Identification of Differentially Expressed Genes and Lipid Metabolism Signaling Pathways between Muscle and Fat Tissues in Broiler Chickens

In this study, signaling pathways and key differentially expressed genes (DEGs) involved in lipid metabolism in muscle and fat tissues were investigated. Muscle and abdominal fat tissues were obtained from 35-day-old female broilers for RNA sequencing. DEGs between muscle and fat tissues were identified. Gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analyses of DEGs were performed. A total of 6130 DEGs were identified to be significantly enriched in 365 GO terms, most of which were involved in biological processes, cellular components, and molecular functions in muscle and fat tissues. Three important lipid signaling pathways (pyruvate metabolism, the insulin signaling pathway, and the adipocytokine signaling pathway) were identified among the fat and muscle tissues of broilers. The key common DEGs in these pathways included phosphoenolpyruvate carboxykinase 2 (PCK2), acetyl-CoA carboxylase 1 alpha and beta (ACACA and ACACB), and the mitogen-activated protein kinase (AMPK) gene family. Hence, our findings revealed the pathways and key genes and gene families involved in the regulation of fat deposition in the muscle and fat tissues of broilers.     

Effect of L-lysine Added in Diluent on the Fresh Semen Quality of Dorper Sheep

In order to explore the effect of different L-lysine levels added in semen dilution on the fresh Dorper sheep quality stored at liquid state,3 health Dorper ram were used to collect semen which were equal-packaged and added to solution containing different levels (0,0.1,0.2,0.3,0.4 and 0.5 g) L-lysine in 120 mL diluent and at 0,6,24,30,48,54,72 and 78 h,the sperm motility,membrane integrity and acrosome integrity were detected.The results showed that the sperm motility in groups added 0,0.1 and 0.2 g L-lysine were higher than other groups,and that in 0.4 and 0.5 g L-lysine groups decreased faster than other groups;The sperm motility in 0.5 g L-lysine group reduced to 0 at 30 h,and it reduced to 0 at 48 h in the group added 0.4 g L-lysine;The effective survival time and survival index in the group added 0.1 g L-lysine was higher than other groups (P< 0.05);The membrane integrity of 0.1 g group was the highest,that had no significant difference with control group from 0 to 54 h (P >0.05),while after 72 h the two groups had significant differences (P< 0.05).The membrane integrity in 0.4 and 0.5 g groups were the worst,and that was significant different with other groups (P< 0.05);The acrosome integrity of 0.1 g group was the highest which had no significant difference with control group (P >0.05),0.4 and 0.5 g groups were the worst,the difference between them was not significant (P >0.05),while was extremely significant difference with other groups except for 0 and 6 h (P< 0.01).The results suggested that dilution added a high concentration of L-lysine could inhibit sperm quality,when the count of L-lysine was more than 0.2 g,sperm quality was significantly decreased,adding 0.1 g L-lysine could improve Dorper sheep fresh sperm quality stored at liquid state.     

BPA不影响卵母细胞减数分裂相关基因Dazl的甲基化

为了探讨环境雌激素BPA对小鼠卵母细胞减数分裂相关基因Dazl甲基化的影响,本研究通过给孕鼠饮用含有BPA的水方式使胎鼠在发育过程中接触BPA,利用重亚硫酸盐测序法,分析了胎鼠生殖嵴卵母细胞不同发育时期Dazl甲基化水平的变化。结果显示:Dazl在减数分裂期间处于低甲基化水平,无论对照组或处理组均低于10%,说明Dazl的低甲基化对维持减数分裂的正常进行有重要作用;对照组与处理组的甲基化水平相当,差异不显著,说明本研究的BPA浓度不影响卵母细胞Dazl的甲基化水平。     

