Diesel exhaust particles (DEPs) are particulate matter from diesel exhaust containing many toxic compounds, such as polyaromatic hydrocarbons (PAHs). Some toxicities of PAH are considered to express via aryl hydrocarbon receptor (AhR). We hypothesized that the male reproductive toxicity of DEPs may depend on PAHs. BALB/c male mice received 24.7, 74.0 or 220 microg/mouse DEP suspension or vehicle injected into the dorsal subcutaneous layer 10 times during 5 weeks. The mice were euthanized, and blood and organs were collected 2 weeks after the last treatment. The epididymis weights, relative epididymis weights per body weight and daily sperm productions and viabilities of the 74.0 and 220 microg/mouse DEP-treated groups decreased significantly compared with those of the vehicle group. The total incidence of sperm abnormalities in the 74.0 and 220 microg/mouse DEP-treated groups increased significantly compared with the vehicle group. The seminiferous epithelium area ratios of the 74.0 and 220 microg/mouse DEP-treated groups were significantly higher compared with the vehicle and 24.6 microg/mouse DEP-treated groups. The ratios of seminiferous tubules with elongated-type spermatids in the 74.0 and 220 microg/mouse DEP-treated groups were significantly decreased compared with the vehicle group. The testosterone level and hepatic ethoxyresorufin-O-deethylase (EROD) activity as an indirect index of AhR activity in the 74.0 microg/mouse DEP-treated group were significantly increased compared with those of the vehicle group. These results clearly demonstrated that DEPs suppress testicular function, especially spermatogenesis and sperm motility. These effects may be AhR dependent. 相似文献

The accident at the Fukushima Daiichi Nuclear Power Plant (FNPP) released a large amount of radioactive substances into the environment. Furthermore, beef contaminated with radioactive cesium above the 500 Bq/kg safety standard was circulated in the food chain in 2011. Japanese consumers remain concerned about the safety of radioactively contaminated food. In our previous study, we detected a linear correlation between radioactive cesium (137Cs) activity in blood and muscle around 500 to 2500 Bq/kg in cattle. However, it was unclear whether the correlation was maintained at a lower radioactivity close to the current safety standard of 100 Bq/kg. In this study, we evaluated 17 cattle in the FNPP evacuation zone that had a 137Cs blood level less than 10 Bq/kg. The results showed a linear correlation between blood 137Cs and muscle 137Cs (Y = 28.0X, R2 = 0.590) at low radioactivity concentration, indicating that cesium radioactivity in the muscle can be estimated from blood radioactivity. This technique would be useful in detecting high‐risk cattle before they enter the market, and will contribute to food safety. 相似文献

Pigs throughout the world are afflicted with leptospirosis, causing serious economic losses and potential hazards to human health. Although it has been known that leptospiral lipopolysaccharide (L‐LPS) is involved in an immunological reaction between an antigen and a host cell, little is known about how the immune system of pigs can respond to L‐LPS. Here, we stimulated pig fibroblasts by L‐LPS and then quantitatively measured gene and protein expression levels of two toll‐like receptors (TLRs), TLR2 and TLR4, by real‐time PCR and Western blotting. As a result, expression of TLR2 was found to be significantly up‐regulated within 24 h after L‐LPS stimulation whereas induction of TLR4 expression was relatively weak. We also revealed that of myeloid differentiation primary response gene 88 (MyD88), interleukin 6 (IL‐6) and IL‐8 gene expressions were markedly up‐regulated by L‐LPS stimulation. These results may suggest that the pig cell can activate TLR2 rather than TLR4 by L‐LPS stimulation, thereby inducing expression of cytokines. 相似文献

Autologous peripheral nerve grafts were used to permit and direct the regrowth of retinal ganglion cell axons from the eye to the ipsilateral superior colliculus of adult hamsters in which the optic nerves had been transected within the orbit. Extracellular recordings in the superior colliculus 15 to 18 weeks after graft insertion revealed excitatory and inhibitory postsynaptic responses to visual stimulation. The finding of light-induced responses in neurons in the superficial layers of the superior colliculus close to the graft indicates that axons regenerating from axotomized retinal ganglion cells can establish electrophysiologically functional synapses with neurons in the superior colliculus of these adult mammals. 相似文献

