Jatropha curcas, a multipurpose shrub has acquired significant economic potential as biodiesel plant. The seeds or pressed cake is toxic due to the presence of toxic substances and is not useful as food/fodder despite having the best protein composition. A simple, efficient, and reproducible method for plant regeneration through direct organogenesis from petiole explants of non-toxic J. curcas was developed using Murashige and Skoog (MS) medium supplemented with different concentrations of thidiazuron (TDZ). The best induction of shoot buds (57.61%), and number of shoot buds (4.98) per explant were obtained when in vitro petiole explants were placed horizontally on MS medium supplemented with 2.27 μM TDZ. The Induced shoot buds were transferred to MS medium containing 10 μM kinetin (Kn), 4.5 μM 6-benzyl aminopurine (BA), and 5.5 μM α-naphthaleneacetic acid (NAA) for shoot proliferation and subsequent elongation was achieved on MS medium supplemented with 2.25 μM BA and 8.5 μM IAA. The elongated shoots could be rooted on half-strength MS medium with 15 μM IBA, 11.4 μM IAA and 5.5 μM NAA with more than 90% survival rate. 相似文献

The main objectives of the “Oryza Map Alignment Project” (OMAP) are to characterize the rice genome from a comparative standpoint by establishing a genus-wide and genome-scale comparative framework from representative species. Here, we report our progress in the analyses of these datasets and emerging “comparative phylogenomics” insights into Oryza evolution at two different resolutions—chromosomal and sequence levels. We demonstrate the abundance and impact of structural variations (SV) on genome diversity using African Oryza as a model. The molecular basis of SV was inferred using three genus-wide vertical sequence datasets. Combined, these data demonstrate that a single reference genome sequence for the genus Oryza is insufficient to comprehensively capture the genomic and allelic diversity present within the genus. Towards this end, we present a strategy to generate high-quality and cost-effective de novo reference sequences of collective Oryza. The application and broader scientific impact of the OMAP resources under an international cooperative effort (I-OMAP) are discussed. 相似文献

Structural modification of photosynthetic reaction centers is an important approach for understanding their charge-separation processes. An unprecedented persistent structural transformation of the special pair (dimer) of bacteriochlorophyll molecules can be produced by light absorption alone. The nonphotochemical hole-burned spectra for the reaction center of Rhodopseudomonas viridis show that the phototransformation leads to a red shift of 150 wave numbers for the special pair's lowest energy absorption band, P960, and a comparable blue shift for a state at 850 nanometers, which can now be definitively assigned as being most closely associated with the upper dimer component. The nucleotide sequence of the region of Gardner-Rasheed feline sarcoma virus (GR-FeSV) encoding its primary translation product, p70gag-fgr, has been determined. From the nucleotide sequence, the amino acid sequence of this transforming protein was deduced. Computer analysis indicates that a portion of P70gag-fgr has extensive amino acid sequence homology with actin, a eukaryotic cytoskeletal protein. A second region of P70gag-fgr is closely related to the tyrosine-specific kinase gene family. Thus, the v-fgr oncogene appears to have arisen as a result of recombinational events involving two distinct cellular genes, one coding for a structural protein and the other for a protein kinase. 相似文献

A method for rearing the southern green stinkbug, (Nezara viridula L.) (Heteroptera: Pentatomidae), using a modified lygus semi-solid artificial diet was developed. First to second-instar nymph were reared in a density of 631.5 ± 125.05 eggs per Petri-dish (4 cm deep × 15 cm diam). Second instar to adult were reared in a density of 535.0 ± 112.46 s instar nymphs per rearing cage (43 × 28 × 9 cm). Mating and oviposition occurred in popup rearing cages (30 × 30 cm), each holding 60–90 mixed sex adults of similar age. Adults emerged 35.88 ± 2.13 d after oviposition and survived for an average of 43.09 ± 9.53 d. On average, adults laid 223.95 ± 69.88 eggs in their lifetime, for a total production of 8,099 ± 1,277 fertile eggs/oviposition cage. Egg fertility was 77.93% ± 16.28. Egg masses held in petri-dishes had a total hatchability of 79.38% ± 20.03. Mortality of early nymphs in petri-dishes was 0.64% ± 0.12 for the first instar and 1.37% ± 0.45 for second instar. Late nymphal mortality in rearing cages was 1.41% ± 0.10, 3.47% ± 1.27, and 4.72% ± 1.29 for the third, fourth, and fifth instars, respectively. Survivorship from nymphs to adults was 88.48% ± 2.76. Using artificial diet for rearing N. viridula could reduce cost by avoiding time-consuming issues with daily feeding fresh natural hosts and insect manipulation. It could increase reliability and simplicity of bug production, which should facilitate mass rearing of its biological control agents. 相似文献
