The purpose of this study was to determine the optimal histamine concentration and 'irritant' allergen threshold concentrations in intradermal testing (IDT) in normal cats. Thirty healthy cats were tested with three different histamine concentrations and four different concentrations of each allergen. The optimal histamine concentration was determined to be 1: 50,000 w/v (0.05 mg mL(-1)). Using this histamine concentration, the 'irritant' threshold concentration for most allergens was above the highest concentrations tested (4,000 PNU mL(-1) for 41 allergens and 700 PNU mL(-1) for human dander). The 'irritant' threshold concentration for flea antigen was determined to be 1:750 w/v. More than 10% of the tested cats showed positive reactions to Dermatophagoides farinae, Dermatophagoides pteronyssinus, housefly, mosquito and moth at every allergen concentration, which suggests that the 'irritant' threshold concentration for these allergens is below 1,000 PNU mL(-1), the lowest allergen concentration tested. Our results confirm previous studies in indicating that allergen and histamine concentrations used in feline IDT may need to be revised.  相似文献

OBJECTIVE: To compare the accuracy of a 3rd (Dolphin Voyager) versus 1st generation pulse oximeter (Nellcor N-180). STUDY DESIGN: Prospective laboratory investigation. ANIMALS: Eight adult dogs. METHODS: In anesthetized dogs, arterial oxygen saturation (SpO(2)) was recorded simultaneously with each pulse oximeter. The oxygen fraction in inspired gas (FiO(2)) was successively reduced from 1.00 to 0.09, with re-saturation (FiO(2) 0.40) after each breathe-down step. After each 3-minute FiO(2) plateau, SpO(2) and pulse rate (PR) were compared with the fractional arterial saturation (SaO(2)) and PR determined by co-oximetry and invasive blood pressure monitoring, respectively. Data analysis included Bland-Altman (B-A) plots, Lin's concordance correlation factor (rho(c)), and linear regression models. RESULTS: Over a SaO(2) range of 33-99%, the overall bias (mean SpO(2) - SaO(2)), precision (SD of bias), and accuracy (A(rms)) for the Dolphin Voyager and Nellcor N-180 were 4.3%, 4.4%, and 6.1%, and 3.2%, 3.0%, and 4.3%, respectively. Bias increased at SaO(2) < 90%, more so with the Dolphin Voyager (from 1.6% to 8.6%) than Nellcor N-180 (from 3.2% to 4.5%). The SpO(2) readings correlated significantly with SaO(2) for both the Dolphin Voyager (rho(c) = 0.94) and Nellcor N-180 (rho(c) = 0.97) (p < 0.001). Regarding PR, bias, precision, and accuracy (A(rms)) for the Dolphin Voyager and Nellcor N-180 were -0.5, 4.6, and 4.6 and 1.38, 4.3, and 4.5 beats minute(-1), respectively. Significant correlation existed between pulse oximeter and directly measured PR (Dolphin Voyager: rho(c) = 0.98; Nellcor N-180: rho(c) = 0.99) (p < 0.001). CONCLUSIONS AND CLINICAL RELEVANCE: In anesthetized dogs with adequate hemodynamic function, both instruments record SaO(2) relatively accurately over a wide range of normal saturation values. However, there is an increasing overestimation at SaO(2) < 90%, particularly with the Dolphin Voyager, indicating that 3rd generation pulse oximeters may not perform better than older instruments. The 5.4-fold increase in bias with the Dolphin Voyager at SaO(2) < 90% stresses the importance of a 93-94% SpO(2) threshold to ensure an arterial saturation of >or=90%. In contrast, PR monitoring with both devices is very reliable.  相似文献

This paper describes the composition of 30 grape-seed oils obtained from France, Italy, and Spain during 2002-2003. Oils were extracted from the seeds using small-scale industrial solvent extraction equipment and analyzed in their unrefined state using standard methods for fatty acids, fatty acids in the triacylglycerol 2-position, tocopherols and tocotrienols, triglycerides, sterols, steradienes, and iodine value. Values for the composition of the sterols, triglycerides, fatty acids, iodine value, and tocopherol composition were generally in good agreement with the results of previous similar surveys. Steradienes (stigmastadiene, campestadiene, stigmastatriene, and campestatriene) were detected in the oil and were probably formed from sterols during the extraction process.  相似文献

Experience can alter synaptic connectivity throughout life, but the degree of plasticity present at each age is regulated by mechanisms that remain largely unknown. Here, we demonstrate that Paired-immunoglobulin-like receptor B (PirB), a major histocompatibility complex class I (MHCI) receptor, is expressed in subsets of neurons throughout the brain. Neuronal PirB protein is associated with synapses and forms complexes with the phosphatases Shp-1 and Shp-2. Soluble PirB fusion protein binds to cortical neurons in an MHCI-dependent manner. In mutant mice lacking functional PirB, cortical ocular-dominance plasticity is more robust at all ages. Thus, an MHCI receptor is expressed in central nervous system neurons and functions to limit the extent of experience-dependent plasticity in the visual cortex throughout life. PirB is also expressed in many other regions of the central nervous system, suggesting that it may function broadly to stabilize neural circuits.  相似文献

