Various anticoagulant were assessed in order to develop a transport media for bovine blood which would improve leukocyte stability. The effects of time, disease in the donor and different personnel and laboratory equipment were also tested.
The transport media most effective for leukocyte stability was found to be Titriplex IV. This is a modification of Titriplex I by the inclusion of acetylsalicyclic acid to prevent cellular clumping and by the removal of methylene blue. Anticoagulant citrate dextroseformalin-ASA performed equally well for transport but cellular morphology was impaired.
Febrile or neoplastic disease in the donor animal did not alter the efficiency of the formalin containing anticoagulants.

The effects of addition of evening primrose oil (EPO) to a mink diet in the breeding season on the reproductive performance and kit and female performance during the lactation period were investigated in an experiment with 4 groups of male and female mink. Matings were carried out so that control males were mated to both control and supplemented females. Similarly, supplemented males mated both control and supplemented females. Reproductive results were evaluated both on a group basis and as an effect of male or female treatment, respectively. After males supplemented with EPO, there was a tendency for reduced rate of stillborn kits and kit losses during the first 21 days of life. These effects could not be explained physiologically. Female treatment did not affect reproductive performance, but there was a tendency for lower weight losses during lactation for EPO-supplemented females. A direct radioimmunoassay procedure for the determination of melatonin in the blood of blue fox has been validated and applied.The assay required 50 μl of sample and standard, 100 ul of antiserum and 100 μl of (3H)melatonin. After overnight incubation at 4°C the antibody bound melatonin was separated from the free hormone with dextran-coated charcoal. Following centrifugation the antibody bound (3H)melatonin was determined in a beta scintillation counter.The antiserum bound 30–35 % of the (3H)melatonin at a final dilution of 1:36000. The non specific binding represented less than 5% of the total radioactivity in all assays. The lowest detectable amount of melatonin was 2.6 fmol/tube, corresponding to 52.5 pmol/1. The inter-assay coefficient of variation at 178 and 510 pmol/1 was 15.6 and 8.8%, respectively. The precision profile, calculated from a 10-replicate standard curve, showed that the coefficient of variation decreased from 43% at 84 pmol/1 to 15% at 336 pmol/1, and remainded at or below 10% for concentrations exceeding 670 pmol/1.Plasma was collected from 2 male blue foxes at about hourly intervals during a 24 h period in September and assayed for melatonin. Background
Vancomycin resistant enterococci are a frequent cause of nosocomial infections and their presence among farm animals is unwanted. Using media supplemented with vancomycin an increase in the proportion of samples from Swedish broilers positive for vancomycin resistant enterococci has been detected. The situation at farm level is largely unknown. The aims of this study were to obtain baseline knowledge about environmental contamination with vancomycin resistant enterococci in Swedish broiler production and the association between environmental contamination and colonisation of birds.Methods
Environmental samples were taken before, during and after a batch of broilers at three farms. Samples were cultured both qualitatively and semi-quantitatively for vancomycin resistant enterococci. In addition, caecal content from birds in the batch following at each farm was cultured qualitatively for vancomycin resistant enterococci.Results
The number of samples positive for vancomycin resistant enterococci varied among the farms. Also the amount of vancomycin resistant enterococci in the positive samples and the proportion of caecal samples containing vancomycin resistant enterococci varied among the farms. Still, the temporal changes in environmental contamination followed a similar pattern in all farms.Conclusion
Vancomycin resistant enterococci persist in the compartments even after cleaning and the temporal changes in environmental contamination were similar among farms. There were however differences among farms regarding both degree of contamination and proportion of birds colonized with vancomycin resistant enterococci. The proportion of colonized birds and the amount of vancomycin resistant enterococci in the compartments seems to be associated. If the factor(s) causing the differences among farms could be identified, it might be possible to reduce both the risk for colonisation by vancomycin resistant enterococci of the subsequent flock and the risk for spread of vancomycin resistant enterococci via the food chain to humans. 相似文献76.

Calonectria ilicicola, Gliocladiopsis sp. and Ilyonectria liriodendri were isolated from diseased roots of young avocado trees. Pathogenicity studies with seedlings of three avocado cultivars, Velvick, Hass and Reed, demonstrated that Calonectria ilicicola is a severe root rot pathogen, reducing the biomass of healthy roots, and reducing plant height over time. Calonectria ilicicola was re‐isolated from diseased roots. Ilyonectria liriodendri and Gliocladiopsis sp. were not pathogenic and plant height was increased after Gliocladiopsis sp. amendment compared to all other treatments in trials with cvs Velvick and Hass. 相似文献