In the field of agriculture, development of evaluation techniques for environmental changes is urgently required for the purpose of finding a balance between growth in agricultural productivity and environmental considerations. The habitat evaluation procedures (HEP) constitute one technique for habitat assessment. While HEP is widely applied to estimate both habitat quality and quantity in an environment, it appears to be necessary to develop an accurate habitat prediction model in order to evaluate environments precisely. In fact, habitat selection by fish is affected by complicated interaction between multiple environmental factors, which makes it difficult to relate physical environments to habitat preference. In the present study, we utilize artificial neural networks (ANNs), which are commonly applied to model complex systems, to predict the habitat selection of Japanese medaka (Oryzias latipes) in agricultural canals. Considering the essential vagueness of fish behavior, fuzzy membership functions are introduced into the input layer, which advances ANN to fuzzy neural network (FNN). In addition, symmetric triangular fuzzy numbers are employed to account for uncertainty in measurement errors and dispersions of physical environment. The FNN model precisely predicts the habitat preference of Japanese medaka in an agricultural canal, and the results show a good agreement between the calculated and observed habitat suitability indices (HSI). The purpose of this research is in solution of two important subjects mutually related; (1) creation of fabrics of various handlings by finish processing and (2) mechanism of generation of itchy feeling acting as the hindrance of a comfortable clothing life and establishment of its objective evaluation method. For the purpose of solution of these two subjects, the same fabrics were used and they were changed by 18 kinds of different physicochemical procedures, which come out 18 levels of fabric handlings ranging from very soft to very stiff. As for handlings of 18 kinds of fabrics, the items of Hand Value were calculated based on all of KES basic properties in terms of KES procedures. Solution of these subjects is also connected with a high level of fabric finishing technology. On the other hand, one of the authors is an expert in fabric finish processing, and has been sure of itchy feeling of fabrics being closely connected with the hardness of fabrics from many years of experience. The degrees of itchy feeling for 18 kinds of fabric were judged by 50 evaluators of different ages and occupations, in terms of five ranks of evaluation method beforehand. As a result, itchy feeling had a high correlation with shear properties and Koshi, and had a negative high correlation with Numeri. It is desirable to increase the flavonoid contents of postharvest vegetables since flavonoids play a beneficial role in human health promotion. In the present study, we show that postharvest vegetables increasingly produced flavonoids when irradiated with light near the absorption wavelength of flavonoids in the plant. Three-day exposure to UV-B for 5 min, 98 μmol m?2 s?1 per day, increased the contents of jaceidin in spinach, kaempherol glycoside in radish sprout, apigenin glycosides in parsley, and isovitexin in Indian spinach after 6 days of storage in a refrigerator, compared to the contents in plants without irradiation. Six days of storage of unripe green strawberry under green light for 5 min, 98 μmol m?2 s?1 per day, enabled them to mature and turn red, accompanied by 3.5-fold increased contents of pelargonidin. Elucidation of the mechanism in parsley found the stimulating expression of the flavonoid synthesis gene, PAL, C4H, 4CL, CHS, and FNS, 6 h after exposure to single irradiation with UV-B for 5 min, and the higher expression was maintained for 24 h. After 3 days irradiation during 6 days of storage, parsley did not show adverse changes in the contents of ascorbic acid, β-carotene, chlorophyll, and moisture. 相似文献

A direct competitive enzyme-linked immunosorbent assay (dc-ELISA) was developed for residue analysis of azoxystrobin in garden crops, for which the maximum residue limits (MRLs) are 0.5-50 mg/kg in Japan. For hapten synthesis, an ethyl carboxyl group was introduced to the 4-position of the 2-cyanophenoxy group in azoxystrobin, and its cyano group was changed to a methyl group. An anti-azoxystrobin monoclonal antibody was prepared from mice immunized with hapten-keyhole limpet hemocyanin conjugate. The dc-ELISA using prepared antibody showed 50-250-fold higher sensitivity compared to the MRLs. The working range of the dc-ELISA was 10-200 ng/mL. The dc-ELISA showed high specificity to azoxystrobin. When methanol extracts from nine kinds of garden crops spiked with azoxystrobin ranging near the MRLs were analyzed, the determined results by the dc-ELISA agreed well with the results of their controls. In addition, azoxystrobin spiked in garden crops homogenates was satisfactorily extracted by methanol solution and easily analyzed. The recovery rate of dc-ELISA was 96-109% and correlated well with the results obtained by HPLC analysis. 相似文献