The phenylpyrazole insecticide, fipronil, is used in seed coating against Agriotes larvae, which infest mainly corn and sunflower. Coating the seeds of the cultivated plants with fipronil has proven its effectiveness against Agriotes populations. In the case of sunflower or even corn, the possible root uptake of this insecticide may lead to a toxic effect against pollinators such as honeybees. In the present report, the uptake and transport of fipronil inside the sunflower seedling was studied in the laboratory. In a first study, sunflower was cultivated on an aqueous medium containing fipronil. An intense root uptake of fipronil occurred, leading to a transport into leaves depending upon transpiration. In a second study, plants were cultivated on a soil in which fipronil was uniformly distributed. Under our soil conditions (20% organic carbon), the partition coefficient between soil and water (K(d)) was found to be equal to 386 +/- 30. The average rate of fipronil transfer from soil water to seedlings was from 2 to 2.6 times lower than water transfer. During the 3 week experiment, 55% of recovered labeled compounds was in the parent form and 35% had been converted to lipophilic metabolites, with either a 4-CF(3)-SO(2) or 4-CF(3)-S substituant, which are also very potent lipophilic insecticides. This paper suggests that the possible uptake of fipronil by sunflower seedlings under agronomic conditions is mainly controlled by the physicochemical characteristics of the seed-coating mixture.  相似文献

In stocking program, the use of artificial incubation conditions in hatcheries from the fertilisation of eggs to the release of unfed fry could reduce their ability to adapt to the natural environment. This study evaluates the effects of three factors on the fitness and physiology of salmon fry at their emergence, the origin of water (river vs drilling), the type of support in the incubator (support matrix vs plastic sheets) and the type of incubators (Californian vs vertical trays), and compares them to a semi-natural incubation method in river. Key biological functions including nutritional and immune status were compared among experimental conditions using biometric parameters, lipid composition and gene expression analyses. Our findings demonstrated that fry incubated in vertical trays supplied with river water had no significant difference in growth and lipid composition compared to those in semi-natural incubators. Besides, fry incubated on a substrate matrix in Californian trays exhibited phenotypic characteristics closest to those incubated in river. This support matrix improved fish growth, lipid consumption and distribution compared to fry on plastic sheets. Moreover, the large amounts of several PUFAs in these fry could allow a better membrane fluidity ensuring a better adaptation to temperature variation under cold conditions. In addition, drilling water improved the survival rate compared to river water due to lower numbers of fine particles, known to be responsible for the clogging of eggs. To conclude, using a substrate combined with drilling water in artificial incubators could increase fry fitness and its adaption to wild life.

As urban development continues to encroach on the natural and rural landscape, land-use planners struggle to identify high priority conservation areas for protection. Although knowing where urban-sensitive species may be occurring on the landscape would facilitate conservation planning, research efforts are often not sufficiently designed to make quality predictions at unknown locations. Recent advances in occupancy modeling allow for more precise estimates of occupancy by accounting for differences in detectability. We applied these techniques to produce robust estimates of habitat occupancy for a subset of forest breeding birds, a group that has been shown to be sensitive to urbanization, in a rapidly urbanizing yet biological diverse region of New York State. We found that detection probability ranged widely across species, from 0.05 to 0.8. Our models suggest that detection probability declined with increasing forest fragmentation. We also found that the probability of occupancy of forest breeding birds is negatively influenced by increasing perimeter-area ratio of forest fragments and urbanization in the surrounding habitat matrix. We capitalized on our random sampling design to produce spatially explicit models that predict high priority conservation areas across the entire region, where interior-species were most likely to occur. Finally, we use our predictive maps to demonstrate how a strict sampling design coupled with occupancy modeling can be a valuable tool for prioritizing biodiversity conservation in land-use planning.  相似文献

In a mesocosm experiment, with bare peat soils exposed to different water levels (WL), we examined whether heterotrophic respiration (Rh) acclimated to a 3 °C temperature increase. Across all WLs, Rh at 15 °C was never lower in the heated treatment than in the unheated treatment, indicating that Rh did not acclimate to the warmer conditions. We hypothesize that this lack of thermal acclimation is due to the unlimited substrate availability in these organic soils. These results imply that peat soils may exhibit a sustained positive feedback to global warming.  相似文献

Respiration of CO2 from soils (Rs) is a major component of the carbon cycle of ecosystems, but understanding is still poor of both the relative contributions of different respiratory sources to Rs, and the environmental factors that drive diurnal variations in Rs. We measured total and litter-free Rs at half-hourly intervals over full 24 h periods, and thereafter twice a month for 10 months in a tropical montane cloud forest (TMCF) in Peru. Total Rs declined by about 61% during the night as a result of variations in respiration rate in the litter, which were partly correlated with the soil surface air temperature. Most of the diurnal variation of Rs in this TMCF appears to be driven by respiration in the litter layer, which contributed 37% to the total soil CO2 efflux. Total Rs rates at this particular site would have been overestimated by 60% if derived from daytime measurements that had not been corrected for diurnal variations in Rs.  相似文献
