Treatment with alkali, particularly overliming, has been widely used as a method for the detoxification of lignocellulose hydrolysates prior to ethanolic fermentation. However, the mechanisms behind the detoxification effect and the influence of the choice of cation have not been well understood. In this study, a dilute acid hydrolysate of spruce and an inhibitor cocktail consisting of six known inhibitors were used to investigate different alkali detoxification methods. The various treatments included the addition of calcium hydroxide, sodium hydroxide, potassium hydroxide, and ammonia to pH 10.0 and subsequent adjustment of the pH to 5.5 with either sulfuric or hydrochloric acid as well as treatment with the corresponding amounts of calcium, sodium, and potassium as sulfate or chloride salts at pH 5.5. An RP-HPLC method was developed for the separation of 18 different inhibitors in the hydrolysate, including furaldehydes and phenolics. Detection and quantification were carried out by means of UV, DAD, and ESI-MS in negative mode. Treatment of the spruce hydrolysate with alkali resulted in up to approximately 40% decrease in the concentration of furaldehydes. The effects on the aromatic compounds were complex. Furthermore, SFE was performed on the precipitate formed during alkali treatment to evaluate the inhibitor content of the precipitate, and the following RP-HPLC analysis implied that potential inhibitors were removed mainly through conversion rather than through filtration of precipitate. Parallel experiments in which sulfuric acid or hydrochloric acid was used for acidification to pH 5.5 after alkali treatment indicated that the choice of anion did not affect the removal of inhibitors. Detoxification with calcium hydroxide and ammonia resulted in better fermentability using Saccharomyces cerevisiae than detoxification with sodium hydroxide. The results from the experiments with the inhibitor cocktail indicated that the positive effects of alkali treatment are difficult to explain by removal of the inhibitors only and that possible stimulatory effects on the fermenting organism warrant further attention.  相似文献

Abstract – Habitat use and diet of Arctic charr (Salvelinus alpinus) coexisting with European whitefish (Coregonus lavaretus) and grayling (Thymallus thymallus) were studied in one deep and two relatively shallow subarctic lakes in northern Norway. Stomach content and stable isotope analyses revealed clear and temporally stable resource partitioning between the species in all three lakes. Arctic charr had a wide and flexible trophic niche and was the only piscivorous species. In contrast, whitefish and grayling had remarkably stable planktivorous and benthivorous niches, respectively. In the deepest lake, Arctic charr together with grayling mainly utilised littoral benthos, while piscivory was more prevalent in Arctic charr in the two shallower lakes. In one of the shallow lakes, whitefish was apparently relegated to the inferior profundal niche because of dominance of the littoral by grayling. Our results suggest that Arctic charr may not necessarily need an extensive profundal zone as a refuge, but can coexist with whitefish if a third competing fish species like grayling occurs in the littoral habitat or if profitable small prey fish are available. The study demonstrates that strong dietary plasticity of Arctic charr is instrumental in the observed coexistence with the commonly competitively superior whitefish.  相似文献

This study investigates the effect of different smolt production strategies on vertebral morphology (radiology), composition (mineral content) and mechanical strength (load-deformation testing) in Atlantic salmon (Salmo salar). Rapid-growing underyearling (0+) smolt were compared with slower-growing yearling (1+) smolt and a reference group of wild smolt (w). The underyearling and yearling smolt were transferred to seawater in October 2002 and May 2003, respectively. The underyearling smolt were reared under continuous light and the yearling smolt under natural light during the first twelve weeks in seawater, at ambient temperatures. Thus, the underyearling smolt hit seawater at 13 °C and were reared at 10-13 °C during the early seawater phase, whereas the yearling smolt hit seawater at 7 °C and were reared at 7-10 °C during the early seawater phase. All groups displayed increased longitudinal growth (up to 9% increase in relative length) of the caudal vertebrae during parr-smolt transformation. However, at transfer to seawater, the underyearling smolt had significantly lower vertebral mineral content (0+ 44%, 1+ 47%, w 50%) and higher incidence of deformed vertebrae (0+ 1.5%, 1+ 0%, w 0%), and at twelve weeks after transfer to seawater significantly lower vertebral mineral content (0+ 36%, 1+ 41%, w 43%), yield-load (0+ 6492 g, 1+ 8797 g, w 9150 g) and stiffness (0+ 7578 g/mm, 1+ 15,161 g/mm, w 20,523 g/mm), and significantly higher incidence of deformed vertebrae (0+ 2.5%, 1+ 0.3%, w 0%). There was a significant correlation between the mineral content and mechanical properties of the vertebrae. The underyearling smolt had significantly elevated plasma concentrations of total Ca, and P and Ca2+ during the parr-smolt transformation and in the early seawater phase.The results show that underyearling smolt may have an increased risk of developing vertebral deformities. It is possible that this risk can be reduced by postponing the start of the short-day treatment. This will reduce the temperature during smoltification, the temperature and daylength during the early seawater phase, and increase the age at smoltification.  相似文献

