The effects of aluminum (Al) stresses of varying intensities and durations on growth parameters were investigated in four Oryza sativa L. genotypes (two Al sensitive, two Al resistant). Our results support the current opinion that the use of only one parameter cannot easily account for differences in Al resistance, since Al concentration or stress duration can affect plant responses in opposite directions. For short stress duration (40 days) at Al 500 μM, weight parameters appear to be more discerning than length parameters. Necrosis evaluation is a more reliable criterion for plants maintained for a long time (80 days) in the presence of high Al concentrations (Al 1000 or 1500 μM).  相似文献

Biochanin A and formononetin are the predominant isoflavones in red clover. In a previous study (J. Agric. Food Chem. 2002, 50, 4783-4790), it was demonstrated that human liver microsomes converted biochanin A and formononetin to genistein and daidzein. This paper now shows CYP1B1-catalyzed O-demethylation of biochanin A and formononetin to produce genistein and daidzein, respectively, which inhibit CYP1B1. Recombinant human CYP1A1 or CYP1B1 was incubated with biochanin A or formononetin. CYP1A1 catalyzed isoflavone 4'-O-demethylation and hydroxylations with similar efficiency, whereas CYP1B1 favored 4'-O-demethylation over hydroxylations. Three of the biochanin A metabolites (5,7,3'-trihydroxy-4'-methoxyisoflavone, 5,7,8-trihydroxy-4'-methoxyisoflavone, and 5,6,7-trihydroxy-4'-methoxyisoflavone) were characterized by 1H NMR spectroscopy and mass spectrometry. Daidzein (Ki = 3.7 microM) exhibited competitive inhibition of CYP1B1 7-ethoxyresorufin O-deethylase activity, and genistein (Ki = 1.9 microM) exhibited mixed inhibition. Biochanin A and/or formononetin may exert anticarcinogenic effects directly by acting as competitive substrates for CYP1B1 or indirectly through their metabolites daidzein and genistein, which inhibit CYP1B1.  相似文献

Over the last two decades, soil cultivation practices in the southern Argentinean Pampas have been changing from a 7 year cash-crop production system alternated with 2–3 years under pasture, to a continuous cropping system. A better understanding of the impact of the period of time a field has been under continuous cropping on a broad spectrum of soil properties related to soil quality is needed to target for sustainable cropping systems. The objectives of this study were to: (i) assess the relationship between physical and chemical soil parameters related to soil quality and (ii) identify soil quality indicators sensitive to soil changes under continuous cropping systems in the Argentinean Pampas.

Correlation analysis of the 29 soil attributes representing soil physical and chemical properties (independent variables) and years of continuous cropping (dependent variable) resulted in a significant correlation (p < 0.05) in 78 of the 420 soil attribute pairs. We detected a clear relationship between hydraulic conductivity at tension h (Kh) and structural porosity (ρe); ρe being a simple tool for monitoring soil hydraulic conditions.

Soil tillage practice (till or no-till) affected most of the soil parameters measured in our study. It was not possible to find only one indicator related to the years under continuous cropping regardless of the cultivation practice. We observed a significant relationship between years under continuous cropping and Kh under no-till (NT) and wheat fallow (p < 0.001, R2 = 0.70). Under these conditions, K−40 diminished as the number of years under continuous cropping increased.

The change in mean weight diameter (CMWD) was the only physical parameter related to the number of years under continuous cropping, explaining 36% of the variability in the number of years under continuous cropping (p < 0.001) The combination of three soil quality indicators (CMWD, partial R2 = 0.38; slope of the soil water retention curve at its inflexion point (S), partial R2 = 0.14 and cation exchange capacity (CEC), partial R2 = 0.13) was able to explain, in part, the years under continuous cropping (R2 = 0.65; p value > 0.001), a measure related to soil quality.  相似文献

The baseflow of tributaries to the Velhas River in the northwest region of Ouro Preto County (São Francisco Basin, Brazil) has been declining in the last years, without a simultaneous decrease in the water input. This can only be attributed to badly planned human activities, which result in soil crusting or compaction, erosion, among other types of impact, such as groundwater overexploitation and wetland drainage. Almost all of the study area has been eroded by hundreds of large-size gullies. In order to analyze the influence of erosion in the hydrological behavior of small streams, two similar and contiguous catchments were selected. The only remarkable difference between them is a gully that occupies circa 42% of one of the catchments (“eroded catchment”). Rainfall, flow rates and chemical data were obtained throughout a hydrological year and the results showed that the eroded catchment presented smaller baseflow rates and bigger but short-lived stormflows. This contrasting flow regime is attributed to the gully, which is causing the partial withdraw of the regolith and the exposure of weathered rock. As the regolith has smaller hydraulic conductivities and higher storage coefficients, the exposure of weathered rock on the gully floor explains the odd flow behavior of the eroded catchment. Other studies in the same region confirm the impact caused by the gullies. Thus, the superposition of many gullies can cause decrease of the baseflow and increase of the stormflow. As gullies are very common erosion features in Brazil, they should be much more studied in order to better understand this and other similar types of environmental impact.  相似文献

