The survival of Leptosphaeria maculans , which causes phoma stem canker (blackleg), on oilseed rape residues ( Brassica napus ) in South Australia was investigated. Using a quantitative polymerase chain reaction (PCR) assay for L. maculans DNA, the pathogen was mainly detected in the upper 5 cm of the soil profile, including residues on the soil surface. As the size of organic matter particles in the soil decreased, so did the quantity of L. maculans detected in them. To obtain representative data for a field, at least 30 subsamples needed to be collected over the 0·81 ha area studied. In a survey of 49 commercial fields in South Australia, most L. maculans was detected in fields 1 year after oilseed rape had been grown, with less detected after 2 years and negligible amounts 3 years or more after cropping. The diagnostic DNA-based assay for L. maculans reduced the time and cost of studying L. maculans survival in soil and increased the sensitivity and accuracy of results compared with estimates of propagule number of colony-forming units on a semiselective medium.  相似文献

Variable-rate fertilizer application requires knowledge of the spatial distribution of soil nutrients within fields. Grid soil sampling might be used for acquiring this information, but is often too expensive for resolving spatial patterns in soil nutrients at the scale of precision fertilizer application. The objective of this study was to determine whether grid sampling efficiency can be improved using cokriging estimates with slope gradient as a secondary variable, which is easily obtained from high-resolution digital elevation models. Soils in two northern Montana wheat fields were sampled at the nodes of a 100-m diagonal grid. Soil test phosphorus and potassium maps were constructed with kriging and cokriging. Co-kriging uses the spatial correlation between two variables to predict for the less intensively sampled variable of interest, often with less estimation error than a univariate method such as kriging. The average estimation variance for cokriging compared to kriging was reduced for all values of the correlation considered. The additional complexity of cokriging might be justified provided a secondary variable exists that is spatially cross correlated with the primary variable of interest.  相似文献

【目的】确定雀嘴茶中6′-O-咖啡酰熊果苷(CA)超声辅助提取的最佳工艺。【方法】采用超声辅助法提取雀嘴茶中的CA,通过单因素试验考察甲醇体积分数(50%,60%,70%,80%,90%)、液(mL)料(g)比(8∶1,10∶1,12∶1,14∶1,16∶1)、超声时间(10,15,20,25,30min)、提取次数(1,2,3,4,5次)对CA提取率的影响,并利用响应面法优化CA的超声辅助提取工艺。【结果】超声辅助法提取CA的理论最佳工艺条件为:甲醇体积分数72.87%、液料比15.87∶1、超声时间18.96min、提取次数3.63次,CA提取率的理论预测值可达到23.12%。根据实际操作的需要,将提取工艺条件修正为甲醇体积分数73%、液料比16∶1、超声时间19min、提取次数4次,在此条件下CA实际提取率为23.03%,与理论值较为接近。【结论】采用响应面法优化雀嘴茶中CA超声辅助提取的最佳工艺完全可行。  相似文献

Data on the extent and dynamics of the viral diseases affecting cereal crops in breeding nurseries have been obtained as a result of the phytovirological monitoring. The deterious pathogens affecting the spring barley and the spring and winter wheat are identified under the conditions of the region. New strategies to enhance growth and productivity of food crops in saline soils represent important research priorities. This study has investigated the role of certain priming techniques to induce salt tolerance of bread wheat. Wheat grains were soaked in 0.2 mm sodium nitroprusside as nitric oxide donor (redox priming), diluted sea water (halopriming) and the combination of both (redox halopriming). Grains were also soaked in distilled water (hydropriming); in addition, untreated grains were taken as control. Our results indicated that priming treatments significantly improved all growth traits and increased leaf pigments concentration as compared to the control. Priming treatments markedly enhanced membrane stability index, proline, total soluble sugars and K+ concentration with simultaneous decrease in the concentration of Na+ and malondialdehyde (MDA). Furthermore, yield and yield‐related traits such as plant height, spike length, total number of tillers, 1000‐grain weight, straw and grain yield considerably affected by priming treatments. Moreover, the grain yield of both genotypes was positively affected by redox halopriming treatment. However, the extent of enhancement was more prominent in Gemmiza‐9 (salt sensitive) than that in Sakha‐93 (salt‐tolerant). Overall, this study clearly indicated that redox halopriming treatment is a promising and handy technique to induce salinity tolerance of wheat genotypes.  相似文献