Alkylresorcinols as markers of whole grain wheat and rye in cereal products   总被引:1,自引:0,他引:1  
The alkylresorcinol (AR) content of Swedish wheat grain samples, as well as of cereal ingredients and cereal foods containing wheat and rye, was determined. The average total AR content in Swedish wheat was 412 microg/g (ranging between 227 and 639 microg/g), which is lower than that in Swedish rye analyzed in a previous study. The relative composition of AR homologues was consistent for wheat samples and differed markedly from that of rye. Notably, the ratio of the homologues C17:0/C21:0 was approximately 0.1 in wheat and approximately 1.0 in rye, indicating that it can be used to distinguish between those two cereals. The AR content in cereal foods commonly consumed in Sweden varied widely, from nondetectable levels in white wheat flour and products not containing the outer parts of wheat and/or rye to >900 microg/g in some whole grain rye products. AR content in cereal foods was calculated from their recipes using average AR values for the cereal ingredients determined in this study. As there was a good correlation between calculated and analyzed AR levels in cereal foods (R2 = 0.91), it is possible to estimate the proportion of whole grain wheat and/or rye in a given cereal product on the basis of AR content and C17:0/C21:0 ratio. ARs appear to be good markers of whole grain wheat and rye in foods, and their analysis may be an objective way to identify foods rich in whole grain wheat and/or rye or brans thereof.  相似文献

Nineteen apparent flavonoids were determined by HPLC-DAD in foliage of a chemotype (G-type) of Barbarea vulgaris , and four were isolated. Two were novel tetraglycosylated flavonols with identical glycosylation patterns, kaempferol 3-O-(2,6-di-O-β-d-glucopyranosyl)-β-d-glucopyranoside-7-O-α-l-rhamnopyranoside (1) and quercetin 3-O-(2,6-di-O-β-d-glucopyranosyl)-β-d-glucopyranoside-7-O-α-l-rhamnopyranoside (2). The identification of d/l configuration was tentatively based on susceptibility to α-l-rhamnosidase and β-d-glucosidases. A characteristic feature of 1 and 2 was appreciable water solubility, an expected consequence of the extensive glycosylation. A less complex pair of flavonols comprised 3-O-β-d-glucopyranoside-7-O-α-l-rhamnopyranosides of kaempferol and quercetin. Two natural chemotypes of B. vulgaris differed in levels of 1 and 2, with the P-type deficient in 1 and 2 and the insect-resistant G-type rich in 1 (ca. 3-4 μmol/g dry wt) and with moderate levels of 2 (ca. 0.3-0.8 μmol/g dry wt). However, there was only modest seasonal variation in flavonols 1 and 2, in contrast to a strong seasonal variation in insect resistance.  相似文献

Application of water saving irrigation strategies in agriculture has become increasingly important. Both modelling and experimental work are needed to gain more insights into the biological and physical mechanisms in the soil-plant system, which regulates water flow in the system and plays a central role in reducing crop transpiration. This paper presented a mechanistic model (Daisy) developed based on data obtained in the SAFIR project on measured leaf gas exchange and soil water dynamics in irrigated potato crops grown in a semi-field environment subjected to different irrigation regimes. Experimental data was compared to simulated results from the new enhanced Daisy model which include modelling 2D soil water flow, abscisic acid (ABA) signalling and its effect on stomatal conductance and hence on transpiration and assimilation, and finally crop yield. The results demonstrated that the enhanced Daisy model is capable of simulating the mechanisms underlying the water saving effects of the partial root-zone drying (PRD) irrigation as compared with the conventional full irrigation (FI). However the simulated effect on both crop yield and water use in this particular experiment was negligible indicating more experimental studies are necessary in order to improve on the model.  相似文献

We report the effect of salinity and temperature on the viability of stored culture-based subitaneous eggs of the calanoid copepod Acartia tonsa for use of copepods in fish larvae culture. Quiescence induction was recorded at 17 and 25 °C, in salinities from 0 to 30. Quiescence was strongly induced at 0 salinity and partially at 5 in both temperatures. Eggs incubated at 0 salinity for up to 12 days at both temperatures showed a decline in the fraction able to be induced into quiescence by abrupt salinity changes. The hatching success of eggs that were able to enter quiescence stabilized after a 1-day incubation and remained ∼25% viable for 12 days in 17 °C. On the contrary, the 25 °C trial showed a gradual decline in viability until stabilizing ∼10% at day 7 and onwards. Longterm 17 °C incubation for 35 days showed that eggs remained quiescent with a viability of ∼14%. Hence, we recommend salinity storage of A. tonsa subitaneous eggs as a relevant shortterm technique, and a suitable alternative to the recently proposed cold storage of eggs when eggs are to be shipped from the copepod producer to a given fish larvae hatchery.  相似文献
