Genetic regulation of mammalian heart size is poorly understood. Hippo signaling represents an organ-size control pathway in Drosophila, where it also inhibits cell proliferation and promotes apoptosis in imaginal discs. To determine whether Hippo signaling controls mammalian heart size, we inactivated Hippo pathway components in the developing mouse heart. Hippo-deficient embryos had overgrown hearts with elevated cardiomyocyte proliferation. Gene expression profiling and chromatin immunoprecipitation revealed that Hippo signaling negatively regulates a subset of Wnt target genes. Genetic interaction studies indicated that β-catenin heterozygosity suppressed the Hippo cardiomyocyte overgrowth phenotype. Furthermore, the Hippo effector Yap interacts with β-catenin on Sox2 and Snai2 genes. These data uncover a nuclear interaction between Hippo and Wnt signaling that restricts cardiomyocyte proliferation and controls heart size.  相似文献

Key message

Pinus pinaster Ait. susceptibility to pinewood nematode significantly differed among provenances, and the two Atlantic provenances of the Iberian Peninsula being the most affected. However, significant provenance × environment interaction was found. Provenance susceptibility was related to basal diameter, number of branches and oleoresin flow, and some climatic parameters.

Context

The pinewood nematode Bursaphelenchus xylophilus, native to North America, is an important pest affecting pine forests throughout Eurasia. In Europe, it has been detected in Portugal and Spain and is primarily associated with Pinus pinaster, an important Mediterranean tree species.

Aims

We have investigated the differences in susceptibility among several P. pinaster provenances in the Iberian Peninsula and France, as well as their relationship to certain growth traits and physiological parameters.

Methods

Three independent inoculation tests were performed on 3 to 4-year-old trees, followed by assessment of growth traits and physiological variables, along with time course destructive sampling for nematode quantification.

Results

The results showed significant differences among provenances for almost all growth traits, wilting, and mortality, though a significant provenance × environment interaction was also detected. Two Atlantic provenances, Noroeste-Litoral and Leiria, displayed the largest susceptibility to pinewood nematode. Changes in susceptibility to B. xylophilus between experiments were influenced by temperature and seasonality. Autumn precipitation and mean maximum temperature during summer at the original provenance sites could be related to provenance susceptibility.

Conclusion

Noroeste-Litoral and Leiria were the most disease-affected provenances. This study emphasizes the need for further research on how tree growth stage influences susceptibility and on the possibility of cross-breeding among provenances.

A historical outbreak of needle blight disease was recorded during 2018 to 2019 in plantations of Pinus radiata and Pinus nigra in the North of Spain. The main pathogens involved in this historical outbreak were identified as Lecanosticta acicola and Dothistroma septosporum. Recently, a variety of tree species in three arboreta planted between 2011 and 2013 in the Basque Country as part of the European project REINFFORCE were showing symptoms of needle blight and defoliation. The aim of this study was to determine which pine species were affected with these pathogens. Tree species sampled included several provenances of P. brutia, P. elliottii, P. nigra, P. pinaster, P. pinea, P. ponderosa, P. sylvestris and P. taeda. Using molecular identification methods, Lecanosticta acicola was confirmed infecting Pinus brutia (Provenance: Alexandropolis, Greece and var. eldarica, Crimea) and represents a new host species for this pathogen. Pinus elliottii (Provenance: Georgia, USA) and P. ponderosa (Provenance: Central California, USA) are new host reports of L. acicola for Spain. Dothistroma septosporum was found for the first time on P. brutia (Provenance: Marmaris, Turkey) and P. ponderosa (Provenance: Oregon, USA) in Spain and was also detected infecting P. nigra (Provenance: Sologne Vayrières, France).  相似文献

The phytopathogenic fungus Nectria galligena Bres. is the most common canker disease agent of hardwood trees. The terpenoids colletochlorin B, colletorin B, ilicicolin C, E, and F, as well as the phytotoxin alpha,beta-dehydrocurvularin have been isolated from liquid cultures of N. galligena obtained from the xylem of infected apple trees in central Chile. Ilicicolin C and F and alpha,beta-dehydrocurvularin were active against Pseudomonas syringae with IC50 values of 28.5, 28.5, and 14.2 microg/mL, respectively, in the same range as streptomycin and penicillin G (11 and 15 microg/mL, respectively). All of the compounds showed moderate inhibitory activity toward the enzymes acetylcholinesterase (AChE) and beta-glucuronidase. The most active enzyme inhibitors were colletochlorin B and ilicicolin C and E, with IC50 values of 30-36 microg/mL in the AChE assay and 32-43 microg/mL in the beta-glucuronidase test. All of the chlorinated compounds showed some toxicity toward human lung fibroblasts, with IC50 values in the range of 64-120 microg/mL. alpha,beta-Dehydrocurvularin proved to be the most toxic compound, showing IC50 values less than 12 microg/mL. The effect of the isolated compounds on seed germination and radicle and epicotyl growth was assessed in lettuce and millet seeds. At 100 and 200 microg/disk, alpha,beta-dehydrocurvularin significantly reduced radicle length and epicotyl growth in Lactuca sativa. This is the first report on the occurrence of colletochlorin B, colletorin B, ilicicolin C, E, and F, as well as alpha,beta-dehydrocurvularin associated to N. galligena.  相似文献

The effect of common bean (Phaseolus vulgaris L.) on the composition of nitrogen fixing bacterial assemblages in soil was studied by comparing planted and unplanted soil. The community composition was studied by terminal restriction fragment length polymorphism (T-RFLP) of the nitrogenase reductase gene (nifH). Principal component analysis (PCA) of T-RFLP profiles showed the separation of profiles from planted and unplanted soil. Terminal restriction fragments (T-RFs) corresponding to rhizobial bacteria were identified preferentially in planted soil; however most nifH T-RFs in soil could not be assigned to T-RFs simulated from a database of known diazotrophs. To specifically study rhizobial bacteria in the soil and nodules, PCR products from the alpha subunit of the nitrogenase enzyme (nifD) were analyzed by denaturing gradient gel electrophoresis (DGGE). DGGE results showed the specific stimulation of the rhizobial microsymbionts in planted soil.  相似文献

We utilized Fire Potential Index (FPI) to assess quantitatively fire season in a Mediterranean and a Temperate ecoregion of Iberian Peninsula by means of Time Series Analysis. The FPI was derived using MODIS NDWI and NDVI time series as indicators of vegetation status. Temperature and relative humidity time series from meteorological stations were interpolated using Universal Kriging. The average FPI time series were computed for each ecoregion for the period running from January 2001 to December 2005 and Time Series Analysis was applied to these series. Results show that FPINDWI and FPINDVI were able to describe fire season in two different ecoregions. Using Time Series Analysis was possible to evaluate and model fire dynamics. By means of spectral analysis and autoregressive models it was possible to detect hidden periodic components and model FPI dynamics with previous FPI information. Large differences between the Mediterranean and the Temperate ecoregions were demonstrated and subtle variability in the Temperate region could be explained. This quantitative approach represents an operational new tool for assessing subtle variations in fire season due to climate change.  相似文献
