and the pellet methods (ii) 11% GLY, following the in‐straw and the pellet methods; and (iii) 8% GLY in the in‐straw method and 3% DMA in the pellet method (i.e. reduced cryoprotectant concentrations). When 6% DMA was used as the cryoprotectant, no differences were seen between the in‐straw and pellet methods in terms of frozen/thawed sperm variables or fertility (10.8% and 12.8%, respectively). The viability and motility variables of the frozen/thawed sperm produced using the in‐straw method with 11% GLY were higher (p < 0.05) than those recorded for the sperm preserved using the same cryoprotectant and concentration in the pellet method. However, fertility was extremely low in both groups (2.1% and 4.2% for the in‐straw and pellet methods, respectively). Finally, the use of 8% GLY in the in‐straw method returned higher sperm viability, intact acrosome and motility values than the use of 3% DMA in the pellet method (p < 0.01). No differences were seen, however, in the fertility results obtained (28.8% and 25.0%, respectively). These results suggest that cryoprotectant concentrations can be reduced and still provide acceptable fertility rates. 相似文献

Dissection is a very useful method of learning veterinary anatomy. However, formaldehyde, which is widely used to preserve cadavers, is an irritant, and it has recently been classified as a carcinogen. In 1997, the Instituto Nacional de Seguridad e Higiene en el Trabajo [National Institute of Workplace Security and Hygiene] found that the levels of formaldehyde in our dissection room were above the threshold limit values. Unfortunately, no optimal substitute for formaldehyde is currently available. Therefore, we designed a new ventilation system that combines slow propulsion of fresh air from above the dissection table and rapid aspiration of polluted air from the perimeter. Formaldehyde measurements performed in 2004, after the introduction of this new system into our dissection laboratory, showed a dramatic reduction (about tenfold, or 0.03 ppm). A suitable propelling/aspirating air system successfully reduces the concentration of formaldehyde in the dissection room, significantly improving safety conditions for students, instructors, and technical staff during gross anatomy learning. 相似文献