We report on the development of a novel alternative method for the assessment of floral origin in honey samples based on the study of honey proteins using immunoblot assays. The main goal of our work was to evaluate the use of honey proteins as chemical markers of the floral origin of honey. Considering that honeybee proteins should be common to all types of honey, we decided to verify the usefulness of pollen proteins as floral origin markers in honey. We used polyclonal anti-pollen antibodies raised in rabbits by repeated immunization of Sunflower (Elianthus annuus) and Eucalyptus (Eucalyptus sp.) pollen extracts. The IgG fraction was purified by immunoaffinity. These antibodies were verified with nitrocellulose blotted pollen and unifloral honey protein extracts. The antibodies anti-Sunflower pollen, bound to the 36 and 33 kDa proteins of Sunflower unifloral honey and to honey containing Sunflower pollen; and the antibodies anti-Eucalyptus sp. pollen bound to the 38 kDa proteins of Eucalyptus sp. unifloral honey in immunoblot assays. Satisfactory results were obtained in differentiating between the types of pollen analyzed and between Sunflower honey and Eucalyptus honey with less cross reactivity with other types of honey from different origin and also with good sensitivity in the detection. This immunoblot method opens an interesting field for the development of new antibodies from different plants, which could serve as an alternative or complementary method to the usual melissopalynological analysis to assess honey floral origin.  相似文献

Soil and subsoil aluminium toxicity has been one of the main limiting factors for soybean and wheat yields in tropical soils. Usually liming is the most effective way to deal with soil acidity and Al toxicity, but in no-till systems the soil is not disturbed making it impossible to incorporate lime in the arable layer, and lime has been usually applied on the soil surface. In this paper soybean and wheat responses to lime applied on the soil surface and/or incorporated in the soil arable layer were evaluated during the transition from conventional tillage to a no-till system. The experiment was conducted for 3 years in Paraná, Brazil, using a wheat–soybean rotation. Lime rates ranging from 0.0 to 9.0 t ha−1 were incorporated down to 20 cm and 4.5 t ha−1 were spread or not on the soil surface. Soil samples were taken down to 60 cm, 39 months after the first lime application. Soil chemical characteristics were affected by lime application down to 60 cm deep in the profile. Soybean responded to lime irrespective of application method, but the highest accumulated yield was obtained when lime was incorporated into the arable layer. For wheat, the more sensitive the cultivar, the greater was the response to lime. During the introduction of a no-till system, lime must be incorporated into the arable layer when the wheat cultivar is Al-sensitive.  相似文献

Purpose

Although the influences of urban land use on water quality have been widely investigated, the impacts of different urbanization patterns, particularly in Mediterranean environments, are not well understood. Focussing on a Portuguese peri-urban catchment with 40 % urban cover, this paper explores (1) the impact of areas with differing urban extent and storm drainage system on streamwater quality and (2) temporal variations driven by season and storm events of differing magnitude, intensity and antecedent weather.

Materials and methods

Water quality was assessed at the catchment outlet (E) and for three upstream tributaries: (1) Porto Bordalo (PB), 39 % urban with a new major road and piping of some overland flow from impervious surfaces directly into the stream; (2) Espírito Santo (ES), 49 % urban, mostly comprising detached houses surrounded by gardens, and with overland flow infiltrating into downslope pervious soils; and (3) Quinta (Q), 22 % urban with partial piping of overland flow from a recent enterprise park area. Water samples were collected at different stages in storm hydrograph responses to ten rainfall events from October 2011 to March 2013. The water quality variables analysed included chemical oxygen demand (COD), nutrients (Kjeldahl nitrogen [Nk-N], ammonium [NH4–N], nitrate [NO3–N] and total dissolved phosphorus (TDP) and heavy metals (Zn and Cu).

Results and discussion

Urban areas had great impact on COD, with the highest median concentrations in ES and the lowest in Q. In ES, fertilizing lawns and gardens may have been responsible for its higher median NO3–N concentrations. High concentrations of heavy metals were recorded in PB and Q, probably due to the piping of road runoff directly into the stream. Generally, higher pollutant concentrations were recorded in the first storm events after the summer drought due to the flushing of accumulated solutes and a lower dilution effect, with Nk-N and NH4–N exceeding water quality standards. Over the wet season, increasing soil moisture favoured greater flow connectivity between runoff processes from pollutant sources and the stream network, leading to a higher proportion of samples exceeding pollution thresholds.

Conclusions

No direct relationship was identified between urban extent and water quality, possibly due to the overriding impact of different storm drainage systems and flow connectivities of different urban patterns. Hydrological regime, linked to seasonal changes, also exerted a major influence on the water quality dynamics. Information on the spatiotemporal dynamics of pollutants, linked to different urban patterns and storm drainage systems, should help enable urban planners to minimize the adverse impacts of urbanization on aquatic ecosystems.