用生物合成的长链开环探针进行SNP分型

蝴蝶兰切花花被不同开放阶段酶活性的变化

以蝴蝶兰鲜切花为试材,对切花花被不同生理阶段3种酶活性的变化进行测量分析,以期为蝴蝶兰切花寿命的延长提供参考依据。结果表明:萼片和花瓣中的过氧化物酶(POD)从花苞期到半开期酶活性变化不明显,从半开期到衰老期酶活性显著升高。花瓣中的超氧化物歧化酶(SOD)从花苞期到半开期酶活性升高,从半开期到衰老期酶活性显著降低;萼片中的超氧化物歧化酶活性变化不显著但也呈先上升后下降趋势。花瓣中的中性蛋白酶(NP)从花苞期到半开期酶活性呈上升趋势,半开期到盛开期呈下降趋势,盛开期到衰老期呈上升趋势;花萼中的中性蛋白酶从花苞期到盛开期酶活性呈下降趋势,从盛开期到衰老期呈上升趋势;总体来看中性蛋白酶的活性呈先下降后上升趋势。     

TCDCA对IL-1β刺激下AA大鼠成纤维样滑膜细胞中PGE2分泌的影响

[目的] 进一步探讨牛磺鹅去氧胆酸（taurochenodeoxycholic acid,TCDCA）在抗炎免疫方面的潜在调节作用。[方法] 以AA大鼠成纤维样滑膜细胞作为研究对象,采用ELISA方法检测TCDCA和IL-1β作用下AA大鼠成纤维样滑膜细胞上清液中PGE₂的含量,分析TCDCA对IL-1β刺激下AA大鼠成纤维样滑膜细胞中PGE₂分泌情况的影响。[结果] TCDCA能够对IL-1β刺激下AA大鼠纤维样滑膜细胞PGE₂的分泌产生下调作用（P<0.05）。[结论] TCDCA对IL-1β刺激下AA大鼠成纤维样滑膜细胞中PGE₂的分泌具有抑制作用,为TCDCA在兽医临床应用提供依据。     

贵州省鸟类新纪录——栗树鸭

2020年4月14日,在贵州省贵阳阿哈湖国家湿地公园保育区内(106°39′0.13″E,26°30′28.46″N,1107 m)开展鸟类监测,于浅水沼泽区拍摄到2只雁鸭类,经鉴定为栗树鸭(Dendrocygna javanica)。通过查阅相关文献,该种在贵州省的分布尚未记录过,属贵州省鸟类新纪录。本文对发现地概况、形态特征、生境特征和分布现状以及对该物种的保护进行了简要讨论。     

Energy metabolism and apoptotic effect of microwave radiation on rat myocardial cells

AIM: To investigate the effect of microwave radiation at different intensities on the rat myocardium and its possible mechanism.METHODS: The rats were radiated by the intensity of 500, 1 000, 1 500 and 2 000 W/m² with 2 450 MHz microwave for 6 min. The heart tissue was collected 6 h after microwave radiation. ATP and mitochondria complex Ⅳ and Ⅴ were measured. The changes of the tissue structures were observed under transmission electron microscope. The apoptosis of the myocardial cells was detected by a cell analyzer. The protein level of cleaved caspase-3 was determined by Western blotting.RESULTS: The concentration of ATP and activity of mitochondria complex Ⅳand Ⅴ signi-ficantly decreased compared with control group in the cardiac tissues. The decreased number, morphological abnormalities such as dissolved cavitation, matrix and obvious tumefaction of mitochondria were observed under transmission electron microscope. The microwave radiation induced the apoptosis of myocardial cells in the rats. The cell apoptotic rate and the protein level of cleaved caspase-3 increased with increasing intensity of microwave radiation(P<0.05).CONCLUSION: Microwave radiation has obvious injury effect on the rat heart, which can cause cardiac energy metabolism dysregulation and cardiac myocyte apoptosis.     

猪用加味玉屏风缓释微丸水提醇沉工艺研究

目的:考察猪用加味玉屏风缓释微丸的水提醇沉工艺。方法:以黄芪甲苷的含量及干膏得率作为评价指标,选用L9(34)进行正交试验。结果:猪用加味玉屏风缓释微丸最佳水提取工艺为药材分别加12倍量水,提取2次,提取时间分别为0.5 h;最佳醇沉工艺为浓缩药液相对密度1.15,加醇浓度70%,静置时间为12 h。结论:优选出的水提醇沉工艺科学、合理。
