Phenolic acids, which are generally esterified with tartaric acid, are natural constituents of grape must and wine and can be released as free acids (principally p-coumaric, caffeic, and ferulic acids) by certain cinnamoyl esterase activities during the wine-making process. Some of the microorganisms present in grape can metabolize the free phenolic acids into 4-vinyl and 4-ethyl derivatives. These volatile phenols contribute to the aroma of wine. The Saccharomyces cerevisiae phenyl acrylic acid decarboxylase gene (PAD1) is steadily transcribed, but its encoded product, Pad1p, shows low activity. In contrast, the phenolic acid decarboxylase (PADC) from Bacillus subtilis and the p-coumaric acid decarboxylase (PDC) from Lactobacillus plantarum display substrate-inducible decarboxylating activity in the presence of phenolic acids. In an attempt to develop wine yeasts with optimized decarboxylation activity on phenolic acids, the padc, pdc, and PAD1 genes were cloned under the control of S. cerevisiae's constitutive phosphoglyceratekinase I gene promoter (PGK1(P)()) and terminator (PGK1(T)()) sequences. These gene constructs were integrated into the URA3 locus of a laboratory strain of S. cerevisiae, Sigma1278b. The overexpression of the two bacterial genes, padc and pdc, in S. cerevisiae showed high enzyme activity. However, this was not the case for PAD1. The padc and pdc genes were also integrated into an industrial wine yeast strain, S. cerevisiae VIN13. As an additional control, both alleles of PAD1 were disrupted in the VIN13 strain. In microvinification trials, all of the laboratory and industrial yeast transformants carrying the padc and pdc gene constructs showed an increase in volatile phenol formation as compared to the untransformed host strains (Sigma1278b and VIN13). This study offers prospects for the development of wine yeast starter strains with optimized decarboxylation activity on phenolic acids and the improvement of wine aroma in the future. 相似文献

Signal peptide peptidase (SPP) catalyzes intramembrane proteolysis of some signal peptides after they have been cleaved from a preprotein. In humans, SPP activity is required to generate signal sequence-derived human lymphocyte antigen-E epitopes that are recognized by the immune system, and to process hepatitis C virus core protein. We have identified human SPP as a polytopic membrane protein with sequence motifs characteristic of the presenilin-type aspartic proteases. Habitats modify the population ecology of species. Here, we show how low water level influences abundance and size of adult anadromous brown trout (Salmo trutta) entering a small, South Norwegian stream for spawning. After smolting, the fish appear chiefly to feed within 10 km of the home stream. In the autumn, South Norwegian streams typically flood because of heavy rainfall, when the anadromous brown trout entered from the sea. Mean annual duration of the upstream migration period was 34 days and ended when the flooding ended and the water temperature dropped to below 4°C. During most of the migration period, on average two trout ascended the river per day. The sexes entered the spawning area concurrently, and the male:female ratio of the anadromous trout was 1.27. No fish entered when the water depth just downstream of the spawning area was below 5 cm, and mean number of fish increased with increasing water depth to ca. 30 cm, but not at higher flows when the ascent gradually decreased. Mean and maximum size of the entering spawners increased with water depth between 5 and 16 cm. Among those that had been to sea, most were recaptured in the home stream, 4% in other streams, but only two of the strays were caught close to spawning time. The present results illustrate that population traits of anadromous brown trout from a small stream differ from those in larger rivers, probably because of selection associated with water flow. 相似文献

Spectrophotometric, morphologic and dielectric properties of polyacrylonitrile (PAN) composite films in the presence of pyrrole
derivatives were reported in this paper. The composite films were fabricated by oxidative polymerization of pyrrole (Py),
N-methyl pyrrole (NMPy) and N-phenyl pyrrole (NPhPy) by cerium(IV) on polyacrylonitrile matrix. The effect of temperature
on the dielectric properties was studied in the frequency range from 0.05 Hz up to 10 MHz and in the temperature range from
0 °C up to 250 °C. Conductivity was increased with temperature due to increase of the mobility of charge carriers in the composite
films. By increasing the temperature, the dipoles become free and respond to the applied electric field in composite structure;
thus, the polarization and dielectric constant increases. PNPhPy-PAN composite films exhibited the highest dielectric constant,

Interaction among plant nutrients can yield antagonistic or synergistic outcomes that influence nutrient use efficiency. To provide insight on this phenomenon, peer-reviewed articles were selected that quantified the interaction effects of nutrients on crop yield levels. In total 94 articles were selected that described 117 interactions between all macro- and micronutrients for different agricultural crops. In 43 cases the interaction was synergistic, in 17 cases the interaction was antagonistic, and in 35 cases the interaction was zero-interaction; the other 23 cases were non-significant (16) or showed a negative response (7). Generally: (a) when the availability of two nutrients is characterized as deficient, a large increase in yield can be expected by diminishing these deficiencies: (b) for most macronutrients the mutual interactions on yield levels are synergistic; and (c) antagonistic effects on yield are often found for divalent cations. Knowledge of nutrient interactions can guide fertilizer design and optimization of fertilization strategies for high yields and high nutrient use efficiencies. 相似文献

Several species of ambrosia beetles (Coleoptera: Scolytidae) have recently started attacking standing, living beeches (Fagus sylvatica) in southern Belgium. In 2001, 1.3 million m3 of apparently healthy trees were struck. So far the outbreak has been limited to the Ardenne, and partly the Gaume, areas, and Brussels has been untouched. The city of Brussels is surrounded by a vast 4300 ha forest, mainly planted with beech, the Forêt de Soignes, of invaluable ecological and recreational value, of which 1600 ha belong to the regional authorities. In the spring 2001, these latter commissionned a study to assess the new threat to the forest. A 500 m×500 m grid of small traps, baited with ethanol and lineatin, was deployed over the regional part of the forest. The main species caught were Trypodendron domesticum, Anisandrus dispar and, in high numbers, Xylosandrus germanus, an exotic species of Asian origin found for the first time in Belgium in 1994. Whilst there was a consistent homogeneity between catches within the same sites (2 traps/site, distant by 2–6 m), there were no spatial relationships between catches at larger distances for T. domesticum and A. dispar. For X. germanus, spatial autocorrelations were observed within distances of 2000 m, suggesting that this species has sufficient mobility to cover this range. The planning of the 2002 trapping campaign will take this information into account: the traps will be deployed within a smaller grid. 相似文献
