• Background and purpose   

A complex interplay between biotic and abiotic factors is believed to be responsible for several oak declines in pedunculate oak (Quercus robur L.). This study aims to clarify the temporal process of oak declines, as well as identifying individual tree and environmental variables that affects growth rate and that may increase the risk of mortality. The decline of N from 15N-labelled mature pea residues was followed in unplanted soil over 16.5 yr. Eight years after residue incorporation, 24% of the residue 15N input was still present in the soil and, after 16.5 yr, 16% of the residue 15N input remained. A double exponential model successfully described the decay of N from 15N-labelled pea residues. The total residual 15N declined with average decay constants of 1.45 yr−1 for the 30 d to 1 yr period and of 0.07 yr−1 for the 1-16 yr period. Sixteen years following incorporation of the residues, indicator plants growing in residues-amended soils were obtaining 1.7% of their N from residue N. This is, to our knowledge, the longest study on decay of N in soils from 15N-labelled crop residues. The current study thus provides a unique data set for our empirical understanding of N-dynamics in agricultural systems, which is a prerequisite to parameterize and validate N-simulation models.  相似文献

The alkylresorcinol (AR) content of 8 commonly consumed cereals, 125 Triticum cultivars, milling fractions of wheat and rye, bread, and other cereal products was analyzed. ARs were found in wheat (489-1429 microgram/g), rye (720-761 microgram/g), triticale (439-647 microgram/g), and barley (42-51 microgram/g), but not in rice, oats, maize, sorghum, or millet. One durum wheat variety was found to have an exceptionally low level of ARs (54 microgram/g) compared to other durum wheat varieties (589-751 microgram/g) and Triticumspecies analyzed. The AR content of milling fractions closely followed the ash content and could be used as a marker of the presence of bran in flour. Using hot 1-propanol extraction, all ARs could be extracted from bread, contrary to previous studies which suggested that ARs were destroyed during baking. Cereal products varied greatly in AR content, with those containing wheat bran or whole rye having the highest content.  相似文献

In temperate regions, climate change is predicted to increase annual mean temperature and intensify the duration and frequency of summer droughts, which together with elevated atmospheric carbon dioxide (CO2) concentrations, may affect the exchange of nitrous oxide (N2O) and methane (CH4) between terrestrial ecosystems and the atmosphere. We report results from the CLIMAITE experiment, where the effects of these three climate change parameters were investigated solely and in all combinations in a temperate heathland. Field measurements of N2O and CH4 fluxes took place 1-2 years after the climate change manipulations were initiated. The soil was generally a net sink for atmospheric CH4. Elevated temperature (T) increased the CH4 uptake by on average 10 μg C m−2 h−1, corresponding to a rise in the uptake rate of about 20%. However, during winter elevated CO2 (CO2) reduced the CH4 uptake, which outweighed the positive effect of warming when analyzed across the study period. Emissions of N2O were generally low (<10 μg N m−2 h−1). As single experimental factors, elevated CO2, temperature and summer drought (D) had no major effect on the N2O fluxes, but the combination of CO2 and warming (TCO2) stimulated N2O emission, whereas the N2O emission ceased when CO2 was combined with drought (DCO2). We suggest that these N2O responses are related to increased rhizodeposition under elevated CO2 combined with increased and reduced nitrogen turnover rates caused by warming and drought, respectively. The N2O flux in the multifactor treatment TDCO2 was not different from the ambient control treatment. Overall, our study suggests that in the future, CH4 uptake may increase slightly, while N2O emission will remain unchanged in temperate ecosystems on well-aerated soils. However, we propose that continued exposure to altered climate could potentially change the greenhouse gas flux pattern in the investigated heathland.  相似文献

This study aimed to find a fast, sensitive and efficient protocol for molecular identification of chicken Eimeria spp. in field samples. Various methods for each of the three steps of the protocol were evaluated: oocyst wall rupturing methods, DNA extraction methods, and identification of species-specific DNA sequences by PCR. We then compared and evaluated five complete protocols. Three series of oocyst suspensions of known number of oocysts from Eimeria mitis, Eimeria praecox, Eimeria maxima and Eimeria tenella were prepared and ground using glass beads or mini-pestle. DNA was extracted from ruptured oocysts using commercial systems (GeneReleaser, Qiagen Stoolkit and Prepman) or phenol-chloroform DNA extraction, followed by identification of species-specific ITS-1 sequences by optimised single species PCR assays. The Stoolkit and Prepman protocols showed insufficient repeatability, and the former was also expensive and relatively time-consuming. In contrast, both the GeneReleaser protocol and phenol-chloroform protocols were robust and sensitive, detecting less than 0.4 oocysts of each species per PCR. Finally, we evaluated our new protocol on 68 coccidia positive field samples. Our data suggests that rupturing the oocysts by mini-pestle grinding, preparing the DNA with GeneReleaser, followed by optimised single species PCR assays, makes a robust and sensitive procedure for identifying chicken Eimeria species in field samples. Importantly, it also provides minimal hands-on-time in the pre-PCR process, lower contamination risk and no handling of toxic chemicals.  相似文献

Purpose

Environmental chemistry of antimony (Sb) is still largely unknown. Many questions remain about its availability to plants and effects of fertilizers on mobility of Sb in the rhizosphere soil. In this work, we focused on the following problems: (1) uptake of Sb by wheat seedlings grown in soil enriched with this metalloid and (2) impact of soil amendments on the plant growth, Sb uptake from soil, and its transfer from roots to upper plant parts.

Materials and methods

To obtain further information on the possible transfer of Sb into plants, greenhouse pot experiments were carried out. Soil was spiked with 15 mg kg?1 of Sb and amended with either chicken manure or natural growth stimulator Energen. Wheat Triticum aestivum L. seedlings were grown in the soil during 17 days. Plants together with rhizosphere soil were collected several times in the course of the experiment. The ICP-OES and ICP-MS techniques were applied to determine the concentrations of macro- and trace elements in the plant and soil material.

Results and discussion

Growth of wheat seedlings in Sb-spiked soil resulted in Sb accumulation in roots and leaves of the plants. Energen and especially chicken manure were capable of stimulating transfer of Sb to more mobile and, as a consequence, more available to the plants form, thus enhancing both uptake of Sb from soil and its transfer from roots to upper plant parts. The accumulation of Sb by plants led to a decrease of Sb concentration in the rhizosphere soil with time, and the most significant decrease was observed after amendment of soil with fertilizers.

Conclusions

Fertilizers may be used to increase phytoextraction of Sb and its removal from contaminated soils. However, such an amendment of soil should be done with caution in order to exclude or at least reduce the negative effects on plants.  相似文献