Parturition was induced in 12 dairy heifers with prostaglandin (PG) F2 alpha about 2 weeks before the expected time of calving. Eight animals gave birth after two injections (group 1), three animals needed more than two injections (group 2) and one animal (cow no. 740) required one injection. All animals in groups 1 and 2 had retained foetal membranes and the time needed to induce parturition was 59 +/- 7 and 149 +/- 10 h, respectively. As cow no. 740 did not have retained foetal membranes and calved 24 h after one PGF2 alpha injection, it was excluded from the results. Udder distension and relaxation of the pelvic ligaments could predict the calving to within 12 h. Furthermore, the pre-calving drop of body temperature could predict the time of parturition to within 16 h. The total white blood cells and polymorphonuclear cells were at their highest values on the day preceding parturition whereas mononuclear cells had a tendency to increase 3 days after calving. Increased levels of haemoglobin were found at the time of parturition, whereas, plasma-calcium levels significantly decreased after parturition (P < 0.001). Progesterone levels markedly decreased after the first PGF2 alpha injection and reached 2 nmol/l at the time of parturition. Plasma levels of oestradiol-17 beta reached the peak at the time of parturition, whereas, the highest levels of the PGF2 alpha metabolite and cortisol were recorded 16 h after calving. 相似文献

Three dairy heifers were examined during three consecutive oestrous cycles (control period, CP). Subsequently, the animals were subjected to 4 and then 5 weeks of twice-weekly ovum pick-up (OPU) (FPP1 and FPP2, respectively) with a recovery period (RP) of two consecutive oestrous cycles between FPP1 and FPP2. After FPP2, the animals were slaughtered and the ovaries were macroscopically examined. Throughout, ovarian activity was monitored by transrectal ultrasonography and concentrations of plasma progesterone. During CP, all the heifers showed normal cyclicity. During FPPs, the heifers occasionally presented oestrous activity. Corpus luteum (CL)-like structures developed from punctured follicles with diameters and life-spans varying from smaller and shorter than those in the CP (P > 0.05) to equal to those in the CP. There was a tendency for a lower number of emerging and punctured follicles in the presence of a CL-like structure. Subsequently to FPP1, all heifers regained normal cyclicity. A thickening of the ovarian tunica albuginea and a slight hardening of the ovaries were found postmortem. In conclusion, dairy heifers can occasionally show cyclic activity and form CL-like structures during twice-weekly OPU. Further, OPU did not seem to cause any major negative effects on ovarian structure and subsequent ovarian function. 相似文献

A Burdizzo castrator was evaluated for the neutering of dogs. Histological and morphological changes of spermatic cells and peripheral serum testosterone after challenge with a GnRH-analogue (gonadorelin) were assessed. There was a control group (G1), a surgically castrated group (G2) and a Burdizzo group (G3) divided in two, G3a receiving two crunches in each spermatic cord and G3b receiving one crunch in each spermatic cord. Sixteen days after application of the Burdizzo blood samples were taken from the dogs at 30 min interval during 2 h; after the second sample the dogs were treated with 1 mug/kg body weight of gonadorelin i.v. The same protocol of gonadorelin challenge was performed in G1 and G2 dogs. The G2 dogs were surgically castrated after the second blood sample, before the gonadorelin treatment, and the G1 dogs after the last blood sample. The excised gonads were examined histologically, and sperm smears were prepared from the caudae epididymidis. The testes and plexus pampiniformis of the G1 and G2 dogs had a normal histological appearance, and they had morphologically normal epididymal sperm cells. In all G3 dogs, there was an acute fibrosis with an inflammatory reaction in the plexus pampiniformis. The testes from the G3a dogs showed diffuse areas of infarction and degeneration of the parenchyma. Similar but less diffuse lesions were seen in group 3b dogs. The deferent ducts from all G3 dogs showed vasitis and/or sperm granulomas. Azoospermia or sperm malformations were observed in the epididymal smears from the G3 dogs. Testosterone concentration in the G1 dogs increased after gonadorelin application (p < 0.0001). The G2 dogs had basal testosterone levels after castration (p < 0.001) and did not respond to gonadorelin. Groups 3a and b showed a slight but non-significant increase in testosterone concentration after gonadorelin challenge, supposedly due to the reduction of testicular blood flow and loss of testicular interstitial tissue. 相似文献

Addition of evening primrose oil (EPO) to a blue fox diet in the reproduction period was evaluated in an experiment with 2 groups, each of 12 male and 25 female blue foxes, regarding the effects on reproductive performance. The experiment was carried out as a field trial and the experimental period lasted from March 10 until the end of the mating season (males) or early July (females). During this period the control group was fed the standard diet of the farm and the experimental group was fed the same diet supplemented with 4.5 g EPO and 2.5 mg zinc sulphate per animal and day. An addition of 10 mg vitamin E per 500 mg EPO was made. The results were evaluated regarding male and female treatment effects. There was an increased rate of abortions in the EPO-group, but simultaneously a non-significant decrease in the frequency of barren females, resulting in a similar level of females without litters in both groups. A tendency for increased litter size in the EPO group was found, mainly as an effect of male treatment, which might indicate an effect on semen quality. 相似文献