The expected temperature rise in late summer/early autumn can change the conditions for acclimation and affect the winter survival of perennial crops. This study examined the effect of the temperature just before the onset of cold acclimation (pre‐acclimation) on freezing tolerance of timothy (Phleum pratense L.), perennial ryegrass (Lolium perenne L.) and red clover (Trifolium pratense L.) populations (both cultivars and breeding populations) adapted to either northern or southern parts of Norway. Using phytotron experiments, we studied whether increasing pre‐acclimation temperature delays growth cessation, affects photoacclimation and reduces freezing tolerance. Furthermore, we assessed whether these effects were related to the latitudinal adaptation of the plant material. The results showed that a rise in pre‐acclimation temperature decreased both cold acclimation capacity and photoacclimation in these species. This affected the freezing tolerance, which was reduced significantly more in northern‐adapted population of timothy and perennial ryegrass compared with southern‐adapted populations. Red clover was less affected by temperature changes than the grasses.  相似文献

Half‐sib progenies of congo signal grass (Brachiaria ruziziensis Germain & Evrard, syn.: B. eminii, Urochloa ruziziensis) were analysed using a mixed model methodology. The objective was to estimate genetic and phenotypic parameters, predict individual genetic values, estimate correlations between characters associated with biomass production and forage nutritive value and use this information to determine the economic weights to compare three multiplicative selection indices. Individual narrow‐sense heritabilities corrected for inbreeding varied from 0.14 to 0.91 for characters associated with biomass production and from 0.04 a 0.24 for nutritional value characters. The correlations among characters associated with annual biomass production were of high magnitude. In contrast, biomass production characters were negatively correlated with crude protein, acid detergent fibre and lignin. Total dry matter yield, crude protein and regrowth ability were weighted separately in evaluations during wet and dry season or annually in selection indices. Index that considered characters annually resulted in better distribution of forage production along the year than indices that separated wet and dry season production.  相似文献

The present study assesses the effects of 5‐aminolevulinic acid (ALA, 0, 0.1, 1 and 10 mg l?1) on the growth of oilseed rape (Brassica napus L. cv. ZS758) seedlings under water‐deficit stress induced by polyethylene glycol (PEG 6000, 0 and ?0.3 MPa). Water‐deficit stress imposed negative effects on seedling growth by reducing shoot biomass, cotyledon water potential, chlorophyll content and non‐enzymatic antioxidants (glutathione and ascorbic acid) levels. On the other hand, water‐deficit stress enhanced the malondialdehyde (MDA) content, reactive oxygen species (ROS) production, enzymatic antioxidants activities, reduced/oxidized glutathione ratio (GSH/GSSG) and reduced/oxidized ascorbic acid (ASA/DHA) ratio in seedlings. Application of ALA at lower dosages (0.1 and 1 mg l?1) improved shoot weight and chlorophyll contents, and decreased MDA in rape seedlings, whereas moderately higher dosage of ALA (10 mg l?1) hampered the growth. The study also indicated that 1 mg l?1 ALA improved chlorophyll content, but reduced MDA content and ROS production significantly under water‐deficit stress. Lower dosages of ALA (0.1 and 1 mg l?1) also enhanced GSH/GSSG and ASA/DHA as compared to the seedlings under water‐deficit stress. The antioxidant enzymes (ascorbate peroxidase, peroxidase, catalase, glutathione reductase and superoxide dismutase) enhanced their activities remarkably with 1 mg l?1 ALA treatment under water‐deficit stress. It was also revealed that 1 mg l?1 ALA treatment alone induced the expression of APX, CAT and GR substantially and under water‐deficit stress conditions ALA treatment could induce the expression of POD, CAT and GR to a certain degree. These results indicated that 0.1–1 mg l?1 ALA could enhance the water‐deficit stress tolerance of oilseed seedlings through improving the biomass accumulation, maintaining a relative high ratio of GSH/GSSG and ASA/DHA, enhancing the activities of the specific antioxidant enzymes and inducing the expression of the specific antioxidant enzyme genes.  相似文献
