Caloric restriction induces body mass loss that is often regained when restriction ends. This study aimed to determine if dietary energy density modulates the extent of post-restriction body mass regain. Water (20% wt:wt) was added to a standard dry commercially available feline diet. Twenty-seven domestic short-haired cats underwent a 20% caloric restriction on this diet. Following restriction, cats were offered the same dry diet ad libitum either without additional water or with 40% added water, therefore maintaining macronutrient composition whilst manipulating energy density. Despite no significant difference in energy intake during ad libitum consumption, post-restriction body mass regain was greater on the high energy dense (0% hydrated), compared to the low energy dense (40% hydrated) diet. The same protocol was repeated with a separate cohort of 19 cats with additional measures of physical activity, gut transit time and energy digestibility. Activity levels on the low energy dense diet were significantly higher than in cats on the high energy dense diet (p=0.030) and were similar to those recorded during caloric restriction. These results suggest that body mass gain following caloric restriction is ameliorated, and physical activity enhanced, by feeding a diet which is low in energy density due to the addition of 40% water. 相似文献

Purpose
The oxidation of ammonium (NH4+) to nitrate (NO3−) in the soil is an important biogeochemical process, which has major environmental implications as it can contribute to NO3− leaching and nitrous oxide (N2O) emissions. Nitrification inhibitors have been used to slow down this process to reduce NO3− leaching and N2O emissions from agricultural land. Methyl ester distribution in pectin homogalacturonan has a major influence on functionality. Enzymatic engineering of the pectin nanostructure for tailoring functionality can expand the role of pectin as a food-formulating agent and the use of in situ modification in prepared foods. We report on the mode of action of a unique citrus thermally tolerant pectin methylesterase (TT-PME) and the nanostructural modifications that it produces. The enzyme was used to produce a controlled demethylesterification series from a model homogalacturonan. Oligogalacturonides released from the resulting demethylesterified blocks introduced by TT-PME using a limited endopolygalacturonase digestion were separated and quantified by high-pressure anion-exchange chromatography (HPAEC) coupled to an evaporative light-scattering detector (ELSD). The results were consistent with the predictions of a numerical simulation, which assumed a multiple-attack mechanism and a degree of processivity ~10, at both pH 4.5 and 7.5. The average demethylesterified block size (0.6-2.8 nm) and number of average-sized blocks per molecule (0.8-1.9) differed, depending upon pH of the enzyme treatment. The mode of action of this enzyme and consequent nanostructural modifications of pectin differ from a previously characterized citrus salt-independent pectin methylesterase (SI-PME). 相似文献

The rapid quantification of flavonoid compounds in onions by attenuated total reflectance (ATR) Fourier transform infrared (FT-IR) spectroscopy combined with multivariate analysis was evaluated as a possible alternative to high-performance liquid chromatography (HPLC) analysis. Quercetin content in onion varieties (yellow, red, and sweet) was quantified using ATR FT-IR (4000 to 400 cm?1) spectroscopy and HPLC methods. Quercetin-3,4'-O-diglucoside (3,4'-Qdg) and quercetin-4'-O-glucoside (4'-Qmg) comprised >80% of the total flavonol content detected in the studied varieties. The quercetin compounds (3,4'-Qdg and 4'-Qmg) and total flavonol conjugates were quantified by HPLC, and results correlated closely with ATR-IR values (R > 0.95). Cross-validated (leave-one-out) partial least-squares regression (PLSR) models successfully predicted concentrations of these quercetins. The standard errors of cross-validation (SECV) of 3,4'-Qdg and 4'-Qmg, total quercetin, and total flavonol contents of onions were 20.43, 21.18, and 21.02 mg/kg fresh weight, respectively. In addition, supervised and unsupervised segregation analyses (principal component analysis, discriminant function analysis, and soft independent modeling of class analogue) were performed to classify onion varieties on the basis of unique infrared spectral features. There was a high degree of segregation (interclass distances > 3.0) for the different types of onion. This study indicated that the IR technique could predict 3,4'-Qdg, 4'-Qmg, total quercetin, and total flavonol contents and has advantages over the traditional HPLC method in providing a valid, efficient, and cost-effective method requiring less sample preparation for the quantification of quercetins in onion. 相似文献

This study investigated soil translocation associated with weeding on steeply sloped swidden fields attended by ethnic Da Bac Tay farmers in Hoa Binh Province in northern Vietnam. Annual soil loss rates of 4–6 Mg ha−1 year−1 were found on 20 m plots located on two separate hillslopes. Median soil flux rates were equivalent to 2.6–3.9 kg m−1 pass−1 for experiments conducted on slopes ranging from 0.54 to 0.84 m m−1. The primary soil translocation process, the mechanical movement of soil via contact with a small hoe (ngheo), contributed approximately 60% of the weeding-related soil flux. Ravel, which is the rolling, bouncing, and sliding of soil clods downslope, was a secondary translocation process that accounted for almost 40% of the soil flux. Soil flux was most appropriately described with an exponential function that could predict the occurrence of ravel on steeper slopes. The observed soil fluxes were much smaller than those determined during weeding and hoeing at other tropical and subtropical sites, primarily because the tillage depth was very shallow (<1 cm) and weed density was low at the time of experimentation. The erosion rates associated with weeding were an order of magnitude lower than reported water erosion rates; therefore, the contribution to landscape change was believed to be small. Combined water and tillage erosion estimates indicated a possible unsustainable increase in soil loss on some steep-slope fields within the last few decades that has resulted from shorter fallow periods, longer periods of cultivation before fallowing, and greater weed pressure. To evaluate the prevalence of methicillin-resistant Staphylococcus aureus (MRSA) in the pig population in Ireland, nasal swabbing was employed in three abattoirs to screen 440 pigs from 41 geographically distributed farms. One hundred individuals involved in the pig industry were also nasally screened. No MRSA isolates were recovered from the pigs and only two of the humans tested were identified as MRSA carriers. Importantly, MRSA was not obtained from pig producers, veterinarians or abattoir employees, but was isolated from individuals working in the wider pig industry. Multi-locus sequence typing revealed that these isolates belonged to sequence types (ST) ST22 and ST1307; the latter is a previously unreported single locus variant of ST5. Five dust samples from each of the three slaughterhouses were culture-negative for MRSA. These results indicate that porcine colonisation by MRSA, and in particular the animal-related strain MRSA-ST398, was not common in Ireland during the period of study. 相似文献

Both interleukin-1 (IL-1) and platelet-derived growth factor (PDGF) induce proliferation of cultured fibroblasts and smooth muscle cells. These polypeptide mediators are released by activated macrophages and other cell types in response to injury and are thought to have a role in tissue remodeling and a number of pathologic processes. Analysis of the kinetics of [3H]thymidine incorporation by cultured fibroblasts demonstrated that the response to IL-1 is delayed approximately 8 hours relative to their response to PDGF. IL-1 transiently stimulated expression of the PDGF A-chain gene, with maximum induction after approximately 2 hours. Subsequent synthesis and release of PDGF activity into the medium was detected as early as 4 hours after IL-1 stimulation, and downregulation of the binding site for the PDGF-AA isoform of PDGF followed PDGF-AA secretion. Antibodies to PDGF completely block the mitogenic response to IL-1. Therefore, the mitogenic activity of IL-1 for fibroblasts and smooth muscle cells appears to be indirect and mediated by induction of the PDGF A-chain gene. 相似文献
