翠绿2号大顶苦瓜的选育

翠绿 2号是用早熟、丰产、优质的强雌性株系B0 7 2 1作母本与品质好、抗逆性强的株系D12 3 1作父本配制而成的大顶苦瓜一代杂种。该品种比对照翠绿大顶苦瓜早熟 2d(天 ) ,前期产量增产 13 .0 % ,总产量增产3 4 .8% ,一般每 667m2 产量 2 0 0 0kg ,适宜华南地区春、秋季栽培。至 2 0 0 1年累计推广种植面积 2 0 0 0hm2 。     

Changes of L-selectin expression on peripheral blood polymorphonuclear leukocytes and their significance in rats with acute lung injury

AIM:To explore the changes of L-selectin expression on peripheral blood polymorphonuclear leukocytes (PMNs) and their significance in rats with acute lung injury (ALI). METHODS:ALI model in rat was established by intravonous injection of E. coli endotoxin (ET). The expression of L-selectin on peripheral blood PMNs was measured by immunofluorescence and flow cytometry.RESULTS:The contribution of L-selectin fluorescence signal was on the surface of PMNs membrane. The expression of L-selectin on poeripheral blood PMN was significantly lower at 5 min after injection of ET and the lowest during 15 min to 30 min, then gradually increased, but the expression of L-selectin on PMN was lower at 60 min after injection of ET than that of control animal.CONCLUSION:In physiological state, L-selectin were expressed on the surface of PMN membrane. The protein expression of L-selectin on PMNs reduced rapidly after injection of ET and the lowest at 15 min, then gradually increased. L-selectin may play a role in the development of ALI.     

Effects of external counterpulsation on renin-angiotensin system and hemodynamics in dogs with myocardial ischemia

AIM:To examine the effect of external counterpulsation(ECP) on renin-angiotensin system(RAS) and hemodynamics in dogs with myocardial ischemia and their relationship. METHODS:Acute myocardial ischemia was induced by occluding the left anterior descending of the dogs. The local renin activity, angiotensin Ⅱ(AngⅡ) level, angiotension-converting enzyme(ACE) activity were tested by biochemical methods and hemodynamics was recorded by a 8-channel physiological recorder.RESULTS:Ischemia could actiivate renin,ACE and AngⅡ in cardiovasculature and ECP reduced them except for renin in ischemic myocardium. Ischemia also could activate RAS in lung and kidney, which play an important role on circulating RAS, and ECP reduced them. Furthermore, ECP could improve hemodynamics and there existed a close relationship between local AngⅡlevel and hemodynamics. CONCLUSION:ECP can reduce local RAS and improve hemodynamics in dogs with myocardial ischemia, which might be one of mechanisms underlying the protective effect of ECP on ischemic myocardium.     

甲基对硫磷人工抗原的合成与鉴定

根据甲基对硫磷的结构特征,分别从甲基对硫磷的硝基一端或磷酸酯基甲氧基一端引入一定长度的间隔臂和活性基团,设计并合成了4种结构的半抗原;针对各种半抗原所带活性基团的性质,通过多种方法制备了与不同的载体蛋白偶联的人工抗原。进行动物免疫后,得到了针对甲基对硫磷的高效价抗体。     

免疫亲合色谱及在农药残留分析中的应用

免疫亲合色谱可用于复杂样本中微量目标分析物的选择性分离富集。综述了免疫亲合色谱的基本原理、方法的建立过程(包括抗体的制备、固相基质的筛选、抗体与基质的偶联、免疫亲合色谱条件的优化)、免疫亲合色谱在农药残留分析中的应用现状及发展趋势。     

重组鸡IL-2增强鸡四联灭活苗免疫效果的研究

使用在原核表达系统中表达的重组鸡白细胞介素 2 (IL 2 )蛋白 ,经过初步的分离纯化 ,与鸡新城疫 禽流感 法氏囊 传染性支气管炎四联油乳剂灭活苗同时使用 ,检测其对疫苗的免疫增强效果。试验共分 5组 ,每组 1 0只鸡。其中对照组只注射四联油乳剂灭活苗 ;4个试验组 ,在注射疫苗的同时分别注射重组鸡IL 2蛋白 0 0 5mg、 0 0 1mg、 0 1 5mg、 0 2 0mg。试验组与对照组同时各注射新城疫等多联油苗 0 5mL。采用HA、HI试验测定鸡新城疫、禽流感抗体滴度。结果表明 :重组鸡IL 2对该四联油乳剂灭活苗有较强的免疫增强作用。以第 3、 4组的增强效果较为明显 ,其中对鸡新城疫病毒和禽流感抗原的抗体效价 ,与对照组相比分别提高 2 42 8和 2 2 3 0个滴度。重组鸡IL 2在使用的过程中 ,没有发现任何毒副作用 ,证明是一种安全高效的免疫增强剂。     

Comparison of different conditions inducing embryonic stem cells

AIM: To evaluate the different conditions inducing mouse embryonic stem cells (ESC) in vitro to differentiate into cardiomyocytes. METHODS: BRL conditioned medium was used to promote the growth of ESC and maintain them in an undifferentiated state. During the inducing process, retinoic acid (RA), DMSO, activin-A and TGF-β₁ were used as inducing reagents, and made up six kinds of differentiating medium. Then a three-step method inducing ESC cultured in hanging drops, in suspension and in plating was used to induce the differentiation of ESC. RESULTS: ESC were induced in vitro to differentiate into cardiomyocytes. Of all groups, the highest differentiating rate was observed in the group induced by activin-A (20 μg/L) and TGF-β₁ (2 μg/L). CONCLUSION: The inducing conditions including activin-A (20 μg/L) and TGF-β₁ (2 μg/L) is very valuable in inducing ESC differentiation into cardiomyocytes.     

噻二唑基-3-哒嗪酮类化合物的合成及生物活性

将取代的二酰基肼环合后,得到中间体2-芳基-5-氯甲基-1,3,4-噻二唑,然后与2-叔丁基-4-氯-5-羟基-3(2H)-哒嗪酮反应,合成了8个未见文献报道的含噻二唑基哒嗪酮类化合物,其化学结构经1H NMR、高分辩质谱和元素分析确认。生物活性测试结果表明,部分化合物对粘虫P.separate W.有较好的抑制生长活性,其中化合物 3b 的EC50值为21 mg/L。     

Bone marrow mesenchymal stem cells are induced to differentiate into cardiomyocytes by hepatocyte growth factor in vitro

AIM: To explore a new method of hepatocyte growth factor (HGF) inducing bone marrow mesenchymal stem cells (MSC) to differentiate into cardiomyocytes. METHODS: Bone marrow MSC was cultured with DMEM media (10% fetal calf serum) 4-6 passages, and induced by HGF (10 μg/L) for 30 d. Automatical beating of the differentiated cells was observed daily with transverse microscopy, or under condition of 0.1% isoproterenol or cal-cium-deprived incubation. Specific cardiac myosin in the cells was indentified by immunochemistry. RESULTS: At 14-20 d of differentiation, bone marrow mesenchymal stem cells formed clones, in 10%-50% of which spontaneous beating cell-mass had come to continuously exist. Isoproterenol increased the beating rate and calcium-deprived media inhibited the beating. The cells were identified to be cardiomyocytes by expression of cardiac myosin heavy chain. CONCLUSION: HGF may induce bone marrow mesenchymal stem cells into cardiomyocytes with high efficiency, but the differentiating pathway of stem cells remains to be further studied.
