Using genetic tools to inform conservation of fragmented populations of Asian elephants (Elephas maximus) across their range in China

A herd of 15 Chinese elephants attracted international attention during their 2021 northward trek, motivating the government to propose establishment of an Asian elephant national park. However, planning is hampered by a lack of genetic information on the remaining populations in China. We collected DNA from 497 dung samples from all 5 populations encompassing the entire range of elephants in China and used mitochondrial and microsatellite markers to investigate their genetic and demographic structure. We identified 237 unique genotypes (153 females, 84 males), representing 81% of the known population. However, the effective population size was small (28, range 25–32). Historic demographic contraction appeared to account for low haplotype diversity (H_d = 0.235), but moderate nucleotide and nuclear diversity (π = 0.6%, H_e = 0.55) was attributable to post-bottleneck recovery involving recent population expansion plus historical gene exchange with elephants in Myanmar, Lao PDR, and Vietnam. The 5 populations fell into 3 clusters, with Nangunhe elephants differing consistently from the other 4 populations (F_ST = 0.23); elephants from Mengyang, Simao, and Jiangcheng belonged to a single population (henceforth, MSJ), and differed from the Shangyong population (F_ST = 0.11). Interpopulation genetic variation reflected isolation by distance and female-biased dispersal. Chinese elephants should be managed as 2 distinct units: Nangunhe and another combining Shangyong and MSJ; their long-term viability will require restoring gene flow between Shangyong and MSJ, and between elephants in China and neighboring countries. Our results have the potential to inform conservation planning for an iconic megafaunal species.     

短期围栏封育对高寒草甸植物群落及土壤理化性质的影响

充分了解围栏封育对植物群落及土壤理化性质的影响,是合理制定草甸恢复和利用相关政策、促进当地畜牧业经济可持续发展的关键。本研究通过对西藏三江并流区封育2年的高寒草甸的群落样方调查与土壤理化性质测定,结果表明,比较自由放牧,围封后地上和地下生物量分别提高26.24%和8.94%,但物种多样性指数略有下降;围封后土壤电导率、盐度和总溶解固体分别下降26.50%,29.03%,30.15%;围栏内地上生物量与土壤盐度、总溶解固体、电导率呈显著正相关(P<0.05),地下生物量与硝态氮、温度呈显著正相关(P<0.05);围栏外地下生物量与介电常数、体积含水率呈显著负相关(P<0.05),与铵态氮呈显著正相关(P<0.05)。研究结果表明,2年短期围封对高寒草甸植物群落及其土壤理化性质均有一定改善,可以作为应对草甸退化的干扰手段与管理措施。     

基于ISSR分子标记的紫云英种质资源遗传多样性及结构分析

紫云英(Astragalus sinicus L.)是我国重要的稻田绿肥作物,为揭示不同地区种质资源遗传特征,本研究利用ISSR分子标记对77份紫云英种质资源进行了遗传多样性及结构分析。研究发现,77份供试紫云英种质资源遗传相似性系数为0.567 7～0.868 6,可划分为6个聚类。不同地区群体间分化系数为0.136 3,达到极显著水平(P<0.001),其中种群间变异占比13.9%,种群内变异占比86.1%。不同ISSR引物扩增信息鉴别紫云英资源的能力存在差异,构建了以P809第3,8,9,10,12,15扩增位点以及P886第1,3,5,9,10,11扩增位点为特征的指纹图谱,能有效区分77份供试紫云英资源。以资源所在省份为单元进行群体分析,供试紫云英资源遗传结构呈现分化特征,河南-安徽-江苏-上海资源遗传结构相似,构成北方亚群;四川-广西-湖南-江西资源相似,构成西南方亚群;福建种质资源则呈现混合型特征。本研究结果可为我国紫云英种质资源的科学利用及新品种选育提供依据。     

右美托咪定抑制NOX4缓解急性应激致大鼠肾损伤

本研究拟探索右美托咪定（dexmedetomidine,DEX）对大鼠急性应激致肾损伤的保护作用,并从氧化应激的角度探索DEX对大鼠肾的保护通路。本研究使用了急性束缚应激模型,其中,大鼠被迫游泳15 min,并束缚3 h。本试验采用生化检测、组织病理学切片观察以评估肾功能,然后测定了氧化应激以及氧化应激的相关通路蛋白。旷场试验证实成功建立了急性应激模型。急性应激引起的肾损伤增加了NOX4,降低了Nrf2/HO-1/NQO1表达水平。DEX可降低NOX4表达,同时升高Nrf2/HO-1/NQO1的表达水平。DEX治疗组与急性应激组相比的肾生化结果明显恢复正常,病理切片观察损伤显著降低。试验结果表明,DEX治疗急性应激可影响NOX4/Nrf2/HO-1/NQO1信号通路,并抑制氧化应激。因此,DEX对急性应激引起的肾损伤具有保护作用,并在应激综合征中具有潜在的临床应用。     

Regression Applied in Continuous Mathematical Forecast

Data Mining can distill the connotative, unknown and potential value information or pattern from many data. It is a problem that must be settled during the developing of Decision Support System how to discover knowledge from Data Warehouse. In order to realize the continuous mathematical forecast that will be used in the Marketing Decision Support System for Industrial Enterprise, the authors use the method of regression to discuss how to design the algorithm for Data Mining, how to design the model for Data Mining, and so on. When the algorithm applied to the sales forecast, the result processing to simulation data indicates it can realize the expected effect.     

Reference Efficiency of Planetary Gear Train

Reference efficiency of a pair of gear drive without planetary motion is the base to calculate efficiency of the whole planetary gear train. The authors find reference efficiency for speed increase is higher than that of the same drive used for speed reduction. This is different from the earlier viewpoint. New equations are derived for correctly calculating the reference efficiency of spur gear and helical gear with internal conjugate and external conjugate either in the state of speed increase or speed reduction respectively. The equations are also used to calculate efficiency of 2K-H planetary gear train.     

Research on the Realization of Remote Controlling for Multi-output Power Supply of Communication Equipment Based on Ethernet

According to the requirements of communication power supply, such as high reliability, wide application and high availability, the authors raise a new remote control design for a multi-output power supply for communication equipment based on TCPIP protocol and embedded technology. Then the hardware design is given for the system is discussed based on Ethernet professional chip PRTL8019AS and related circuits. The firmware software design for the salve-computer based TCPIP technology, and the soft design of communieation and control for the master-computer are proposed based on Winsoek and MFC technology in detail A test machine is built to do the testing, the test results prove that the test machine works stably and well, satisfies the design rules and also provides a feasible method for the remote supervise in communication power supplier whose requirement includes remote supervise, no-people on duty and so on.

Expert System for Artificial Intelligence Blast Furnace Smelting Process

In order to upgrade traditional industry and realize the automation of furnace condition judgement and standardization of operation, an expert system for various furnace conditions and breakdown judgement and operation guidance on blast furnace are developed based on the theory of artificial intelligence and knowledge engineering. The system is fulfilled through C++Builder and combined with the actual production picture. The system has efficient and convenient man-machine interface by using menu operation method. The testing and operation of actual production data in simulation system indicate that the system is exact and reliable and can provide both the warning of various furnace breakdown and corresponding operation guidance.
